1H nuclear magnetic resonance spectroscopy-based studies of the metabolism of food-borne carcinogen 2-amino-3-methylimidazo[4,5-f]quinoline by human intestinal microbiota Inra
Humblot, C.; Combourieu, B.; Väisänen, M.J.; Furet, J.P.; Delort, A.M.; Rabot, S..
2-Amino-3-methylimidazo[4,5-f]quinoline (IQ) is a mutagenic/carcinogenic compound formed from meat and fish during cooking. Following ingestion, IQ is metabolized mainly by liver xenobiotic-metabolizing enzymes, but intestinal bacteria may also contribute to its biotransformation. The aim of this study was to investigate the metabolism of IQ by the human intestinal microbiota. Following incubation of IQ (200 microM) under anoxic conditions with 100-fold dilutions of stools freshly collected from three healthy volunteers, we quantified residual IQ by high-pressure liquid chromatography (HPLC) analysis and characterized the production of IQ metabolites by in situ (1)H nuclear magnetic resonance ((1)H-NMR) spectroscopic analysis of crude incubation media. In...

20-Hydroxyecdysone deters oviposition and larval feeding in the European grapevine moth, Lobesia botrana Inra
Calas, D.; Thiery, D.; Marion-Poll, F..
European grapevine moth females (Lobesia botrana, Lepidoptera Tortricidae) select an oviposition site by tasting the host plant surface and then gluing a single egg on berries from grapes or from several other host plant species. In doing so, females should avoid ovipositing on plants that are detrimental to their progeny. Do they sense the same deterrent compounds as larvae, despite the fact that they do not have access to the same compartments of the plants? We tested this hypothesis with 20-hydroxyecdysone (20E), purified from Leuzea carthamoides. Phytoecdysteroids are usually found inside plant tissues and accessible to larvae in an aqueous phase, while adults would access them only through the epicuticular wax. We first confirmed that larvae avoid...

2-methylthiazolidine and 4-ethylguaiacol, male sex pheromone components of the cockroach Nauphoeta cinerea (Dictyoptera, Bladeridae) : A reinvestigation Inra
Sirugue, D.; Bonnard, O.; Le Quere, J.L.; Farine, J.P.; Brossut, R..
In Nauphoeta cinerea, male calling behavior is associated with sex pheromone release by the sternal glands. The male pheromone that attracts females from a distance is a mixture of 2-methylthiazolidine and 4-ethylguaiacol. It is active at very low concentrations, 0.05 and 0.01 ng, respectively. Two other compounds, 3-hydroxy-2-butanone and 2-methyl-2thiazoline, act at close range, keeping the female in the vicinity of the male. The function of the volatile pheromone and those of previously described contact pheromones are discussed in regard to their possible involvement in the establishment of male dominant-subordinate relationships.

3D virtual plants to phenotype differences among genotypes: Taking into account plant-environment interactions to better understand genetic variability in leaf development response to light Inra
Chenu, K.; Lecoeur, J..
Incident light affects orgnaogenesis and morphogenesis processes involved in leaf development through the amount of radiation absorbed by the plant (Chenu et al., 2005). The genetic variability of these responses was investigated on Arabidopsis thaliana ecotypes (Col, Di-m, Ler, Ws) and mutants (se-1, rot3-1, ron2-2, p70S-KOR) displaying contrasted architectures and radiation use efficiencies (Fig. 1).Plants were grown under various levels of incident light, with a stable light quality. The local plantenvironmentinteractions were estimated for each genotype, from plant emergence to the end ofrosette expansion, using an architectural model (Barczi et al., 1997) coupled with a radiative model (Dauzat and Eroy, 1997). Leaf development was assessed in terms of...

