Caracterização do sistema antioxidativo no estabelecimento da associação micorrízica Castanea sativa Mill. / Amanita muscaria IPB - Escola Superior Agrária

As micorrizas são associações mutualistas que se estabelecem entre fungos, pertencentes sobretudo às divisões Basidiomycota, Ascomycota e Zigomycota, e raízes de plantas vasculares. Com o estabelecimento da associação micorrízica, ocorrem várias alterações morfológicas, fisiológicas e ecológicas em ambos os intervenientes, que têm sido objecto de estudo por diversos autores. Contudo, os mecanismos que controlam o processo de colonização e como este processo se inicia é completamente desconhecido. Mycorrhizae are symbiotic associations between soil borne fungi and the root system of higher plants having a beneficial role on plant growth. The fungus receives carbon from plant, while the plant increased nutrient uptake mediated via the fungus. In agroforestry systems, most of the mycorrhizae belong to ectomycorrhizal (ECM) group being the mechanisms controlling its development poorly defined. During arbuscular mycorrhizal (AM) establishment, some evidences suggest that a temporal and spatial activation of different defence mechanisms by plants are activated. No Nordeste Transmontano, a cultura do castanheiro, Castanea sativa Mill., tem uma grande importância, económica, social, cultural e paisagística. Esta espécie caracteriza-se por estabelecer associações simbióticas com fungos do solo, resultando na formação de ectomicorrizas que, na sua maioria, são produtoras de estruturas reprodutoras macroscópicas, os macrofungos. Diversity and fruiting pattern of macrofungi associated with chestnut tree (Castanea sativa Mill.) in the Trás-os-Montes region (Northeast Portugal) IPB - Escola Superior Agrária

The chestnut (Castanea sativa) agro-ecosystem is of great social, economic and landscape importance in NE Portugal. There are multiple resources associated with this crop, among them fruit and wood production and mushroom harvesting. However, information about the diversity and ecology of macrofungi is very scarce. In this context, the aim of this study was to assess the macrofungal diversity associated with chestnut trees over 4 y. Carpophore surveys were conducted in a non-tilled C. sativa orchard located in Braganc¸a, Portugal, from Sep. 2002 to Dec. 2005. Efeito de extractos de raízes eliciadas de Castanea sativa no crescimento do fungo ectomicorrízico Pisolithus tinctorius IPB - Escola Superior Agrária

O castanheiro (Castanea sativa Mill.) estabelece associação com numerosas espécies de fungos ectomicorrízicos, estando descritos os efeitos benéficos para a planta após micorrização com Pisolithus tinctorius. Neste processo é essencial que ocorra a troca de sinais entre os simbiontes, para que seja reconhecida a sua compatibilidade e para que ocorra a formação dos órgãos ectomicorrízicos. Neste trabalho são fornecidas evidências que sugerem a capacidade de extractos de raízes de castanheiro, nos estádios iniciais de contacto com P. tinctorius regularem o crescimento do fungo micorrízico.

Effect of soil tillage on diversity and abundance of macrofungi associated with chestnut tree in the Northeast of Portugal IPB - Escola Superior Agrária

European chestnut trees (Castanea sativa Mill.) are of great economic importance for wood and fruit production in the northeast of Portugal. In order to maintain the soil free of weeds, farmers traditionally practice several superficial tillages along the year. Phytosanitary problems mainly related with increasing Phytophtora cinnamomi contamination of soils are progressively changing these practices.

In vitro evidences of mycoparasitism of the ectomycorrhizal fungus Pisolithus tinctorius by Hypholoma fasciculare IPB - Escola Superior Agrária

In the northeast of Portugal macrofungi community associated to chestnut tree (Castanea sativa Mill.) is rich and diversified. Among fungal species, Pisolithus tinctorius and Hypholoma fasciculare are common in this habitat. In vitro interaction between P. tinctorius and H. fasciculare was investigated, to ascertain the potential mycoparasitic capabilities of H. fasciculare in the growth suppression of ectomycorrhizal P. tinctorius. The results show that in co-culture, the growth of P. tinctorius was drastically inhibited by H. fasciculare. Preliminary studies seem to evidence P. tinctorius hyphal injury and stress response in the contact zone between both mycelia. Interaction between Hypholoma fasciculare and the ectomycorrhizal fungus Pisolithus tinctorius IPB - Escola Superior Agrária

In the past few years, cord-forming basidiomycetes have received considerable attention in their growth strategies in the presence of other fungi (saprotrophic and pathogenic). In the present study, the in vitro interaction between the cord-forming basidiomycete Hypholoma fasciculare and the ectomycorrhizal fungus Pisolithus tinctorius was studied. Dual-cultures were established and the radial fungal growth, hyphae morphology and production of volatile compounds were evaluated during interaction. Involvement of reactive oxygen species during early stages of ectomycorrhiza establishment between Castanea sativa and Pisolithus tinctorius IPB - Escola Superior Agrária

Evidence for the participation of reactive oxygen species (ROS) and antioxidant systems in ectomycorrhizal(ECM) establishment is lacking. In this paper, we evaluated ROS production and the activities of superoxide dismutase (SOD) and catalase (CAT) during the early contact of the ECM fungus Pisolithus tinctorius with the roots of Castanea sativa (chestnut tree). Roots were placed in contact with P. tinctorius mycelia, and ROS production was evaluated by determining the levels of H2O2 and O2 I− during the early stages of fungal contact. Three peaks of H2O2 production were detected, the first two coinciding with O2 I− bursts. During mycorrhiza establishment morphological, physiological and ecological alterations occur in both symbionts.
