Une série de nouvelles techniques, fondées sur la PCR, pour l'isolement des gènes à expression différentielle ont été récemment décrites pour l'isolement de gènes parasitaires dont la spécificité est liée au stade du parasite ou à la réaction de l'hôte. Nous avons utilisé deux de ces techniques, l'affichage différentiel et de cDNA-AFLPs, pour l'étude des changements de l'expression des gènes durant l'éclosion et le développement ultérieur des nématodes à kystes de la pomme de terre, #Globodera rostochiensis$ et #G. pallida$. The sequencing of prfA, encoding the transcriptional regulator of virulence genes, in 26 low virulence field Listeria monocytogenes strains showed that eight strains exhibited the same single amino-acid substitution: PrfAK220T. These strains exhibited no expression of PrfA-regulated proteins and thus no virulence. This substitution inactivated PrfA, since expression of the PrfAK220T mutant gene in an EGDΔprfA strain did not restore the haemolytic and phosphatidylcholine phospholipase C activities, in contrast to the wild-type prfA gene. The substitution of the lysine at position 220 occurred in the helix αH. However, the data showed that the PrfAK220T protein is dimerized just as well as its wild-type counterpart, but does not bind to PrfA boxes....

A novel family of calmodulin-binding transcription activators in multicellular organisms Inra

Screening of cDNA expression libraries derived from plants exposed to stress, with35S-labeled recombinant calmodulin as a probe, revealed a new family of proteins containing a transcription activation domain and two types of DNA-binding domains designated the CG-1 domain and the transcription factor immunoglobulin domain, ankyrin repeats, and a varying number of IQ calmodulin-binding motifs. A unigene catalogue of 5700 expressed genes in cassava Inra

Two economically important characters, starch content and cassava bacterial blight resistance, were targeted to generate a large collection of cassava ESTs. Two libraries were constructed from cassava root tissues of varieties with high and low starch contents. Other libraries were constructed from plant tissues challenged by the pathogen Xanthomonas axonopodis pv. manihotis. We report here the single pass sequencing of 11 954 cDNA clones from the 5' ends, including 111 from the 3' ends. Cluster analysis permitted the identification of a unigene set of 5700 sequences. Lactic acid bacteria and particularly Lactococcus lactis are widely used for the production of lactic acid in fermented foods. Control of the catabolic rate in L. lactis, i.e., the rate of lactic acid production, appears to be determinant for dairy product quality. While the mechanisms involved in control have not been totally elucidated, they seem to depend upon the strain and the growth conditions. Furthermore, it remains unclear whether the catabolic rate is controlled at the level of transcription, translation or enzyme activity. ANAIS is a user-friendly web-based tool for the processing of NimbleGen expression data. The interface reads single-channel microarray files generated by NimbleGen platforms and produces easily interpretable graphical and numerical results. It provides biologists six turnkey analysis modules—normalization, probe to gene, quality controls, differential expression, detection, queries and clustering—to explore quickly, freely and without the need for computer programming, NimbleGen transcriptome data.

Analysis of CATMA transcriptome data identifies hundreds of novel functional genes and improves gene models in the Arabidopsis genome Inra

Background: Since the finishing of the sequencing of the Arabidopsis thaliana genome, the Arabidopsis community and the annotator centers have been working on the improvement of gene annotation at the structural and functional levels. In this context, we have used the large CATMA resource on the Arabidopsis transcriptome to search for genes missed by different annotation processes. Probes on the CATMA microarrays are specific gene sequence tags (GSTs) based on the CDS models predicted by the Eugene software. Among the 24 576 CATMA v2 GSTs, 677 are in regions considered as intergenic by the TAIR annotation. Background: Invasion of the mosquito salivary glands by Plasmodium is a critical step for malaria transmission. From a SAGE analysis, we previously identified several genes whose expression in salivary glands was regulated coincident with sporozoite invasion of salivary glands. To get insights into the consequences of these salivary gland responses, here we have studied one of the genes, PRS1 (Plasmodium responsive salivary 1), whose expression was upregulated in infected glands, using immunolocalization and functional inactivation approaches. Plant growth and development are sensitive to light. Light-responsive DNA-binding transcription factors have been functionally identified. However, how transcription initiation complex integrates light signals from enhancer-bound transcription factors remains unknown. In this work, we characterized mutations within the Arabidopsis HAF2 gene encoding TATA-binding protein-associated factor TAF1 (or TAFII250). The mutation of HAF2 induced decreases on chlorophyll accumulation, light-induced mRNA levels, and promoter activity.