(3Z,6Z,8E)-3,6,8-dodecatrien-1-ol : Sex pheromone in a higher fungus-growing termite, Pseudacanthotermes spiniger (Isoptera, Macrotermitinae) Inra
Bordereau, C.; Robert, A.; Bonnard, O.; Le Quere, J.L..
The female sex pheromone of the fungus-growing termitePseudacanthotermes spiniger (Termitidae, Macrotermitinae) was isolated from sternal glands of alates. The compound inducing attraction and excitation in males was identified as the (3Z,6Z,8E)-3,6,8-dodecatrien-1-ol by GC-MS, microhydrogenation, GC-FTIR, and NMR. This unsaturated alcohol is present in both sexes but in much higher quantities in females than in males (about 10 times). The hypothesis is suggested that this alcohol, which is detected at extremely low concentrations by the workers ofP. spiniger, may be used either as a trail-following pheromone or a sex pheromone according to concentrations and to target castes. The presence of this alcohol in Macrotermitinae reinforces the idea of a...

A 3D model for simulating the spatial and temporal distribution of temperature within ellipsoidal fruit Inra
Saudreau, M.; Sinoquet, H.; Santin, O.; Marquier, A.; Adam, B.; Longuenesse, J.J.; Guilioni, L.; Chelle, M..
A physical model simulating the spatio-temporal distribution of temperature in an ellipsoidal fruit has been developed. It is based on the numerical resolution of the three-dimensional (3D) unsteady heat conduction equation with unsteady and non-homogeneous heat fluxes as boundary conditions. The numerical scheme and the physical models have been tested by comparing with an analytical solution for simple configurations. The model quality has been assessed by comparing model outputs to temperature measured with thermocouples at several locations in isolated peach and apple fruits. The root mean square error (RMSE) for temperature simulated at a 1 min time step was about 0.8 °C. A sensitivity analysis showed that accurate estimations of the surface...

A 454 multiplex sequencing method for rapid and reliable genotyping of highly polymorphic genes in large-scale studies Inra
Galan, M.; Guivier, E.; Caraux, G.; Charbonnel, N.; Cosson, J.F..
Background: High-throughput sequencing technologies offer new perspectives for biomedical, agronomical and evolutionary research. Promising progresses now concern the application of these technologies to large-scale studies of genetic variation. Such studies require the genotyping of high numbers of samples. This is theoretically possible using 454 pyrosequencing, which generates billions of base pairs of sequence data. However several challenges arise: first in the attribution of each read produced to its original sample, and second, in bioinformatic analyses to distinguish true from artifactual sequence variation. This pilot study proposes a new application for the 454 GS FLX platform, allowing the individual genotyping of thousands of samples in one...

A bio-economic modeling approach to asses the environmental and economic sustainability of farming practices in a tunisian watershed Inra
Belhouchette, H.; Blanco, M.; Hengsdijk, H.; Wéry, J.; Flichman, G..
In agriculture, short-term objectives to increase farm income often conflict with long-term objectives to maintain soil fertility. In semi-arid regions such as Northern Tunisia, deficit irrigation to reduce water use and nitrate leaching may reduce the long-term production potential due to soil salinization. Until now, the impact of water management on nitrate leaching, soil salinization, production potential and economic performance of irrigated farms has not been quantified. This study aims at analyzing the effects of different water and nutrient management on three farm performance indicators, i.e. nitrate leaching, soil salinization and farm income. We use a new analytical bio-economic approach to assess the environmental and economic performance of...

A carbon-14-glucose assay to compare microbial activity between rhizosphere samples Inra
Nguyen, C.; Henry, F..
The present work investigated to what extent the utilisation of 14C-glucose by soil microorganisms could be used to compare microbial activity between rhizosphere samples. A very low amount of 14C-glucose was added to the soil to be tested. After incubation for 6 days at a constant temperature in the dark, we determined the mineralised fraction (14CO2) and the 14C of the microbial biomass rendered extractable after fumigation with chloroform vapour (14CFE). In unplanted soils, we observed that the 14CO2/14CFE ratio increased when microbial growth was stimulated by addition of unlabelled glucose (20-500 µg C g-1 soil) compared to the control (no unlabelled glucose added). When 14C-glucose was added immediately after unlabelled glucose amendments, the...
