Background: Although bivalves are among the most-studied marine organisms because of their ecological role and economic importance, very little information is available on the genome sequences of oyster species. This report documents three large-scale cDNA sequencing projects for the Pacific oyster Crassostrea gigas initiated to provide a large number of expressed sequence tags that were subsequently compiled in a publicly accessible database. This resource allowed for the identification of a large number of transcripts and provides valuable information for ongoing investigations of tissue-specific and stimulus-dependant gene expression patterns. These data are crucial for constructing comprehensive DNA microarrays, identifying single nucleotide... |

We investigated the role of oyster gonadal TGF beta (og-TGF beta) in the reproduction of Crassostrea gigas, using an in vivo RNA interference approach. We designed double-stranded RNA targeting og-TGF beta, which is specifically expressed in the somatic cells surrounding germ cells in the gonad of both male and female oysters. In vivo injection of this og-TGF beta dsRNA into the gonad led to knock-down phenotypes for both sexes, with significant reduction (77.52% relative to controls) of the gonad area, lowered reproductive effort and germ cell under-proliferation. Interestingly, half of the injected females halted their vitellogenesis, since we were only able to observe pre-vitellogenic oocytes. In addition, apoptotic germ cells and haemocytes infiltrated... |

To study the complex and non identified host causes of summer mortality in C. gigas, divergent selection criteria based upon summer survival have been applied to produce resistant (R) and susceptible (S) oysterfamilies(Samain et al., 2007). A previous analysis by suppression-subtractive hybridization has characterized 46 differentially-expressedcDNAsbetween R and S families(Huvet et al., 2004). The present study reports the screening, using nylon array, of the 46 oyster SSHcDNAsfrom R and S selected families. Among the few differentially expressed genes characterized between R and S samples, one encodes a Transforming-Growth-Factor-β-related (TGFβ) protein. Members of the TGFβsuperfamily are critical growth factors regulating a variety ofimportant... |

Through differential screening between oyster families selected for high and low summer survival, we have characterized a new transforming growth factor-beta (TGF-beta) superfamily member. This novel factor, named oyster-gonadal-TGF beta-like (og-TGF beta-like), is synthesized as a 307 amino acid precursor and displays 6 of the 7 characteristic cysteine residues of the C-terminal, mature peptide. Sequence comparison revealed that og-TGF beta-like has a low percentage of identity with other known TGF-beta superfamily members, suggesting that og-TGF beta-like is a derived member of this large superfamily. Real-time PCR (RT-PCR) analysis in different oyster tissues showed that og-TGF beta-like is specifically expressed in both male and female gonads, at... |

Occupying a prime position in the French coastal landscape, oyster farming is the leading contributor to the French aquaculture sector with an estimated production of 85,000 tonnes in recent years. However, since 2008, this species has been affected by recurrent massive mortality episodes triggered by the presence of viral infectious agents (of the type Herpes virus OsHV-1 μVar) in conjunction with environmental factors (i.e. temperature rise). This viral infection induces an alteration of the antimicrobial defenses and a destabilization of its microbiota. This process leads to secondary infection by opportunistic bacteria that are potentially pathogenic and multiply rapidly, causing irreversible tissue damage leading to the death of the animal. At the... |

Of all environmental factors, seawater temperature plays a decisive role in triggering marine diseases. Like fever in vertebrates, high seawater temperature could modulate the host response to pathogens in ectothermic animals. In France, massive mortality of Pacific oysters, Crassostrea gigas, caused by the ostreid herpesvirus 1 (OsHV-1) is markedly reduced when temperatures exceed 24°C in the field. In the present study we assess how high temperature influences the host response to the pathogen by comparing transcriptomes (RNA sequencing) during the course of experimental infection at 21°C (reference) and 29°C. We show that high temperature induced host physiological processes that are unfavorable to the viral infection. Temperature influenced the... |

The intertidal zone is characterized by a sharp vertical gradient of environmental stress, which structures species distribution and their interactions. Few studies, however, have examined the influence of tidal height on host–pathogen interactions. Here, we investigated how the tidal height influence outbreak of the Ostreid herpesvirus type 1 (OsHV-1) affecting the Pacific oyster. A volunteer network composed of 20 oyster growers monitored the survival of 28 batches of oysters during an epizootic event in Southern Brittany, France. Oysters were spat from wild collection or hatchery production. The sampling sites were spread over a 150-km2 area with a tidal height ranging from 0.98 to 2.90 m. Concomitantly, we followed survival of oyster spats in relation... |

Summer mortality of the Pacific oyster Crassostrea gigas is the result of a complex interaction between oysters, their environment, and pathogens. Heredity appears to be a major factor determining the sensitivity of oysters to summer mortality, allowing resistant (R) and susceptible (S) lines to be produced. We conducted genome-wide expression profiling of R and S gonads during the 3-month period preceding a summer mortality event, using a cDNA microarray that we designed. ANOVA analysis revealed that 34 genes were differentially expressed between R and S lines on four dates preceding the mortality event. Annotation of some of these genes highlights reproduction and its allocation and antioxidant defenses as the main pathways that operate differentially... |

The transforming growth factor (TGF)-beta superfamily is a group of important growth factors involved in multiple processes such as differentiation, cell proliferation, apoptosis and cellular growth. In the Pacific oyster Crassostrea gigas, the oyster gonadal (og) TGF-beta gene was recently characterized through genome-wide expression profiling of oyster lines selected to be resistant or susceptible to summer mortality. Og TGF-beta appeared specifically expressed in the gonad to reach a maximum when gonads are fully mature, which singularly contrasts with the pleiotropic roles commonly ascribed to most TGF-beta family members. The function of og TGF-beta protein in oysters is unknown, and defining its role remains challenging. In this study, we develop a... |

In summer, oysters suffer mortality that decimates this year about 60% of the population in France. We previously demonstrated a genetic basis for summer survival thatallowed us to produce resistant (R) and susceptible (S) oyster familiesby divergent selection (Samainet al., 2007). At the physiological level, R families can survive summer mortality because they are reproductively less active than S families (Fleury et al., 2008).In order to identify the molecular basis of the summer survival,a transcriptomicapproach on R and S families was performed using microarray analysis and 34 differentially-expressed genes were identified between R and S in the gonad (Fleury et al., in prep). Among these genes, we found an EST encoding a neuropeptide Y related... |

A transplant is a complex surgical procedure performed to obtain a pearl after about 12 to 18 months. One of the key steps in this procedure is the transfer of the graft (piece of mantle) from the donor oyster. A functional relationship exists between the coating nature of the pearl and the cellular structure of the pearl bag portion of the graft. We have identified two genetic markers of biomineralisation in Pinctada margaritifera, the pearl oyster: Perline and Calcine, responsible for secretion of aragonite and calcite, respectively. After isolating and sequencing a portion of each of these genes, we verified the specificity of these sequences using molecular methods (RT-PCR, in situ hybridisation) and biocomputing methods (alignments, phylogenetic... |
