El presente trabajo constituye el primer reporte sobre el estudio del sistema proteolítico del hongo Moniliophthora roreri, agente causal de la moniliasis del cacao. El trabajo inició con la identificación de la cepa aislada a partir de frutos de cacao infectados. La amplificación y secuenciación de un fragmento del gen 18S rDNA, permitió la identificación molecular de la cepa aislada como M. roreri, designada como MRO1. A continuación, se evaluó el crecimiento celular de M. roreri en los medios de cultivo líquidos mineral y V8 enriquecido, indicando que en este último se obtiene mayor peso seco en mg mL-1. La determinación de los niveles de las actividades proteolíticas de aspartil proteasa (mrAP), aminopeptidasa (mrAPE), carboxipeptidasa (mrCP) y dipeptidil aminopeptidasa (mrDAP), fue evaluada siguiendo las cinéticas de producción de proteasas desde las 0 hasta las 144h de incubación. Dichas determinaciones se realizaron a partir de extractos enzimáticos obtenidos de la fracción intracelular y extracelular empleando sustratos específicos. En ambos medios ensayados, se encontró actividad enzimática intracelular y extracelular de mrAP, mrAPE, mrDAP y mrCP las cuales podrían participar en diversos eventos celulares, y en el caso específico de las proteasas extracelulares, podrían participar en la fitopatogénesis de M. roreri. Los ensayos con diferentes inhibidores específicos, indicaron que es probable que cada una de las actividades enzimáticas de mrAP, mrAPE, mrDAP y mrCP, sean codificadas por múltiples genes, como se ha descrito en otros hongos. Los ensayos de localización subcelular indicaron que a nivel intracelular las enzimas mrAPE y mrDAP se encuentran asociadas mayoritariamente a la fracción citoplásmica, mientras que la mrCP se encuentra asociada mayoritariamente a la fracción membranal, en ambos medios de cultivo.__________The present work is the first report on the study of the protelic system of the phytopathogenic fungus Moniliophthora roreri causing frosty pod rot (moniliasis disease). The work beginning with the identification of the isolated strain from infected cocoa pods. The amplification and sequencing of a fragment of the gene 18S rDNA, allowed to the molecular identification of the isolated strain as M. roreri, designated as MRO1. Later, the cellular growth of M. roreri was evaluated in mineral and enriched V8 liquid medium, indicating that in this last, one greater dry weight in mg mL-1 is obtained. The determination of the levels of the proteolytic activities of aspartil protease (mrAP), aminopeptidase (mrAPE), carboxypeptidase (mrCP) and dipeptidil aminopeptidase (mrDAP), was evaluated following the kinetic production of proteases from the 0 to 144 h of incubation. These determinations were made from enzymatic extracts obtained of the intracellular and extracellular fraction using specific substrates. In both tried media, was intracellular and extracellular enzymatic activity in mrDAP mrAP, mrAPE, mrDAP and mrCP, in M. roreri which could participate in diverse cellular events, and in the specific case of the extracellular proteases, could participate in the pathogenesis process. The tests with different specific inhibitors indicated that it is probable that each one of the enzymatic activities of mrAP, mrAPE, mrDAP and mrCP, are codified by multiple genes, since has been described in other fungi. The tests of subcellular location indicated that at intracellular level the enzymes mrAPE, mrDAP are associate mainly to the citoplasmic fraction, whereas mrCP is associate mainly to the membranal fraction, in both means of culture.
