Abstract Prostate cancer (PCa) is one of the leading causes of death among men. Genes such as PCA3, PSA, and Fra-1 are suggested to serve as potential tools for the detection of PCa, as they are deregulated during this pathology. A similar event occurs with small non-coding RNAs, called miRNAs, specifically miR-195-5p, miR-133a-3p, and miR-148b-3p, which were analyzed in a Chinese population and suggested to be possible candidates for PCa diagnosis. We evaluated the expression levels of three miRNAs and three genes in tissue samples of PCa and benign prostate disease, such as benign prostatic hyperplasia, or prostatitis, in order to determine their potential as candidates for PCa detection. Our results showed a statistically significant overexpression of...

The Arctic sea ice biomarker IP25: a review of current understanding, recommendations for future research and applications in palaeo sea ice reconstructions ArchiMer
Belt, Simon T.; Mueller, Juliane.
In recent years, a novel proxy for the past occurrence of Arctic sea ice has been proposed that is based on the variable marine sedimentary abundance of an organic geochemical lipid derived from sea ice diatoms in the spring. This lipid, termed IP25 (Ice Proxy with 25 carbon atoms), is a highly branched isoprenoid mono-unsaturated alkene that appears to be sufficiently stable in sediments to permit meaningful palaeo sea ice reconstructions to be carried out over short- to long-term timescales. Since the first proposed use of IP25 as a proxy for palaeo sea ice by Belt et al. (2007), a number of laboratories have measured this biomarker in Arctic sediments and it is anticipated that research activity in this area will increase further in the future. The...

A Feedback Mechanism to Control Apoptosis Occurs in the Digestive Gland of the Oyster Crassostrea gigas Exposed to the Paralytic Shellfish Toxins Producer Alexandrium catenella ArchiMer
Rolland, Jean-luc; Medhioub, Walid; Vergnes, Agnes; Abi-khalil, Celina; Savar, Veronique; Abadie, Eric; Masseret, Estelle; Amzil, Zouher; Laabir, Mohamed.
To better understand the effect of Paralytic Shellfish Toxins (PSTs) accumulation in the digestive gland of the Pacific oyster, Crassostrea gigas, we experimentally exposed individual oysters for 48 h to a PSTs producer, the dinoflagellate Alexandrium catenella. In comparison to the effect of the non-toxic Alexandrium tamarense, on the eight apoptotic related genes tested, Bax and BI.1 were significantly upregulated in oysters exposed 48 h to A. catenella. Among the five detoxification related genes tested, the expression of cytochrome P450 (CYP1A) was shown to be correlated with toxin concentration in the digestive gland of oysters exposed to the toxic dinoflagellate. Beside this, we observed a significant increase in ROS production, a decrease in...

Utilisation des métallothionéines comme biomarqueur de la contamination métallique : variabilité entre sites et organes chez l'huı̂tre Crassostrea gigas ArchiMer
Geret, F; Cosson, Rp.
Heavy metals (Cd, Cu, Hg and Zn) and metallothionein levels were determined in oysters Crassostrea gigas from two sites: the Gironde Estuary (contaminated site) and Bourgneuf Bay (uncontaminated site). Metallothionein levels led us to suspect the presence of cadmium in the environment when quantified in the gills. Soluble cadmium and metallothioneins were positively correlated in the digestive gland of oysters from Gironde (contaminated site), but metallothionein levels in this organ were not significantly different from levels observed in the digestive gland of oysters from Bourgneuf Bay (uncontaminated site). Conversely, no correlation was noticed between metals and metallothioneins in the gills of oysters from Gironde despite metallothionein levels...

Traitement par lagunage a haut rendement algal (LHRA) des effluents piscicoles marins recycles: évaluation chimique et écotoxicologique ArchiMer
Deviller, Genevieve.
World aquaculture is a fast growing activity and the control of its waste becomes a key issue. Compared to traditional systems, recirculating systems allow to reduce replacement water and waste volumes. Nevertheless their effluents are more concentrated in dissolved nutrients (nitrate, phosphate). The high rate algae pond (HRAP) is one of the possible treatment technique to remove those nutrients. During this work, three rearing systems: one recycled, one recycled with HRAP and one open as a standard, were operated for one year to assess the impact of: (1) the reuse of treated effluent on survival, growth and nutrition of sea bass (2) the rearing system on fish health using multibiomarkers and trace metals accumulation responses and (3) best conditions for...

