Coffee is one of the main agricultural commodities in the country, and it is important to conservation of plant material for breeding programs. Cryopreservation is a promising alternative for preserving in the long-term the germplasm of species considered recalcitrant. However, studies should be performed to achieve maximum survival of seedlings after immersion in liquid nitrogen. The objective of this work was to find a cryopreservation protocol for storing seeds of Coffea canephora, studying two methods of cryopreservation, slow and fast cooling. Seeds of the Coffea canephora species are considered more recalcitrant than those of the Coffea arabica species. They tolerate partial dehydration and they cannot be stored in conventional seed banks at -18ºC because they are sensitive to freezing temperatures. Cryopreservation is a reliable method for storing seeds with these characteristics for long periods. However, preliminary studies are necessary to determine ideal storage conditions. Cryopreservation of the mangaba tree (Hancornia speciosa Gomes): a protocol for long-term storage Agronomy

The aim of this study was to evaluate the efficiency of vitrification and droplet vitrification for the cryopreservation of Hancornia speciosa shoot tips. The shoot tips were subjected to four different periods of exposure (15, 30, 45 and 60 min.) to plant vitrification solution 2 (PVS2) before plunging into liquid nitrogen. We evaluated the regrowth of H. speciosa shoot tips that were cryopreserved by the classical vitrification technique and by the droplet vitrification technique. Araucaria angustifolia seeds are recalcitrant, and their metabolism remains high during storage. This research aimed to describe the initiation of germination in A. angustifolia seeds during storage in order to standardize the assessment of physiological quality and to promote seed conservation. Seeds were collected from two populations and stored for 270 days in the natural laboratory environment and cold chamber. The remarkably short storability of recalcitrant seeds has prevented their inclusion in programs of ex situ conservation. The causes of their desiccation sensitivity and rapid decline in viability during storage are not fully elucidated yet. In this study the highly recalcitrant, fully viable embryos of the tree species Inga vera subsp. affinis were stored under various conditions and analyzed physiologically and cytologically at intervals, in order to obtain more insights in their loss of viability during storage. Com o objetivo de identificar a tolerância à dessecação de sementes de Euterpe espiritosantensis, dois lotes de frutos foram colhidos na coleção de palmeiras do Instituto Agronômico de Campinas, no município de Ubatuba, estado de São Paulo e transportados em embalagem impermeável até a Faculdade de Ciências Agronômicas/UNESP, município de Botucatu, estado de São Paulo, onde foram despolpados. As sementes foram submetidas à secagem em sílica gel a partir das testemunhas não desidratadas (51,4% e 46,6% de teor de água), retirando-se amostras a cada 12 ou 24 horas.
