One Perkinsus species may hide another: characterization of Perkinsus species present in clam production areas of France ArchiMer
Arzul, Isabelle; Chollet, Bruno; Michel, Justine; Robert, Maeva; Garcia, Celine; Joly, Jean-pierre; Francois, Cyrille; Miossec, Laurence.
Although clam populations in France are known to be infected with protozoans of the genus Perkinsus, no molecular characterization was previously performed on these parasites. Considering that several members of this genus have been associated with mortalities of molluscs worldwide, a study was undertaken in order to characterize these parasites in France. For that purpose, clams, Ruditapes philippinarum and R. decussatus, collected from different production areas and found to be infected with Perkinsus sp. in thioglycolate culture medium, were selected for PCR-RFLP tests and sequencing. Perkinsus olseni was detected in all the investigated areas and results also suggested the presence of P. chesapeaki in Leucate, a lagoon on the Mediterranean coast and in...

Can survival of European flat oysters following experimental infection with Bonamia ostreae be predicted using QTLs? ArchiMer
Harrang, Estelle; Heurtebise, Serge; Faury, Nicole; Robert, Maeva; Arzul, Isabelle; Lapegue, Sylvie.
The present study identifies quantitative trait loci (QTLs) in response to an experimental infection with the parasite responsible for bonamiosis, Bonamia ostreae, in two segregating families of the European flat oyster, Ostrea edulis. We first constructed a genetic-linkage map for each studied family and improved the existing genetic-linkage map for the European flat oyster with a set of SNP markers. This latter map now combines the best accuracy and the best estimate of the genome coverage available for an oyster species. Secondly, by comparing the QTLs detected in this study with those previously published for O. edulis in similar experimental conditions, we identified several potential QTLs that were identical between the different families, and also...

Created in 1986, the REPAMO network (Network of mollusc pathology), is a national zoosanitary surveillance network of shellfish health status along the French coasts. Its activities are in keeping with two European Directives, 91 /67/EEC and 95/70/EC and are a patt of the institutional tasks ofIfremer. The aims of the network are to prevent the introduction and spread of exotic pathogens and to survey the evolution of notifiable pathogens already present in France. The network focused on the survey of listed diseases (bonamiosis and marteiliosis of flat oyster) in two areas under agreement process (Banc de Granville and zone X). In 2004, no sample couId be analysed to these two areas because of the difficulty to obtain flat oysters (sparse oyster beds,...

A monitoring of the natural and high density populations of commercial molluscs is performed on an annual basis within the scope of the Repamo mollusc pathology monitoring network The aim of this surveillance is (1) to monitor the health status of the molluscs along the French coast and to produce a reference picture, and (2) to study the evolution of the noticeable pathogenic agents existing on the national territory. Different sampling strategies have been introduced for the past 15 years; each strategy is based on an active monitoring including the systematic study of natural mollusc deposits. Indeed, the natural and high density mollusc populations are interrelated and pathogenic agents can be and have been exchanged between these populations with...

Bonamia ostreae-induced mortalities in one-year old European flat oysters Ostrea edulis: experimental infection by cohabitation challenge ArchiMer
Lallias, Delphine; Arzul, Isabelle; Heurtebise, Serge; Ferrand, Sylvie; Chollet, Bruno; Robert, Maeva; Beaumon, Andy; Boudry, Pierre; Morga, Benjamin; Lapegue, Sylvie.
Bonamiosis is a parasitic disease (causative agent: Bonamia ostreae) affecting the European flat oyster Ostrea edulis, responsible for a drastic decline in its aquaculture production. Selective breeding programs for resistance to bonamiosis have been undertaken to counter this disease. In the present study, a 6-month cohabitation challenge experiment was performed in order to transmit the disease from wild oysters injected with the parasite to two tested families of oysters (20 and 8-month old at the beginning of the experiment, with different genetic backgrounds) originating from a selective breeding program developed by IFREMER in France. Mortalities were checked daily and ventricular heart smears were performed on dying or moribund oysters to detect the...

Rapport régional du REPAMO 1999: résultats de la cellule de veille zoosanitaire de La Tremblade dans les régions du sud de la Loire ArchiMer
Thebault, Anne; Chollet, Bruno; Robert, Maeva.

Molecular characterization of parasites of the genus Perkinsus present in clams from French producing areas ArchiMer
Arzul, Isabelle; Michel, Justine; Chollet, Bruno; Robert, Maeva; Miossec, Laurence; Garcia, Celine; Francois, Cyrille.
Production of clams is the third most important bivalve production in France and was estimated at 3400 metric tons in 2001 among which 60% came from aquaculture and 40% from fisheries. Surveillance of clam diseases mainly relies on histology and thioglycolate culture, two techniques allowing detection of parasites of the genus Perkinsus but not specific enough to determine the parasite species. Based on a previous study aiming at estimating distribution and prevalence of perkinsosis in France, we selected 34 infected clams from 4 different producing areas in order to characterize the parasite species. Molecular characterization was done by PCR-RFLP targeting the ITS region as described by Abollo et al. (2006). 16 PCR products were cloned and 10 to 20...

