Aggregatibacter actinomycetemcomitansarcB influences hydrophobic properties, biofilm formation and adhesion to hydroxyapatite BJM
Longo,PL; Ota-Tsuzuki,C; Nunes,ACR; Fernandes,BL; Mintz,K; Fives-Taylor,P; Mayer,MPA.
The regulation of gene expression in the oral pathogen Aggregatibacter actinomycetemcomitans is still not fully elucidated. ArcAB is a two-component system which allows facultative anaerobic bacteria to sense various respiratory growth conditions and adapt their gene expression accordingly. This study investigated in A. actinomycetemcomitans the role of arcB on the regulation of biofilm formation, adhesion to saliva coated hydroxyapatite (SHA) and the hydrophobic properties of the cell. These phenotypic traits were determined for an A. actinomycetemcomitansarcB deficient type and a wild type strain. Differences in hydrophobic properties were shown at early and late exponential growth phases under microaerobic incubation and at late exponential phase under...

Cloning and characterization of the gene encoding the PepF endopeptidase from the aquatic bacterium Caulobacter crescentus BJM
Braz,Vânia S.; Lang,Elza A.S.; Marques,Marilis V..
The metallopeptidases have a very important role in bacteria, being involved in several processes that rely on protein turnover, such as nutrition, degradation of signal peptides, protein localization and virulence. We have cloned and characterized the gene of the metalloendopeptidase PepF from the aquatic bacterium Caulobacter crescentus. The gene upstream of pepF (orf1) encodes a conserved hypothetical protein found in Mycobacterium and Streptomyces. pepF is co-transcribed with the gene downstream (orf3), which encodes a protein that belongs to the ABC1 protein kinase family, suggesting that these two proteins may share a common function in the cell. The C. crescentus PepF protein possesses the conserved HEXGH motif present in zinc binding domains of...

COX-2 gene expression and methylation profile in Sapajus apella as an experimental model for gastric adenocarcinoma Genet. Mol. Biol.
Rosário Pinheiro,Danilo do; Harada,Maria Lucia; Rodriguez Burbano,Rommel Mario; Nascimento Borges,Barbara do.
Abstract Gastric cancer (GC) remains one of the main causes of cancer-related death worldwide. There are two distinct histological types of GC: diffuse and intestinal. The latter is characterized by the presence of pre-neoplastic lesions. One of the most frequently altered enzymes in intestinal GC is COX-2, an important lesion marker. This work aimed to study COX-2 methylation and expression in N-methyl-N-Nitrosurea (MNU)-induced intestinal GC in six Sapajus apella animals. The partial promoter sequence of S. apella COX-2 gene was obtained and used to identify transcription factors and cis-regulatory element binding sites. The COX-2 methylation pattern was assessed using Methylation-Specific PCR (MSP), and expression was analyzed by immunohistochemistry...

Effects of BST and high energy diet on gene expression in mammary parenchyma of dairy heifers R. Bras. Zootec.
Lew,Betina Joyce; Oliveira,Mauro Dal Secco de; Freitas Júnior,José Esler de; Carvalho,Marina Vieira de; Rêgo,Aníbal Coutinho do; Rennó,Francisco Palma.
The objective of this study was to determine the effects of dietary energy and recombinant bovine somatotropin (bST) injection to identify genes that might control mammogenesis. Total RNA was extracted from the parenchymal tissue of 32 heifers randomly assigned to one of four treatments: two diets (a standard diet and a high energy, high protein diet), each with or without bST. To perform microarray experiments, RNA samples were pooled (2 animals/pool) before reverse transcription and labeling with Cy3 or Cy5. A 4-node loop design was used to examine the differential gene expression among treatments using a bovine-specific cDNA microarray (National Bovine Functional Genomics Consortium Library, NBFGC) containing 18,263 unique expressed sequence tags (EST)....

Identification in silico of putative damage responsive elements (DRE) in promoter regions of the yeast genome Genet. Mol. Biol.
Silva,Walkiria Luckwu de Santana; Cavalcanti,Andre Ricardo de Oliveira; Guimarães,Katia Silva; Morais Jr.,Marcos Antonio de.
We report an in silico analysis to identify nucleotide sequence motifs in DNA repair genes that may define a binding site for regulatory proteins during the induction of those genes by mutagens. The damage responsive elements (DRE) weight matrix generated in this analysis was used to search for homologous sequences in the promoter region of all genes, including putative gene and hypothetical open reading frames (ORFs), in the Saccharomyces Genome Data Base (SGD). The results demonstrated that over one third of the yeast genes in the database presented at least one 15-bp sequence in their promoter region with 85% or more of similarity to the DRE consensus sequence. The presence of the DRE sequence in the promoter region of regulatory genes and its high...

