Toxicity assessment of peptaibols and contaminated sediments on Crassostrea gigas embryos ArchiMer

Peptaibols are known membrane-modifying peptides that were recently detected in marine sediments and mussels collected from a shellfish farming area (Fier d'Ars, Atlantic coast, France). In this investigation, embryotoxicity bioassays with oysters (Crassostrea gigas) were performed to assess acute toxicity of alamethicin and different groups of peptaibols produced by a Trichoderma longibrachiatum strain isolated from marine environment. C gigas embryos appeared very sensitive to all the metabolites examined with higher toxic effects for long-sequence peptides (EC50 ranging from 10 to 64nM). Estimation de la qualité biologique de l'eau sur le littoral atlantique du Sud-Ouest de la France à l'aide des tests de croissance des larves de Mytilus edulis ArchiMer

The quality of the seawater off the southwestern Atlantic coast of France was assessed by studying the effects of the water from three sites (Arcachon Bay, Mimizan and Capbreton) on larvae of the mussel (Mytilus edulis). Mussel larvae were raised in water samples taken at all three sites on March 12, 1991 and larval growth was monitored during 10 days. The results indicate that the water quality was best in the Bay of Arcachon, followed by Mimizan, whereas the water off Capbreton was possibly contaminated by pollutants.

In vitro test-based comparison of pesticide-induced sensitivity in marine and freshwater phytoplankton ArchiMer

This study aims to assess the effects of two pesticides, namely the insecticide carbofuran and the herbicide isoproturon, on monospecifically cultivated marine and freshwater phytoplankton according to standard methods. In the presence of pesticide, growth rates were lower in marine species Chaetoceros gracilis and Phaeodactylum tricornutum than in freshwater species Chlorella vulgaris and Selenastrum capricornutum after 72 hours. The EC50 values were obtained with the REGTOX Macro software, and the NEC values by applying the DEBtox model.

Taux de réussite du développement embryonnaire de la moule et de l'huitre creuse en présence d'eau ou de sédiment cotier de la baie de Seine ArchiMer

Une étude préalable à la mise en route, dans le cadre du RNO, d'un programme de surveillance des effets biologiques a été menée en recherchant l'effet de la présence d'eau ou de sédiment côtier de la baie de Seine sur le développement embryonnaire de la moule et de l'huître creuse. Les résultats montrent que ce test est très intéressant à proposer dans le cadre d'une surveillance biologique, après mise au point spécifique du protocole expérimental pour le suivi du sédiment.

Marine water quality assessment using transplanted oyster larvae ArchiMer

Active bio-monitoring in terms of biornarkers was attempted using Crassostrea gigas larvae produced in the laboratory and transplanted using special containers to two sites at the entrance (A) and inner part (P) of the harbour of Arcachon (French Atlantic Coast). The larvae were kept in the medium for 48 h. Cette synthèse bibliographique, non exhaustive, a été réalisée dans le cadre du projet "rade de Brest" grâce à la collaboration d'Antoine JUDAS, contractuel de la CUB en 1995 (premier trimestre). Elle fait partie du projet "Critères physiologiques de la qualité du milieu" réalisé au laboratoire d'Ecotoxicologie de la Direction de l'Environnement Littoral de Brest. Ce document se présente sous la forme de listes dans lesquelles sont reportés les effets létaux et sublétaux de substances chimiques. Pour chaque substance sont indiqués: la forme chimique de la substance, l'espèce testée, le type d'essai réalisé, les concentrations entraînant 50% de létalité (LC50) ou 50% d'effet sublétal (CE50) ainsi que la référence des publications d'où proviennent les valeurs.

Toxicity of Epoxiconazole to the Marine Diatom Chaetoceros calcitrans: Influence of Growth Conditions and Algal Development Stage ArchiMer

The triazole fungicide epoxiconazole is extensively used to control fungi on crops and may present some potential risk from runoff on coastal ecosystems located close to agricultural areas. Phytotoxicity assessments were conducted on the marine diatom Chaetoceros calcitrans using both the active ingredient and its formulated product (Opus). The 3-day EC50 using cell count was 2.31 mg/L for epoxiconazole active ingredient and 2.9 mu g/L for epoxiconazole-formulated. The fungicide produced an increase of cellular volume, pigment (chlorophylls a, c, and carotenoids) content, ATP synthesis, and rates of photosynthesis and respiration. Le projet à pour but de modéliser la dispersion des immersions des boues de dragage, à partir de modèles numériques de manière à pouvoir évaluer le devenir de la charge particulaire et des contaminants associés. Les résultats des expérimentations en laboratoire montrent, qu'à l'échelle représentative du temps de séjour des particules dans la colonne d'eau du champ proche (quelques heures), on ne constate pas de solubilisation des métaux. Determination of peptaibol trace amounts in marine sediments by liquid chromatography/electrospray ionization-ion trap-mass spectrometry ArchiMer

Extraction followed by reverse phase liquid chromatography (LC)/electrospray ionization-ion trap-mass spectrometry (ESI-IT-MS) analysis has been successfully developed for the determination of peptaibols, fungal toxic metabolites, in marine sediments. Spiking experiments showed that the mean recovery of target compounds exceeded 85% at a spiking level of 10 ng/g of sediment (wet weight). Detection and quantification limits were 250 and 830 pg/g of sediment, respectively. The method developed constituted the first sensitive assay for quantification of peptaibol trace amounts in a natural environment. A concentration of 5 ng/g in sediment samples collected from Fier d'Ars was found.

Evaluation of biomarkers in oyster larvae in natural and polluted conditions ArchiMer

Crassostrea gigas D-shaped larvae were subjected to different conditions of temperature and salinity for 24 h and four biomarkers (acetylcholinesterase (AChE) activity, thiobarbituric acid reactive substances (TBARS) levels, glutathione S-transferase (GST) and catalase (CAT) activities) were measured. AChE activity decreased when salinity increased from 25 to 30 and 35 psu at 20 and 25 degreesC. Temperature did not seem to have an influence on AChE activity. TBARS levels increased as a function of salinity when the temperature was maintained at 20 degreesC, whereas at 25 degreesC no effect of salinity could be observed. The aim of the present study was to evaluate marine contamination and toxicity in sediment and pore water from the areas of Izmir Bay (Turkey) and Mytilene Harbor (Greece). The evaluation was performed in terms of selected toxic-metals, polycyclic aromatic compounds (PAR) and toxicity to sea urchin (Paracentrotus lividus) early development and fertilization. In-situ microcosms, a tool for assessment of pesticide impacts on oyster spat (Crassostrea gigas) ArchiMer

Effects of the herbicide Basamais (bentazon) and the fungicide Opus (epoxiconazole) on oyster spat (Crassostrea gigas) were assessed using in-situ microcosms in a field experiment lasting 13 days. Six-week-old hatchery spat (mean size 1.1 mm), previously collected on PVC plates, was immersed in glass bottles filled with 200 mu m filtered seawater. Bottles were maintained underwater at 6 m depth and their water content changed every other day. Growth, measured as shell area index increase, was 126 +/- 4% in the control bottles.
