Acute metal stress in Populus tremula x P. alba (717-1B4 genotype): leaf and cambial proteome changes induced by Cd2+ Inra

The comprehension of metal homeostasis in plants requires the identification of molecular markers linked to stress tolerance. Proteomic changes in leaves and cambial zone of Populus tremula×P. alba (717-1B4 genotype) were analyzed after 61 days of exposure to cadmium (Cd) 360 mg/kg soil dry weight in pot-soil cultures. The treatment led to an acute Cd stress with a reduction of growth and photosynthesis. Cd stress induced changes in the display of 120 spots for leaf tissue and 153 spots for the cambial zone. It involved a reduced photosynthesis, resulting in a profound reorganisation of carbon and carbohydrate metabolisms in both tissues. Influence of exogenous L-proline on embryogenic cultures of larch (Larix leptoeuropaea Dengler), sitka spruce (Picea sitchensis (Bong.) Carr.) and oak (Quercus robur L.) subjected to cold and salt stress Inra

Des cultures embryogènes de mélèze hybride, d'épicéa de sitka et de chêne ont été soumises à différentes conditions de culture et leur croissance étudiée en fonction de l'ajout de L-proline au milieu de culture (1 mM, 10 mM, et 100 mM). Si des températures basses (4 °C) inhibent totalement la croissance des cultures, celle-ci redevient partiellement normale en présence de proline. Nos résultats montrent que les cultures non seulement survivent à de basses températures mais sont aussi capables de croître activement au cours de la durée d'application du froid. De même, la croissance est inversement corrélée à la concentration en sel avec sa complète inhibition en présence de 200 mM de NaCl. In the tropics, maize ( Zea mays L.) is often grown under low N conditions. Information on the respective role of N uptake and partitioning at anthesis in determining grain yield under low N is scarce. Senescence traits have been proposed as secondary traits to select for low N tolerance, but the stability of their association with yield under different environmental conditions has been rarely described. Overproduction of proline in transgenic hybrid larch (Larix x leptoeuropaea (Dengler) cultures renders them tolerant to cold, salt and frost Inra

Cold, salt and frost are important environmental stresses in forest trees and may significantly reduce productivity. Elevated levels of proline are associated with these stresses and may help alleviate their effects. Transgenic hybrid larch Larix leptoeuropaea has been produced expressing a Vigna aconitifolia gene for pyrroline 5-carboxylate synthase, the rate-limiting step in proline synthesis. Embryogenic masses of hybrid larch were co-cultivated with Agrobacterium tumefaciens harbouring a binary vector expressing the gene. The integration of the gene into the plant genome was confirmed by Southern blot and by proline content analysis. There was an approximately 30-fold increase in proline level in transgenic tissue compared to non-transformed controls....

Stress tolerance of plants is conditioned by their ability to preserve the function of theirtissues and cells. The mechanisms involved in the metabolism defence partly depend on thephysiological status and on the stress sensing and signalling pathways.