Influence of dietary phospholipids on early ontogenesis of fish ArchiMer
Cahu, Chantal; Gisbert, Enric; Villeneuve, Laure A. N.; Morais, Sofia; Hamza, Neila; Wold, Per-avid; Zambonino, Jose-luis.
The aim of this paper is to provide explanations of how dietary phospholipid (PL) globally improves fish larval development, including growth and survival, digestive functions and skeletal development, and to propose optimal PL levels and sources in fish larval diets. Dietary incorporation of 8-12% PL related to dry matter (d.m.) promotes growth and enhanced survival in various species. Marine source PL, incorporating highly unsaturated fatty acids, was most efficient than soybean lecithin. This beneficial effect was explained by an enhancement in digestive functions, assessed by digestive enzyme activities and histomorphology. Nevertheless, 1.5-2.5% highly unsaturated fatty acids related to diet d.m. supplied by PL improved growth, survival and skeletal...

Oxidative stress: molecular perception and transduction of signals triggering antioxidant gene defenses BJMBR
Scandalios,J.G..
Molecular oxygen (O2) is the premier biological electron acceptor that serves vital roles in fundamental cellular functions. However, with the beneficial properties of O2 comes the inadvertent formation of reactive oxygen species (ROS) such as superoxide (O2<FONT FACE=Symbol>·-</FONT>), hydrogen peroxide, and hydroxyl radical (OH<FONT FACE=Symbol>·</FONT>). If unabated, ROS pose a serious threat to or cause the death of aerobic cells. To minimize the damaging effects of ROS, aerobic organisms evolved non-enzymatic and enzymatic antioxidant defenses. The latter include catalases, peroxidases, superoxide dismutases, and glutathione S-transferases (GST). Cellular ROS-sensing mechanisms are not well understood, but a number of...

Role of non-coding RNAs in non-aging-related neurological disorders BJMBR
Vieira,A.S.; Dogini,D.B.; Lopes-Cendes,I..
Protein coding sequences represent only 2% of the human genome. Recent advances have demonstrated that a significant portion of the genome is actively transcribed as non-coding RNA molecules. These non-coding RNAs are emerging as key players in the regulation of biological processes, and act as "fine-tuners" of gene expression. Neurological disorders are caused by a wide range of genetic mutations, epigenetic and environmental factors, and the exact pathophysiology of many of these conditions is still unknown. It is currently recognized that dysregulations in the expression of non-coding RNAs are present in many neurological disorders and may be relevant in the mechanisms leading to disease. In addition, circulating non-coding RNAs are emerging as...

Studies toward the identification of transcription factors in cassava storage root Braz. J. Plant Physiol.
Souza,Cláudia Regina Batista de; Almeida,Elionor Rita Pereira de; Carvalho,Luiz Joaquim Castelo Branco; Gander,Eugen Silvano.
Transcription factors play important roles in several physiological processes. In recent years many transcription factors have been isolated from plants and they are emerging as powerful tools in the manipulation of plant traits. In this work we initiated studies in order to isolate transcription factors from cassava (Manihot esculenta Crantz), an important tropical and subtropical crop. Our results show three kinds of proteins expressed differentially in cassava storage root and immunologically related to the opaque-2 transcription factor from maize. Southwestern experiments showed two proteins capable of interacting in vitro with the DNA sequence of the be2S1 gene from the Brazil nut tree.

The central role of RNA in the genetic programming of complex organisms Anais da ABC (AABC)
Mattick,John S..
Notwithstanding lineage-specific variations, the number and type of protein-coding genes remain relatively static across the animal kingdom. By contrast there has been a massive expansion in the extent of genomic non-proteincoding sequences with increasing developmental complexity. These non-coding sequences are, in fact, transcribed in a regulated manner to produce large numbers of large and small non-protein-coding RNAs that control gene expression at many levels including chromatin architecture, post-transcriptional processing and translation. Moreover, many RNAs are edited, especially in the nervous system, which may be the basis of epigenome-environment interactions and the function of the brain.
