A rapid method for sequencing of rRNA gene(s) amplified by polymerase chain reaction using an automated DNA sequencer IRD

A method for DNA sequencing of ribosomal RNA (rRNA) genes, amplified by polymerase chain reaction (PCR), using internal primers, designed on the basis of conserved regions of rRNA genes for determining a near complete sequence (99%) of the gene using an automated DNA sequencer (Applied Biosystem Incorporation, USA) is described. The procedure is extremely rapid as cloning of the gene is not required for sequence determination. In addition time consuming steps such as ethanol precipitation and hazardous steps such as phenol/chloroform extractions are excluded from the protocol for the purification of extension products after Taq cycle sequencing using the ABI dye terminator chemistry. A Rhodococcus qsdA-Encoded Enzyme Defines a Novel Class of Large-Spectrum Quorum-Quenching Lactonases Inra

A gene involved in N-acyl homoserine lactone (N-AHSL) degradation was identified by screening a genomic library of Rhodococcus erythropolis strain W2. This gene, named qsdA (for quorum-sensing signal degradation),encodes an N-AHSL lactonase unrelated to the two previously characterized N-AHSL-degrading enzymes, i.e., the lactonase AiiA and the amidohydrolase AiiD. QsdA is related to phosphotriesterases and constitutes thereference of a novel class of N-AHSL degradation enzymes. It confers the ability to inactivate N-AHSLs with an acyl chain ranging from C6 to C14, with or without substitution at carbon 3. Presented herein is an updated sheep cytogenetic map that contains 452 loci (291 type Iand 161 type II) assigned to specific chromosome bands or regions on standard R-bandedideograms. This map, which significantly extends our knowledge of the physical organizationof the ovine genome, includes new assignments for 88 autosomal loci, including 74type I loci (known genes) and 14 type II loci (SSRs/microsatellite marker/STSs), by FISHmappingand R-banding. Comparison of the ovine map to the cattle and goat cytogeneticmaps showed that common loci were located within homologous chromosomes andchromosome bands, confirming the high level of conservation of autosomes amongruminant species. L'apomixie diplosporique est un mode de reproduction asexuée par graines caractéristique des formes polyhaploïdes du genre #Tripsacum$, apparenté sauvage du maïs. L'introduction du ou des gènes responsables de ce caractère chez les plantes cultivées ouvrirait de nouvelles perspectives pour l'amélioration des plantes et l'agriculture. Dans le cadre du programme de transfert de l'apomixie au maïs conduit au CIMMYT, différentes espèces de #Tripsacum$ ont été croisées avec le maïs et de nombreux hybrides F1 ont été obtenus. Effets du gène « crête en pois » et d’autres gènes à effet visible sur le poids des coqs adultes Inra

Dans un troupeau expérimental du type « pondeuse à œufs teintés » mi-lourd, de 1961 à 1984, des génotypes relatifs à des locus de coloration ou à effet morphologique ont été comparés sur des couples de frères ou demi-frères. Sur 42 couples de coqs Pp+ (crête en pois) et p+p+ (crête simple), le poids moyen des premiers à 40 semaines dépasse celui des seconds d’environ 250 g, soit 7,8 % du poids moyen du génotype homozygote récessif (P < 0,001). Cette différence provient d’un plus fort gain de poids tardif (après 8 semaines d’âge) et n’existe pas chez les poules adultes d’après des données antérieures. Des recherches ultérieures sont suggérées.
