El maíz (Zea mays L.) de calidad proteínica (QPM) es nutricionalmente superior en comparación con el maíz normal, porque produce semillas con casi el doble de los aminoácidos (aa) lisina (Lys) y triptófano (Trp), esenciales para la síntesis endógena de la vitamina B niacina. Actualmente, la conversión de una línea de maíz normal a una QPM, se acelera mediante la selección asistida por marcadores moleculares (MAS) y métodos de detección química. En este estudio se reporta el desarrollo y validación de marcadores basados en polimorfismos de un solo nucleótido (SNPs) para los loci opaque-2 (o2) y Ask-2, involucrados en la genética de los QPM, así como la interacción de ambos loci para el contenido de 11 aa en el endospermo de dos poblaciones F2 de maíz,...

Evaluation of high quality protein maize (QPM) hybrids obtained by conversion of normal inbred lines. Repositório Alice

Two experimental QPM hybrids obtained by lhe partial conversion of three elite inbred lines by modified backcrossing were evaluated and compared to the normal versions of the same hybrids. Thirteen agronomic and 12 grain quality traits were evaluated. The hybrids were also anaJyzed by SSR markers in order to evaluate the genetic relationship among them. Results demonstrated that the converted QPM hybrids presented a better protein quality and a similar agronomic performance for most of the evaluated traits, including grain yield, in comparison to their normal versions. However, some of the deficiencies generally ascribed to QPM hybrids, such as a higher lodgíng percentage, thicker cobs, and shorter kernels were maintained up to this stage of the conversion...

Studies toward the identification of transcription factors in cassava storage root Braz. J. Plant Physiol.

Transcription factors play important roles in several physiological processes. In recent years many transcription factors have been isolated from plants and they are emerging as powerful tools in the manipulation of plant traits. In this work we initiated studies in order to isolate transcription factors from cassava (Manihot esculenta Crantz), an important tropical and subtropical crop. Our results show three kinds of proteins expressed differentially in cassava storage root and immunologically related to the opaque-2 transcription factor from maize. Southwestern experiments showed two proteins capable of interacting in vitro with the DNA sequence of the be2S1 gene from the Brazil nut tree.

Quality protein maize (QPM) contains the opaque-2 gene for twice tryptophan content along with modifying genes for kernel vitreousness. The QPM hybrid needs the inbred lines containing with opaque-2 gene. Therefore, the objectives of this study were to improve the corn inbred lines with high tryptophan by the opaque-2 gene that detected by molecular marker, and to estimate combing ability of these inbred lines for the yield. Eight lines of BC1S5 containing with opaque-2 gene detected by phi057 were used in the experiment. Tryptophan content in the endosperm of 8 selected lines and 28 of their diallel hybrids were analyzed using glyoxylic method. การเพิ่มปริมาณทริปโตแฟนด้วยยีน opaque-2 ในข้าวโพดข้าวเหนียวร่วมกับการใช้เครื่องหมายโมเลกุลช่วยในการคัดเลือก Thai Agricultural

Crosses were made between waxy corn and quality protein corn as female and male parents, respectively. In segregating progenies of two crosses, Kwi no.1 x PopII no.53 and Kwi no.9 x PopII no.53, selfing and backcrossing methods were used and then marker-assisted selection of opaque-2 gene (o2o2) by phi057 and umc1066 and of waxy (wxwx) by phi022 and phi061 were applied to obtain the opaque-waxy corn (o2o2wxwx). For the first step of MAS, phi057 and umc1066 were employed to detect the opaque-2 gene (o2o2) in both of BC1S1 and S1 generations. The genotypic ratio of opaque-2 gene (O2O2:O2o2:o2o2) was 37:46:31 for BC1S1 and 42:146:42 for S1 from the cross of Kwi no.1 x PopII no.53 while it was 41:51:41 for BC1S1 and 41:169:35 for S1 from the cross of Kwi no.9...
