Metodologia alternativa para extração de DNA genômico de Staphylococcus aureus. Abstract: This study describes a rapid procedure for the isolation of genomic DNA from Staphylococcus aureus that yielded a good amount of high quality DNA for the amplification of staphylococcal enterotoxins genes ( A, B, C, D, and E) and the TSST-1 gene as well as enzymatic restriction ( HaeIII) from environmental isolates. With this method, it was possible to detect these genes in a sample containing as little as 105 cells with positive PCR reactions obtained from approximately 10pg of DNA in a final reaction volume of 25 mu l. Mangabeira (Hancornia speciosa) is a native fruit tree found mainly in the Cerrado biome and shows great economic potential due to its multiple uses; the fruits are used in agriculture, are important as a food resource, and can be consumed in natura or processed. Due to a reduction in the area of ecosystems where it occurs, mangabeira is threatened by genetic erosion in Brazil. The characterization of the genetic diversity of plants can provide the basis for strategies to protect and conserve endangered populations, like mangabeira. Abstract Theobroma cacao L. and its products are consumed worldwide. Those products are of great research interest due to antioxidant properties of some of their polyphenolic constituents. The amount of these polyphenols and polysaccharides has shown that can interfere with the high quality and quantity of nucleic acids for molecular research.
