Background: Whole-genome physical maps facilitate genome sequencing, sequence assembly, mapping of candidate genes, and the design of targeted genetic markers. An automated protocol was used to construct a Vitis vinifera 'Cabernet Sauvignon' physical map. The quality of the result was addressed with regard to the effect of high heterozygosity on the accuracy of contig assembly. Its usefulness for the genome-wide mapping of genes for disease resistance, which is an important trait for grapevine, was then assessed. Results: The physical map included 29,727 BAC clones assembled into 1,770 contigs, spanning 715,684 kbp, and corresponding to 1.5-fold the genome size. Anatomical, histological, and histochemical changes in grape seeds from Vitis vinifera L. cv cabernet franc during fruit development Inra

Cabernet franc berries were sampled at five stages from berry set to harvest from an experimental vineyard in mid Loire Valley. Seeds were collected from representative berries in term of stage of development. The evolution of seed was followed both macro- and microscopically. For microscopy analysis, seeds were cut, put in a fixation solution, and cut into thin sections with a microtome. Five staining solutions were used for each seed sample: toluidine blue O, phloroglucinol, periodic acid-Schiff's reagent and naphtol blue black, vanillin, and p-dimethylaminocinnamaldehyde. Soil and climate are major constituents of the French notion of Terroir. This concept implies that there is a strong relationship between the composition of the grape, the characteristics of the wine and the territory of production. To study this link, a new method of characterization of the Terroir, including geological and pedological factors, was investigated. It uses a field model based on depth and clay content of soil, together with the degree of weathering of the parent rock. This is a critical step in accessing all the genes of this species and increases the chances of exploiting the natural genetic diversity through association genetics. Several studies have shown that the proanthocyanidins are responsible for some major flavour properties of the red wines such as colour, bitterness and astringency. Nevertheless, their synthesis and properties are still misunderstood. Some studies have suggested that the proanthocyanidin content is set before veraison. This paper describes the setting of the proanthocyanidins, until the veraison stage, and the relations that exist with the environment. La modélisation présentée ici a subi une double validation : expérimentale, par exploitation de bassins fortement contrastés en végétation, et méthodologique, par exploitation simultanée d'un modèle régional (descriptif synthétique) à démarche ascendante (QdF) et d'un modèle conceptuel global à démarche descendante (GR3J). Le bassin versant de l'Ardières à Beaujeu (54,5 km2) est un petit bassin versant montagneux, recouvert sur 90 % de sa superficie d'une forêt de sapins, de feuillus ainsi que de prairies et sur les 10 % restants de vignobles. A proximité de celui-ci, le bassin versant de la Vauxonne à Buyon (49,3 km2), dont 70 % de la superficie sont plantés en vigne et 30 % sont occupés par la forêt et les prairies, présente des crues bien différenciées,...
