Development of a BALB/c mouse model of Helicobacter pylori infection with fresh and frozen bacteria Biol. Res.
Rabelo-Gonçalves,Elizabeth M A; Nishimura,Nancy F; Murilo R Zeitune,José.
An experimental model for H. pylori infection was established by intragastrically challenging BALB/c mice with 1 ml (10(8) CFU/ml) of suspension for two consecutive days. Animals were divided into three groups. GA: mice inoculated with fresh bacteria; GB: mice inoculated with frozen bacteria, and GC: mice inoculated with brucella broth (control group). Animals were killed at 7, 14, 21, 28, 35 and 60 days pi and fragments of stomach and duodenum were collected, paraffin embedded and stained by hematoxylin-eosin and Giemsa. The results showed that challenged mice exhibited mild duodenitis and gastritis. In group GA, infiltration in the duodenum was lymphoplasmacytic until day 35; in group GB, it was lymphomonocytic for 60 days pi. In the stomach, H. pylori...

Experimental evidence of dietary ciguatoxin accumulation in an herbivorous coral reef fish ArchiMer
Clausing, Rachel J.; Losen, Barbara; Oberhaensli, Francois R.; Darius, H. Taiana; Sibat, Manoella; Hess, Philipp; Swarzenski, Peter W.; Chinain, Mireille; Bottein, Marie-yasmine Dechraoui.
Ciguatoxins (CTXs) are potent algal toxins that cause widespread ciguatera poisoning and are found ubiquitously in coral reef food webs. Here we developed an environmentally-relevant, experimental model of CTX trophic transfer involving dietary exposure of herbivorous fish to the CTX-producing microalgae Gambierdiscus polynesiensis. Juvenile Naso brevirostris were fed a gel-food embedded with microalgae for 16 weeks (89 cells g.1 fish daily, 0.4 ¦Ìg CTX3C equiv kg.1 fish). CTXs in muscle tissue were detectable after 2 weeks at levels above the threshold for human intoxication (1.2 ¡À 0.2 ¦Ìg CTX3C equiv kg.1). Although tissue CTX concentrations stabilized after 8 weeks (¡«3 ¡À 0.5 ¦Ìg CTX3C equiv kg.1), muscle toxin burden (total ¦Ìg CTX in muscle tissue)...

Infection dynamics of a V. splendidus strain pathogenic to Mytilus edulis : In vivo and in vitro interactions with hemocytes ArchiMer
Ben Cheikh, Yosra; Travers, Marie-agnes; Le Foll, Frank.
The pathogenic strain V. splendidus 10/068 1T1 has previously been reported for its virulence to the blue mussel and for its capacity to alter immune responses. In this study, we expanded the knowledge on hemocyte-pathogen interactions by using in vitro and in vivo assays. V. splendidus 10/068 1T1 severely inhibited cell adhesion and acidic vacuole formation unlike the innocuous phylogenetically related V. splendidus 12/056 M24T1 which had no effect on these cell functions. Furthermore, the virulent bacteria decreased hemocyte viability (59% of viability after 24 h). Infection dynamics were explored by using a model based on water tank cohabitation with septic mussels infected by GFP-tagged V. splendidus 10/068 1T1. Experimental infections were...

Paraquat poisoning: an experimental model of dose-dependent acute lung injury due to surfactant dysfunction BJMBR
Silva,M.F.R.; Saldiva,P.H.N..
Since the most characteristic feature of paraquat poisoning is lung damage, a prospective controlled study was performed on excised rat lungs in order to estimate the intensity of lesion after different doses. Twenty-five male, 2-3-month-old non-SPF Wistar rats, divided into 5 groups, received paraquat dichloride in a single intraperitoneal injection (0, 1, 5, 25, or 50 mg/kg body weight) 24 h before the experiment. Static pressure-volume (PV) curves were performed in air- and saline-filled lungs; an estimator of surface tension and tissue works was computed by integrating the area of both curves and reported as work/ml of volume displacement. Paraquat induced a dose-dependent increase of inspiratory surface tension work that reached a significant two-fold...

Somatic, Biochemical and Hepatic Alterations in Wild Type Mice Chronically Fed High Fat Diet International Journal of Morphology
Neves,Renata Heisler; Barros Alencar,Alba Cristina Miranda de; Aguila,Marcia Barbosa; Mandarim-de-Lacerda,Carlos Alberto; Machado-Silva,José Roberto; Gomes,Delir Corrêa.
This study evaluated whether a high fat diet (HFC group) induces overweight, hepatic steatosis and plasma lipoproteins level alteration compared to standard chow diet (SC group). Female mice were submitted to each diet over 6 months. Body mass and food intake were evaluated weekly throughout the experiment. Total cholesterol, TG, LDL-c, HDL-c and VLDL-c were analyzed. Mice were sacrificed to remove liver, spleen, heart and intestine. The volume of the organs was determined according to the submersion method. Fixed livers were embedded in paraffin and stained with hematoxylin and eosin and Masson's trichrome. The analysis used a video microscope system and a test-system with 42 test-points. The volume density was estimated for hepatocytes, steatosis and...