Evaluation of the impacts of land-based contaminants on the benthic faunas of Jakarta Bay, Indonesia ArchiMer
Rees, Jg; Setiapermana, D; Sharp, Va; Weeks, Jm; Williams, Tm.
To evaluate the impact of land-based contaminants on benthic faunas, as part of the Land-Ocean Contamination Study (LOCS), a study transect was established in Jakarta Bay, Indonesia during the 1996 southeasterly monsoon. The transect extended 72 km between the city's main port, Tanjung Priok, and the Pilau Seribu Island chain in the Java Sea. The dissolved concentrations of Pb, Cu, Zn, Cr and Ni in seawater, in suspended particulate matter (SPM) and surficial sea bed sediments were measured along the transect at nine sites. In addition, metal concentrations were measured in tissues of the corals Goniopora lobata and Lobophyllia hemprichii at five sites, and of the green mussel, Perna viridis at six sites. An assessment of the impact of contaminants on the...

Etude du comportement à court terme de produits chimiques flottants après leur déversement en mer et recherche d'un indicateur d'exposition biologique ArchiMer
Mamaca, Emina.
Large volumes of chemicals are produced each year. Shipment of these chemicals by boats and barge transiting along coastal marine route are numerous. Recent examples of chemical spills as Eiwa Maru (2002) sinking with 5000 T of xylène from 20 nm of Tokyo and less far from our coastline the Ievoli Sun (2000) which sank and release 1000 T of styren has raised concern about the general lack of information of the fate of chemicals at sea and the impact on fauna and flora. The goal of this study is to characterize the behaviour of some selected chemicals at sea in order to evaluate the cinetic of natural dispersion, emulsification and evaporation. This study is done in three scale : in laboratory with a new methodologie created for this purpose, in a pilot...

Rapid metabolic adaptation in European sea bass (Dicentrarchus labrax) juveniles fed different carbohydrate sources after heat shock stress ArchiMer
Enes, P; Panserat, S; Kaushik, Sadasivam; Oliva Teles, A.
A study was conducted to evaluate the effect of two dietary carbohydrate sources (waxy maize starch and glucose) on the metabolic adaptation of sea bass juveniles (initial weight: 24 g) to a heat shock treatment (temperature rise from 18 degrees C to 25 degrees C within 24 h). Two isonitrogenous and isolipidic diets were fon-nulated to contain 20% waxy maize starch (WS diet) or 20% glucose (GLU diet). Triplicate groups of fish were fed to near satiation for 4 weeks at both temperatures (18 degrees C and 25 degrees C). Then, fish previously maintained at 18 degrees C were submitted to a heat shock (18 degrees C to 25 degrees C) and continued to be fed with the same diets during I more week. The higher water temperature significantly improved growth...

A 27 kyr terrestrial biomarker record in the southern Indian Ocean ArchiMer
Sicre, Marie-alexandrine; Labeyrie, Laurent; Ezat, Ullah; Mazaud, Alain; Turon, Jean-louis.
Terrestrial inputs were reconstructed using high molecular weight n-alkane concentrations recorded in the sub-Antarctic Indian Ocean core MD94-103 (45 degrees 35'S, 86 degrees 31'E, 3560 m) to examine regional changes in the atmospheric circulation over the last 27 kyr. This record was compared to the dust content of EPICA-Dome C ice and continental data from South Africa ( e. g., pollen sequences and isotope records in speleothems) to get a comprehensive understanding of atmospheric links between low and midlatitudes of the Indian Ocean. Terrestrial n-alkanes indicate higher glacial than Holocene inputs and marked glacial oscillations. Minimum values during the Last Glacial Maximum (LGM) are consistent with colder and drier climate and presumably caused...

