pH controls spermatozoa motility in the Pacific oyster ( Crassostrea gigas ) ArchiMer

Investigating the roles of chemical factors stimulating and inhibiting sperm motility is required to understand the mechanisms of spermatozoa movement. In this study, we described the composition of the seminal fluid (osmotic pressure, pH, and ions) and investigated the roles of these factors and salinity in initiating spermatozoa movement in the Pacific oyster. The acidic pH of the gonad (5.82 ± 0.22) maintained sperm in the quiescent stage and initiation of flagellar movement was triggered by a sudden increase of spermatozoa external pH (pHe) when released in seawater (SW). The pollack Pollachius pollachius is a commercially important fish, considered as a candidate for aquaculture in France. Rearing attempts are promising (Suquet et al., 1996), and more information is required about its reproduction, to contribute to a better management of breeders for this species. This study was designed to examine the first maturity in pollack and the effects of conditions such as origin and delayed reproduction season, on gonad maturation, reproductive and growth performances of adults.

Influence of dry diets on reproductive performance and egg lipid composition during the first spawning season of captive pollack ArchiMer

Four‐year‐old pollack Pollachius pollachius L., previously fed on dry pellets since their juvenile stage, were divided into three experimental groups two months prior their first spawning season. They were fed a commercial broodstock pellet enriched with 6% added oils, either: (1) capelin oil (control), (2) capelin oil plus arachidonic acid, or (3) DHA rich tuna oil. Spawning performance was determined in each group and egg lipids were analysed. During the vitellogenic period, the estradiol levels in plasma increased with oocyte enlargement, indicating that captivity and pelleted feed did not affect reproductive capacity. Sperm features of captive Atlantic bluefin tuna (Thunnus thynnus) ArchiMer

P>The present study aimed to establish some basic characteristics of Atlantic bluefin tuna sperm from captive mature males, treated or untreated by gonadotropin releasing hormones agonist (GnRHa). Intratesticular milt was collected from treated and untreated fish (mean weight +/- SD: 122.9 +/- 29.2 kg, n = 21). There was no significant effect of GnRHa treatment on GSI (1.33 +/- 0.70%, n = 21) or on sperm concentration (3.8 +/- 1.3 x 1010 spermatozoa ml-1, n = 21) estimated by optical density at 260 nm. Involvement of Mitochondrial Activity and OXPHOS in ATP Synthesis During the Motility Phase of Spermatozoa in the Pacific Oyster, Crassostrea gigas ArchiMer

In the Pacific oyster, spermatozoa are characterized by a remarkably long movement phase (i.e., over 24 h) sustained by a capacity to maintain intracellular ATP level. To gain information on oxidative phosphorylation (OXPHOS) functionality during the motility phase of Pacific oyster spermatozoa, we studied 1) changes in spermatozoal mitochondrial activity, that is, mitochondrial membrane potential (MMP), and intracellular ATP content in relation to motion parameters and 2) the involvement of OXPHOS for spermatozoal movement using carbonyl cyanide m-chlorophenyl hydrazone (CCCP).       The biology of gametes and larvae in brooding mollusc species remains little explored. The aim of this study is to present the results obtained in 2016, on biological characteristics of gametes and larvae of European flat oyster.       Measurements were carried out on 50 oysters, collected by divers in the Rade de Brest (Finistère, France), during the spawning period of this species. Gonad pH is acidic (6.58±0.54). After the dissociation of spermatozeugmata, an acellular structure to which the spermatozoa are clustered, the percentage of motile spermatozoa ranged from 20 to 34% and the Velocity of the Average Path from 70 to 85µm/s. Short-term storage and cryopreservation of turbot (Scophthalmus maximus) sperm ArchiMer

Short-term storage over several days as well as cryopreservation of turbot (Scophthalmus maximus) sperm were studied. Two extenders, Ringer 200 and artificial seminal liquid (ASL2), are suggested for semen collection in order to avoid the deleterious effect of urine contamination, and for the purpose of short-term storage between 0 and 15 degrees C. Oxygen atmosphere is not suitable for turbot sperm storage. Turbot spermatozoa undergo cryopreservation with a high rate of success especially in a sucrose solution with 10% dimethyl sulfoxide (DMSO) and 10% egg yolk.

Effect of stripping frequency on turbot spawn quality. ArchiMer

In production operations the application of daily examination of each female of broodstock should be a determinant progress in hatchery management in terms of fry output. The eggs expelled shortly after ovulation by means of a systematic fivefold a week stripping attempt of females, presented a high viability rate (72%) compared to that of eggs collected twice a week (46%) whereas the treatments did not have any effect on female fecundity. Increase of the stripping rhythm brought the hatching rates and broodstock fertilities respectively from 11% to 35% and from 20,000 to 65,000 larvae/kg of spawning female. The enhancement of egg quality and of the resulting larva production is discussed.

Pacific oyster Crassostrea gigas is one major species for aquaculture, and the development of breeding programs and the need for preservation of wild stock genetic resources prompted the need for larvae cryopreservation. The objective of the present study was to choose the most reliable protocol from several existing publications, to test its biological and practical limitations, and to adapt it to hatchery conditions. The selected protocol was characterized by a very slow freezing rate without seeding, and by the use of ethylene glycol and sucrose as cryoprotectant. Sperm features in turbot (Scophthalmus maximus): a comparison with other freshwater and marine fish species ArchiMer

This paper presents some features of turbot sperm and compares them with data reported in the literature on other freshwater and marine fish. In captivity, turbot spermiation period lasts 6 months. When compared to other fish species, turbot males are characterized by low values of maximum gonadosomatic index (0.6-0.8%) and are poor sperm producers in terms of volume (0.2-2.2 ml), concentration (0.7-11.0 x 10(9) spermatozoa/ml) and total sperm number released per stripping (0.2-12.0 x 10(9) spermatozoa). Compared to most freshwater species, the duration of cell movement is long (1-17 min). The spermatozoon of turbot can be considered to be of a primitive type. Optimizing sperm cryopreservation in great scallop (Pecten maximus) ArchiMer

Anaesthesia and gonad sampling in the European flat oyster (Ostrea edulis) ArchiMer

Ce rapport fait état d'une étude réalisée au laboratoire de Génétique et Pathologie de l'IFREMER dans le cadre de l'analyse d'épisodes de mortalité anormale survenue en écloserie privée de crevettes pénéides. Plusieurs hypothèses ont été avancées. La présence de Vibrio harveyi a amené à constituer une collection de 40 souches à partir desquelles une identification bactériologique a été effectuée. Elle montre en effet une dominance de souches appartenant à un groupe homogène de Vibrio harveyi.
