Several methodologies in molecular biology have been used in the investigation of Malassezia pachydermatis and its differentiation into subtypes. Recent molecular research of this species includes the use of samples isolated from canine otitis externa and dermatitis, as well as from healthy animals, having in view an epidemiological study of the yeast. The aim of this study was to identify molecular differences in M. pachydermatis samples isolated from dogs with otitis externa. The M. pachydermatis strains were analyzed by means of the Random Amplification Primer DNA - Polimerase Chain Reaction (RAPD–PCR) for molecular heterogeneity research. DNA extraction was carried out with phenol-chloroform and the RAPD technique using the AGAATCCGCC primer. A...

Production and partial characterization of proteases from Mucor hiemalis URM3773 Biological Sciences
Ribeiro, Roana Cecília dos Santos; Ribeiro, Thaís Rafaelle dos Santos; Souza-Motta, Cristina Maria de; Medeiros, Erika Valente; Moreira, Keila Aparecida.
The current study evaluated the proteases production from 11 fungal species belonging to the genera Mucor, Rhizomucor and Absidia. The species were obtained from the Collection of Cultures URM at the Mycology Department-UFPE, Brazil. The best producing species was Mucor hiemalis URM 3773 (1.689 U mL-1). Plackett-Burman design methodology was employed to select the most effective parameter for protease production out of 11 medium components, including: concentration of filtrate soybean, glucose, incubation period, yeast extract, tryptone, pH, aeration, rotation, NH4Cl, MgSO4 and K2HPO4. Filtrated soybean concentration was the significant variable over the response variable, which was the specific protease activity. The crude enzyme extract showed optimal...

Screening of an adapted culture medium composed by different carbon sources for heterotrophic cultivation of Chlorella vulgaris using a microplate assay Biological Sciences
Fonseca, Gustavo Graciano; Silva, Alisson Alves da.
The ability of microalgae to grow heterotrophically lies in the ability to oxidize organic compounds present in the external environment. The aim of this work was to evaluate the growth of Chlorella vulgaris under heterotrophic culture conditions using Basal Bold and NPK media supplemented with different sole carbon sources (glucose, fructose, sucrose, glycerol or acetate). The kinetic parameters obtained were maximum specific growth rate (µmax), doubling time (DT), maximal absorbance, which was also converted to cell concentration values using a linear relation, and cell productivity (PX). Among all the treatments analyzed, the highest maximum specific growth rate found was 0.030 hour-1 (0.72 day-1) in the treatment using Basal Bold medium supplemented...

Socio-environmental and microbiological assessment of the river Piancó, Pombal, Brazil Biological Sciences
Lacerda, Eulina Kelly Guilherme; Silva, Edevaldo da; Medeiros, Rosália Severo.
The River Piancó in the municipality of Pombal on the western stretch of the state of Paraíba, Brazil, receives contaminated effluents. Current assay evaluates the microbiological quality of the river water and analyzes the social and environmental aspects of the riverine community with regard to the water and its use of the river. The water´s microbiological quality was verified at four sites on the river in five two-monthly collections throughout the year (totaling 20 samples) for the following analyses: Most Probably Number (MPN), Total (TC) and Fecal Coliform (FC) and identification of Escherichia coli. The social and environmental aspects were assessed by a 15-question questionnaire given to thirty homes. Results report Fecal Coliforms (> 240 to...
