Dispersal of Cotesia flavipes in sugarcane field and implications for parasitoid releases Bragantia

Diatraea saccharalis Fabr. (Lepidoptera: Crambidae) is a major sugarcane pest in Brazil. The management of infested areas is based on the release of Cotesia flavipes (Cameron) (Hymenoptera: Braconidae), a parasitoid of D. saccharalis larvae, but there are doubts about the effectiveness of C. flavipes, primarily regarding its rate of dispersal in sugarcane fields. Thus, the objective of this study was to evaluate the dispersal of C. flavipes in a sugarcane field and suggest a release method that provides higher parasitoid efficiency. The migration rate of esterases and their substrate specificity for 4-methylumbelliferyl esters (acetate, propionate, and butyrate) and alpha- and beta-naphthyl esters were analyzed in Diatraea saccharalis by starch gel electrophoresis. Substrate preference of esterases was observed with Est-2 and Est-8 isozymes showing substrate specificity for 4-methylumbelliferyl esters and Est-4 isozyme showing specificity for 4-methylumbelliferyl butyrate and alpha-naphthyl butyrate. Horizontal transmission and effect of the temperature in pathogenicity of Beauveria bassiana against Diatraea saccharalis (Lepidoptera: Crambidae) BABT

The aim of this work was to evaluate the effect of temperature on the pathogenicity of Beauveria bassiana against Diatraea saccharalis and evaluate the horizontal transmission capacity among the sugarcane borer larvae. The strains were most pathogenic at 26ºC, followed by 32ºC than at 20ºC, with respective mortalities of 100, 50 and 30.3%. The lethal time was also affected by the temperature, with the shortest LT 50 observed at 26ºC (2.2 days) and the longest at 20ºC (16.2 days). Induction of a gloverin-like antimicrobial polypeptide in the sugarcane borer Diatraea saccharalis challenged by septic injury BJMBR

Diatraea saccharalis (Fabricius, 1794) (Lepidoptera: Crambidae) is an important pest for Brazilian sugarcane. In the present study, we detected two distinct spots in hemolymph from septic injured larvae (HDs1 and HDs2), which are separated by 2DE gel electrophoresis. Both spots were subjected to in-gel tryptic digestion and MALDI-TOF/TOF analysis, which revealed the sequence VFGTLGSDDSGLFGK present in both HDs1 and HDs2. This sequence had homology and 80% identity with specific Lepidoptera antimicrobial peptides called gloverins. Analyses using the ImageMaster 2D software showed pI 8.94 of the HDs1 spot, which is similar to that described to Hyalophora gloveri gloverin (pI 8.5). The sugarcane borer, Diatraea saccharalis (Fabr.) is the main sugarcane pest in Brazil. In the State of São Paulo, the main active population control of D. saccharalis is by inundative releases of the exotic parasitoid, Cotesia flavipes (Cam.). Morphological regional differences of epithelial cells along the midgut in Diatraea saccharalis Fabricius (Lepidoptera: Crambidae) larvae Neotropical Entomology

The sugarcane borer, Diatraea saccharalis Fabricius, is a pest to sugarcane and many other crops. This work aims to characterize morphological variability in the epithelial cells (columnar, goblet and regenerative) along the midgut of D. saccharalis larvae. Fragments of the midgut (anterior, middle and posterior regions) were fixed and processed by light and scanning electron microscopy. There are both cytochemical and ultrastructural differences in the morphology of the epithelial cells, depending on their localization along the midgut. Diatraea saccharalis (Fabr.) (Lepidoptera: Pyralidae) spends the largest part of its life cycle inside the stalk of the host plant,which provides protection against the action of conventional control methods. Biological control has been considered a viable alternative to control this pest in sugarcane (Saccharum officinarum L.) and corn (Zea mays L.), two pest preferential hosts. Commercial sorghum hybrids and Embrapa's lines were evaluated regarding resistance to sugarcane borer. Two field experiments were conducted in Sete Lagoas, MG, under natural infestation, during the second cropping season in a randomized complete block with 3 replicates. The plants were evaluated regarding to stand, maturity cycle, plant height, and number of broken or lodging stalks and yield. The borer incidence, number of galleries, number of healthy and damaged nodes were also evaluated to calculate the Infestation Index and Intensity Infestation Index. Resistance of sugarcane cultivars to Diatraea saccharalis PAB

The objective of this work was to evaluate the oviposition preference of Diatraea saccharalis and the effect of ten sugarcane cultivars on larval development. Oviposition preference was assessed under greenhouse conditions by three releases of couples of moths, with subsequent counting of egg masses and eggs per plant. In order to evaluate the effect of the cultivars on larval development, each plant was infected with about 150 eggs, and, 29 days later, the total number of internodes, number of bored internodes, number of life forms found, larval and pupal weight and length, and the width of larval head capsule were evaluated. Sequence and analysis of the mitochondrial DNA control region in the sugarcane borer Diatraea saccharalis (Lepidoptera: Crambidae) BABT

This study aimed at the sequence and analysis of the mtDNA control region (CR) of the Diatraea saccharalis. The genome PCR amplification was performed using the complementary primers to the flanking regions of Bombyx mori CR mitochondrial segment. The sequencing revealed that the amplified product was 568 bp long, which was smaller than that observed for B. mori (725 bp). Within the amplified segment, a sequence with 338 nucleotides was identified as the control region, which displayed a high AT content (93.5%). The D. saccharalis mtDNA CR multiple sequence alignment analysis showed that this region had high similarity with the Lepidoptera Cydia pomonella.

This work aimed to evaluate the effects of Metarhizium anisopliae on Diatraea flavipennella and investigate their immune response. Was observed the effect of M. anisopliae against larvae of D. flavipennella sprayed at concentrations of 103, 104, 105 conidia / mL, in which showed differences relative the larval period, extending up to 72.0 days in treatment and 25.0 days in the control. Ultrastructure of the Lyonet's glands in larvae of Diatraea saccharalis Fabricius (Lepidoptera: Pyralidae) Biocell

The Lyonet's gland is found in Lepidoptera larvae, close to the excretory duct of the silk gland. The role played by this gland is still uncertain. This work aims to describe the ultrastructure of the Lyonet's gland in Diatraea saccharalis larvae, offering suggestions regarding its possible function. The insects were reared under laboratory-controlled conditions. The glands were conventionally prepared for transmission (TEM) and scanning (SEM) electron microscopy. SEM showed that Lyonet's glands are paired small structures located in the ventral side of the head. They are composed by clustered long cells resembling leaves.
