Characterization of cellulolytic activities of Bjerkandera adusta and Pycnoporus sanguineus on solid wheat straw medium Electron. J. Biotechnol.

Cellulolytic properties of two white rot fungi, Bjerkandera adusta and Pycnoporus sanguineus, cultivated on wheat straw agar medium, were characterized and compared. Optimal growing parameters for maximum enzyme production for both fungi were wheat straw medium pH 5 and 28ºC. B. adusta showed, on the 6th day of culture, carboxymethylcellulose (CMC)ase activity levels 1.6 times higher than maximal P. sanguineus activity, achieved on the 8th day. B. adusta supernatants also displayed higher activity levels towards xylan (3.6-fold) compared to those of P. sanguineus. Degradación in vitro de la pared celular, variables ruminales y actividad enzimática de miroorganismos adheridos a sólidos del complejo Lolium-Festuca. Colegio de Postgraduados

En este estudio se evaluó el efecto del complejo Lolium-Festuca (CL-F), cosechado a 28 (T1) y 35 d (T2) de rebrote, en la desaparición in vitro de la materia seca (DIVMS), cinética de producción de gas (CPG), y en la actividad de carboximetilcelulasas (CMCasas) y de xilanasas de microorganismos adheridos a sólidos a las 12, 16, 24, y 48 h de incubación. La degradación del tejido foliar a las 12, 24, y 48 h de incubación se documentó mediante Microscopía de Electrónica de Barrido (MEB); la lignina se detectó con la reacción floroglucinol-HCl. El modelo logístico y el procedimiento NLIN se usaron para obtener los datos de CPG. Digestibilidad in situ de dietas con harina de nopal deshidratado conteniendo un preparado de enzimas fibrolíticas exógenas. Repositório Alice

Se evaluó el efecto de un preparado de enzimas fibrolíticas exógenas (celulasas y xilanasas) en la degradabilidad in situ de la materia seca (DisMS), fibra detergente neutro (DFDNr) y fibra detergente ácido residual (DFDAr), en dietas altas o bajas en harina de nopal deshidratado. Se aplicaron concentraciones de 0, 1, 2 y 3 g de enzima por kilogramo de materia seca al inicio y 24 horas antes de la degradación in situ. Se determinó la concentración de ácidos grasos volátiles totales y de nitrógeno amoniacal a las 0, 3, 6, 9, 12 y 24 horas después de aplicarse la enzima. No se observaron efectos en DisMS, DFDNr y DFDAr; la aplicación al inicio de la degradación in situ mostró valores más altos que a 24 horas para DisMS y DFDNr, pero fue menor para DFDAr. No...

Extremely thermophilic microorganisms and their polymer-hidrolytic enzymes Rev. Microbiol.

Thermophilic and hyperthermophilic microorganisms are found as normal inhabitants of continental and submarine volcanic areas, geothermally heated sea-sediments and hydrothermal vents and thus are considered extremophiles. Several present or potential applications of extremophilic enzymes are reviewed, especially polymer-hydrolysing enzymes, such as amylolytic and hemicellulolytic enzymes. The purpose of this review is to present the range of morphological and metabolic features among those microorganisms growing from 70oC to 100°C and to indicate potential opportunities for useful applications derived from these features.

Los residuos agrícolas son los desechos obtenidos del proceso de la producción de los alimentos y fibras vegetales, y su utilización está limitada principalmente por su bajo contenido de proteína y elevados niveles de lignina. Los hongos basidiomicetos como Pleurotus spp disminuyen los componentes de la pared celular debido a su capacidad para sintetizar enzimas fibrolíticas. Los pre-tratamientos alcalinos hidrolizan la hemicelulosa y lignina de materiales fibrosos. Purification and characterization of xylanases from Trichoderma inhamatum Electron. J. Biotechnol.

Background Two xylanases, Xyl I and Xyl II, were purified from the crude extracellular extract of a Trichoderma inhamatum strain cultivated in liquid medium with oat spelts xylan. Results The molecular masses of the purified enzymes estimated by SDS-PAGE and gel filtration were, respectively, 19 and 14 kDa for Xyl I and 21 and 14.6 kDa for Xyl II. The enzymes are glycoproteins with optimum activity at 50°C in pH 5.0-5.5 for Xyl I and 5.5 for Xyl II. The xylanases were very stable at 40°C and in the pH ranges from 4.5-6.5 for Xyl I and 4.0-8.0 for Xyl II. The ion Hg2+ and the detergent SDS strongly reduced the activity while 1,4-dithiothreitol stimulated both enzymes. Thermal stability of xylanases produced by Aspergillus awamori BJM

The effect of temperature on the activity and stability of endoxylanase and beta-xylosidase from Aspergillus awamori was investigated. The growth of A. awamori in milled sugar cane bagasse produced predominantly extracellular endoxylanase (30 U/ml) and lower amounts of beta-xylosidase (1.3 U/ml). Grown in sugar cane bagasse as the principal carbon source, the microorganism produced a quite stable beta-xylosidase in a temperature range of 35-55°C, but it exhibited a lower thermostable endoxylanase. The thermostability of endoxylanase was enhanced through addition of polyhydric alcohols, mainly 2 M xylitol and sorbitol solutions.
