Crassostrea gigas mortality in France: the usual suspect, a herpes virus, may not be the killer in this polymicrobial opportunistic disease ArchiMer
Petton, Bruno; Bruto, Maxime; James, Adele; Labreuche, Yannick; Alunno-bruscia, Marianne; Le Roux, Frederique.
Successive disease outbreaks in oyster (Crassostrea gigas) beds in France have resulted in dramatic losses in production, and subsequent decline in the oyster-farming industry. Deaths of juvenile oysters have been associated with the presence of a herpes virus (OsHV-1 µvar) and bacterial populations of the genus Vibrio. Although the pathogenicity of OsHV-1 µvar, as well as several strains of Vibrio has been demonstrated by experimental infections, our understanding of the complexity of infections occurring in the natural environment remains limited. In the present study, we use specific-pathogen-free (SPF) oysters infected in an estuarine environment to study the diversity and dynamics of cultured microbial populations during disease expression. We observe...

Detection and description of a particular Ostreid herpesvirus 1 genotype associated with massive mortality outbreaks of Pacific oysters, Crassostrea gigas, in France in 2008 ArchiMer
Segarra, Amelie; Pepin, Jean-francois; Arzul, Isabelle; Morga, Benjamin; Faury, Nicole; Renault, Tristan.
Ostreid herpesvirus 1 (OsHV-1) infections have been reported around the world and are associated with high mortalities of the Pacific oyster (Crassostrea gigas). In the summer 2008, abnormal mortality rates ranging from 80% to 100% were reported in France and affected only Pacific oysters. Analyses of oyster samples collected during mortality outbreaks demonstrated a significant detection of OsHV-1 (75% of analysed batches), which appeared stronger than previous years. DNA sequencing based on C and IA regions was carried out on 28 batches of OsHV-1 infected Pacific oysters collected in 2008. Polymorphisms were described in both the C and IA regions and characterized a genotype of OsHV-1 not already reported and termed OsHV-1 μVar. A microsatellite zone...

Development of a polymerase chain reaction for the detection of abalone herpesvirus infection based on the DNA polymerase gene ArchiMer
Chen, M.h.; Kuo, S. T.; Renault, Tristan; Friedman, C. S.; Chang, P. H..
A 5781-base pair (bp) fragment of genomic DNA from the Taiwanese abalone herpesvirus was obtained and showed 99% (5767/5779) homology in the nucleotide sequence and 99% (1923/1926) in the amino acid sequence with the DNA polymerase gene of the abalone herpesvirus strain Victoria/AUS/2007. Homology of the amino acid sequence with the DNA polymerase of ostreid herpesvirus 1 was 30% (563/1856). In this study, a PCR-based procedure for detecting herpesvirus infection of abalone, Haliotis diversicolor supertexta, in Taiwan was developed. The method employed primer sets targeting the viral DNA polymerase gene, and was able to amplify DNA fragments of the expected size from infected samples. Primer sets of 40f and 146r were designed for amplification of an...

Fonds d'Aide à la Recherche et à l'Innovation (FARI) - Décision d'Aide à la Recherche n° 95-07-01 - Rapport final ArchiMer
Renault, Tristan.

Gomes,Christiane Maria Moreira; Giraldo,Paulo César; Gomes,Francis de Assis Moraes; Amaral,Rose; Passos,Mauro Romero Leal; Gonçalves,Ana Katherine da Silveira.
Female genital ulcer is a disease that affects a large number of women, and its etiologic diagnosis can be difficult. The disease may increase the risk of acquiring HIV. Genital ulcer may be present in sexually transmitted diseases (STD) - syphilis, chancroid, genital herpes, donovanosis, lymphogranuloma venereum and other non-STD disorders (NSTD) - Behçet's syndrome, pemphigus, Crohn's disease, erosive lichen planus and others. This study evaluated the clinical-histopathologic-microbiologic characteristics of female genital ulcers. A cross-sectional descriptive prospective study was conducted during a six-month period to investigate the first 53 women without a definitive diagnosis, seeking medical care for genital ulcers at a genital infections...

Les virus infectant les mollusques marins : un exemple d'actualité, les herpèsvirus ArchiMer
Renault, Tristan.
Although viruses have been associated with mortality outbreaks in different mollusc species, little information is available on their exact affiliation and taxonomic position. Herpes viruses have attracted particular attention because of their economic and ecological impact on both farmed and wild marine molluscs over the past 20 years. Investigations based on molecular approaches have been carried out on the herpes virus infecting Pacific oysters in France. Molecular and ultra-structural data were used to classify the virus under the name ostreid herpesvirus 1 (OsHV-1), as the first known species in the family Malacoherpesviridae. Since 2008, massive mortality outbreaks were reported in several Crassostrea gigas oyster farms in France, Ireland, and the...

This study aims to improve understanding of causes of mortalities in oyster Crassostrea gigas in pertuis charentais related to environmental parameters and the kinetics of targeted infectious agents, such as bacteria Vibrio splendidus (group ) and V. aestuarianus and Ostreid herpesvirus OsHV-1. Two sampling campaigns were carried out on the site of Agnas in 2009 and 2010, before, during or following mortality events of spat from March to September. The approach is to detect and quantify in short time step infectious agents from tissues of farmed oysters placed on rack and in various samples taken nearby: native oysters and other species (wild mussels and barnacles), seawater and sediment. With physical and chemical automatic probes placed on the site,...

QTL for resistance to summer mortality and OsHV-1 load in the Pacific oyster (Crassostrea gigas) ArchiMer
Sauvage, Christopher; Boudry, Pierre; De Koning, D. -j.; Haley, C. S.; Heurtebise, Serge; Lapegue, Sylvie.
P>Summer mortality is a phenomenon severely affecting the aquaculture production of the Pacific oyster (Crassostrea gigas). Although its causal factors are complex, resistance to mortality has been described as a highly heritable trait, and several pathogens including the virus Ostreid Herpes virus type 1 (OsHV-1) have been associated with this phenomenon. A QTL analysis for survival of summer mortality and OsHV-1 load, estimated using real-time PCR, was performed using five F-2 full-sib families resulting from a divergent selection experiment for resistance to summer mortality. A consensus linkage map was built using 29 SNPs and 51 microsatellite markers. Five significant QTL were identified and assigned to linkage groups V, VI, VII and IX. Analysis of...

RESCO - REseau d'Observations Conchylicoles : Campagne 2012 ArchiMer
Fleury, Elodie; Bedier, Edouard.

RESCO - Réseau d'observations Conchylicoles : Campagne 2013 ArchiMer
Fleury, Elodie.
The French network RESCO was developed in 2009, following the widespread mortalities in French oysters stocks of Crassostrea gigas that killed a majority of young cultivated oysters in summer 2008. The main objective of this network was to develop a tool of estimation of the mortalities at national scale, through a standardized monitoring of sentinel oysters In 2013, similar batches of spat and 18 month old oysters from different origins were deployed in thirteen sites located all along the French coasts to measure the spatial and temporal evolution of the mortality and growth rates. At the same time, the program provides a high-frequency recording of environmental parameters and the detection of the main infectious agents associated to the oyster...
