A first-generation microsatellite linkage map of the Japanese quail Inra

A linkage map of the Japanese quail (Coturnix japonica) genome was constructed based uponsegregation analysis of 72 microsatellite loci in 433 F2 progeny of 10 half-sib familiesobtained from a cross between two quail lines of different genetic origins. One line wasselected for long duration of tonic immobility, a behavioural trait related to fearfulness,while the other was selected based on early egg production. Fifty-eight of the markers wereresolved into 12 autosomal linkage groups and a Z chromosome-specific linkage group,while the remaining 14 markers were unlinked. Stephens and Donnelly have introduced a simple yet powerful importance sampling scheme for computing the likelihood in population genetic models. Fundamental to the method is an approximation to the conditional probability of the allelic type of an additional gene, given those currently in the sample. As noted by Li and Stephens.. the product of these conditional probabilities for a sequence of draws that gives the frequency of allelic types in a sample is an approximation to the likelihood, and can be used directly in inference. Bread, beer and wine: Saccharomyces cerevisiae diversity reflects human history Inra

Fermented beverages and foods have played a significant role in most societies worldwide for millennia. To better understand how the yeast species Saccharomyces cerevisiae , the main fermenting agent, evolved along this historical and expansion process, we analysed the genetic diversity among 651 strains from 56 different geographical origins, worldwide. Their genotyping at 12 microsatellite loci revealed 575 distinct genotypes organized in subgroups of yeast types, i.e. bread, beer, wine, sake. Challenges of microsatellite isolation in fungi Inra

Although they represent powerful genetic markers in many fields of biology, microsatellites have been isolated in few fungal species. The aim of this study was to assess whether obtaining microsatellite markers with an acceptable level of polymorphism is generally harder from fungi than in other organisms. We therefore surveyed the number,nature and polymorphism level of published microsatellite markers in fungi from the literature and from our own data on seventeen fungal microsatellite-enriched libraries, and in five other phylogroups (angiosperms, insects, fishes, birds and mammals). Characterization of demographic expansions from pairwise comparisons of linked microsatellite haplotypes Inra

This work extends the methods of demographic inference based on the distribution of pairwise genetic differences between individuals (mismatch distribution) to the case of linked microsatellite data. Population genetics theory describes the distribution of mutations among a sample of genes under different demographic scenarios. However, the actual number of mutations can rarely be deduced from DNA polymorphisms. The inclusion of mutation models in theoretical predictions can improve the performance of statistical methods. Nous avons examiné le polymorphisme électrophorétique de 47 locus enzymatiques dans 24 populations naturelles et domestiques de truite commune de France ainsi que la variabilité à 5 locus microsatellites dans 4 de ces populations. La proportion de locus enzymatiques polymorphes sur l’ensemble des échantillons est de 57% et varie de 0 à 37% entre populations. Ce polymorphisme permet de distinguer 2 sous-espèces distinctes occupantrespectivement les bassins atlantique et méditerranéen et dont la distance génétique standard (Nei) s’établit, en moyenne, à 0,10. They have been used as tools for a large number of applications ranging from gene mapping to phylogenetic studies and isolate typing. Genetic differentiation of Anopheles gambiae populations from East and West Africa : comparison of microsatellite and allozyme loci IRD

Genetic variation of #Anopheles gambiae$ was analysed to assess interpopulation divergence over a 6000 km distance using short tandem repeat (microsatellite) loci and allozyme loci. Differentiation of populations from Kenya and Senegal measured by allele length variation at five microsatellite loci was compared with estimates calculated from published data on six allozyme loci (Miles, 1978). The average Wright's F(ST) of microsatellite loci (0.016) was lower than that of allozymes (0.036). Slatkin's R(ST) values for microsatellite loci were generally higher than their F(ST) values, but the average R(ST) value was virtually identical (0.036) to the average allozyme F(ST). Genetic structure and functioning of alien ship rat populations from a Corsican micro-insular complex Inra

The detrimental effects of the introduced ship rat on bird species on the Lavezzi Mediterranean archipelago has led to the decision to eradicate the rodent from the main island, Lavezzu (73 ha), as well as from several neighbouring islets. A genetic study using eight microsatellite markers has revealed some of the dynamics of this rat population. First, it has been shown that the rat population was genetically stable (no departure from Hardy-Weinberg equilibrium and no linkage disequilibrium) and as suggested by paleontological data, established for a long time.
