Activated charcoal and graphite for the micropropagation of Cattleya bicolor Lindl. and a orchid double-hybrid ‘BLC Pastoral Innocence’ - doi: 10.4025/actasciagron.v34i2.12257 Agronomy

No previous studies have investigated the use of graphite instead of activated charcoal in orchids. In this work, different concentrations of activated charcoal or graphite were added to KC medium to darken the culture medium and stimulate the in vitro propagation of Cattleya bicolor and of a double hybrid orchid (‘BLC Pastoral Innocence’). The seedlings were inoculated on growth regulator-free KC medium; the effects of activated charcoal (0.0, 1.5, 3.0, 4.5, 6.0, and 7.5 g L-1) and graphite (0.0, 1.5, 3.0, 4.5, 6.0, and 7.5 g L-1) concentrations were tested separately. Each flask contained fifteen seedlings, and the experiments had a randomised, two-way factorial design. The two tested variables were culture medium and illumination levels; eleven culture...

Assessment of DNA extraction methods for detection of arbuscular mycorrhizal fungi in plant roots by nested-PCR Biological Sciences
Diédhiou, Abdala Gamby; Borges, Wardsson Lustrino; Sadio, Oumar; de Faria, Sergio Miana.
DNA extraction methods were evaluated for the yield and purity of DNA recovered from mycorrhized roots and whether the recovered DNA is suitable for amplification of arbuscular mycorrhizal (AM) fungal SSU rDNA. The DNeasy Plant Mini Kit and three extraction buffers were used alone or in combination with either polyvinylpyrrolidone (PVP), polyvinylpolypyrrolidone (PVPP) and/or activated charcoal (AC). Among the extraction methods tested, those based on the CTAB buffers yielded more DNA than those based on the TE buffer and the DNeasy Plant Mini Kit. Moreover, the use of AC alone or in combination with PVPP reduced DNA yield, while it significantly improved the purity of recovered DNA, whatever the extraction buffer. On the other hand, the success of...

Cryopreservation of Cyrtopodium hatschbachii Pabst (Orchidaceae) immature seeds by encapsulation-dehydration Biocell
Surenciski,Mauro Rodrigo; Flachsland,Eduardo Alberto; Terada,Graciela; Mroginski,Luis Amado; Rey,Hebe Yolanda.
The aim of the present study was to investigate the efficiency of the encapsulation-dehydration technique for cryopreservation of Cyrtopodium hastchbachii Pabst seeds. Immature seeds of this species were cryopreserved by an encapsulation-dehydration technique. Seeds of five immature pods, 120 days after pollination, were encapsulated in 3% calcium alginate matrix and pretreated in liquid medium supplemented with 0.08 M sucrose (24 h), 0.15 M sucrose (24 h), 0.25 M sucrose (48 h), 0.5 M sucrose (24 h) and 0.75 M sucrose (24 h) in shaker at 60 rpm. Alginate beads were dehydrated 5 h in silicagel and immersed in liquid nitrogen for 12 h. Cryopreserved beads were thawed at 30°C for 1 min, rehydrated using the same liquid mediums (0.75 M sucrose (24 h), 0.5 M...

Simple and robust DNA extraction method for the large-scale analysis of genotypes containing high polyphenolic content, such as landracesof Solanum tuberosum and Zea mays Ciencia e Investigación Agraria
Barra,Macarena; Salazar,Erika; Beltrán,María; Sagredo,Boris.
The extraction of high-quality DNA is essential for studies conducted at the molecular level in species with an abundance of contaminants in their tissues, such as some landraces of potato and maize, in which it is difficult to extract good-quality genomic DNA. Compounds such as polyphenols interfere with the amplification of DNA during polymerase chain reaction (PCR), which makes it difficult to conduct PCR-based studies ofmolecular markers. This article describes a simple and robust protocol for DNA isolation that was applied to potato and maize landraces and resulted in a high DNA concentration with excellent purity and quality. This method is based on the method of Krizman et al. (2006) with some modifications and was tested on potato leaves and young...