Effects of temperature and salinity on the survival of Bonamia ostreae parasite infecting falt oyster Ostrea edulis in sea water ArchiMer
Arzul, Isabelle; Bond, Celine; Gagnaire, Beatrice; Chollet, Bruno; Morga, Benjamin; Ferrand, Sylvie; Robert, Maeva; Renault, Tristan.
Bonamiosis due to the intrahaemocytic protistan parasite Bonamia ostreae is a European endemic disease affecting flat oysters Ostrea edulis. The parasite has been described in different ecosystems from estuaries to open sea and no clear correlations could be demonstrated between the development of the disease and environmental parameters such as temperature or salinity. The parasite life cycle, including its survival outside the host is not completely known. Nevertheless, the infection can be directly transmitted by cohabitation between infected and non infected oysters suggesting that the parasite does not need intermediate host to complete its cycle. In the present study, the impact of temperature and salinity on the survival of purified parasites...

REPAMO: a surveillance tool for mollusc health ArchiMer
Francois, Cyrille; Garcia, Celine; Arzul, Isabelle; Joly, Jean-pierre; Miossec, Laurence; Chollet, Bruno; Ferrand, Sylvie; Robert, Maeva.
At the present time losses due to diseases are one of main factors limiting the aquaculture development. Unceasing increase of exchanges between different countries favours the risk of introduction and propagation of new pathogen agents. At the beginning of the 90’s the European Union laid the surveillance of mollusc health inside each Member State in order to protect the safety of exchanges. Indeed few measures are available to protect molluscs from diseases. Only the prophylactic approach is possible; this approach is based on a zoosanitary surveillance of mollusc populations and/or a development of resistant populations against some diseases. Created in 1986, the French network for surveillance and monitoring of mollusc health (REPAMO) ensures the...

Characterization of the protozoan parasite Marteilia refringens infecting the dwarf oyster Ostrea stentina in Tunisia ArchiMer
Elgharsalli, Refka; Aloui-bejaoui, Nejla; Sala, Hedi; Chollet, Bruno; Joly, Jean-pierre; Robert, Maeva; Couraleau, Yann; Arzul, Isabelle.
Marteilia refringens is a protozoan parasite recognized as a significant pathogen of the European flat oyster Ostrea edulis. The life cycle of this species is still poorly known, although there is evidence of the need for intermediate host(s). In the present study, we have used molecular approaches to identify this parasite in samples of the dwarf oyster Ostrea stentina after reports of massive mortality along the Tunisian coasts. In 2009 we evaluated the status of O. stentina from Monastir and checked if there was an infection with M. refringens, using polymerase chain reaction assays. Of the 103 tested O. stentina, 85 were PCRpositive using a Marteilia genus-specific assay. Additional assays were subsequently carried out on some samples collected in 2010...

In April 2010, the sampling of juvenile Pacific cupped oysters originating from France was undertaken from a farm located off the coast of the Marche region in Italy. Samples were sent to the national reference laboratory for mollusc diseases for bacteriological, histological, electron microscopical and molecular analysis. Classical and Real-Time PCR indicated the presence of the microvariant (OsHV-1 mu var) of Type 1 Ostreid Herpes virus (OsHV-1) despite the absence of clinical and pathological signs normally associated with the presence of this variant. Further molecular and sequencing analysis revealed the presence of OsHV-1 par and OsHV-1 DNA in one oyster indicating that simultaneous detection of both viruses is possible. This is the first report of...

Contribution to the understanding of the cycle of the protozoan parasite Marteilia refringens ArchiMer
Arzul, Isabelle; Chollet, Bruno; Boyer, S.; Bonnet, D.; Gaillard, Juliette; Baldi, Yoann; Robert, Maeva; Joly, Jean-pierre; Garcia, Celine; Bouchoucha, Marc.
The paramyxean parasite Marteilia refringens infects several bivalve species including European flat oysters Ostrea edulis and Mediterranean mussels Mytilus galloprovincialis. Sequence polymorphism allowed definition of three parasite types ‘M’, ‘O’ and ‘C’ preferably detected in oysters, mussels and cockles respectively.Transmission of the infection from infected bivalves to copepods Paracartia grani could be experimentally achieved but assays fromcopepods to bivalves failed. In order to contribute to the elucidation of the M. refringens life cycle, the dynamics of the infection was investigated in O. edulis, M. galloprovincialis and zooplankton over one year in Diana lagoon, Corsica (France). Flat oysters appeared non-infected while mussels were infected...