Evaluation of biomarkers in oyster larvae in natural and polluted conditions ArchiMer
Damiens, Gautier; His, Edouard; Gnassia Barelli, Mauricette; Quiniou, Francoise; Romeo, Michèle.
Crassostrea gigas D-shaped larvae were subjected to different conditions of temperature and salinity for 24 h and four biomarkers (acetylcholinesterase (AChE) activity, thiobarbituric acid reactive substances (TBARS) levels, glutathione S-transferase (GST) and catalase (CAT) activities) were measured. AChE activity decreased when salinity increased from 25 to 30 and 35 psu at 20 and 25 degreesC. Temperature did not seem to have an influence on AChE activity. TBARS levels increased as a function of salinity when the temperature was maintained at 20 degreesC, whereas at 25 degreesC no effect of salinity could be observed. Variations in GST and CAT activities were not significant with salinity and temperature except that catalase activity was higher at 25...

Genotoxicity and activation of cellular defenses in transplanted zebra mussels Dreissena polymorpha along the Seine river ArchiMer
Chatel, Annelle; Faucet-marquis, Virginie; Gourlay-france, Catherine; Pfohl-leszkowicz, Annie; Hubert, Francoise.
The aim of the present study was to confirm the relevance of studying DNA adduct formation in a field study. In that context, freshwater mussels Dreissena polymorpha, collected in a reference station, were transplanted in different sites with a pollution gradient. After one and two months, mussels were collected and DNA adduct formation was analyzed using the 32P post labelling technique on both gills and digestive glands. In addition, the expression of genes involved in the detoxification system (catalase (CAT), superoxide dismutase (SOD), glutathione S-transferase (GST), HSP70, aryl hydrocarbon receptor (AHR), P glycoprotein (PgP), metallothionein (MT)) was assessed by RT-PCR. DNA adducts were observed at amount comparable to data from literature....

Natural factors to consider when using acetylcholinesterase activity as neurotoxicity biomarker in Young-Of-Year striped bass (Morone saxatilis) ArchiMer
Durieux, Eric; Farver, Thomas B.; Fitzgerald, Patrick S.; Eder, Kai J.; Ostrach, David J..
Acetylcholinesterase (AChE) activity is one of the most common biomarkers of neurotoxicity used in aquatic organisms. However, compared to its extensive use as biomarker, the effects of natural factors onAChE activity remain unclear especially in estuarine fishes. The aim of this study was to evaluate the effects of natural factors on AChE activity of striped bass (Morone saxatilis) juveniles. Brain AChE activity was measured in YOY (Young-Of-Year) individuals collected monthly from August 2007 to January 2008 at 12 different sites in the San Francisco Estuary system. The spatio-temporal variability of AChE was analyzed relative to water temperature and salinity as well as fish size. AChE activity was highly positively correlated with water temperature and...

Many maritime countries in Europe have implemented marine environmental monitoring programmes which include the measurement of chemical contaminants and related biological effects. How best to integrate data obtained in these two types of monitoring into meaningful assessments has been the subject of recent efforts by the International Council for Exploration of the Sea (ICES) Expert Groups. Work within these groups has concentrated on defining a core set of chemical and biological endpoints that can be used across maritime areas, defining confounding factors, supporting parameters and protocols for measurement. The framework comprised markers for concentrations of, exposure to and effects from, contaminants. Most importantly, assessment criteria for...

The variations of catalase activity, which is a biochemical biomarker of exposure to an oxidative stress, were followed month by month (from May 1998 to May 1999) in Mediterranean clams (Ruditapes decussatus) collected at three stations (J, A and F), and mussels (Mytilus galloprovincialis) collected at two stations (J and C) in Lake Bizerte (Tunisia). Several oceanographic parameters were measured together with catalase activity in the animals to evaluate the quality of waters in the lake. Whatever the station taken into consideration, catalase activity in clams significantly varied as a function of the season, with marked increases in September, when the quality of waters was poor. Clams originating from station A presented the highest mean activity...

A 49-month-old Holstein cow with anorexia, tachypnea, enlarged peripheral lymph nodes, and difficulty standing up was suspected of bovine leukosis. Hematological examination revealed lymphocytosis with the presence of neoplastic cells. Increased total lactate dehydrogenase (LDH) activity, isozymes of LDH-2 and LDH-3 activities and thymidine kinase activity were observed. Cytological findings of fine needle aspiration of subiliac lymph nodes indicated lymphosarcoma. Histopathology and antibody analysis confirmed the diagnosis of enzootic bovine leukosis, a B-cell bovine lymphoma caused by bovine leukemia virus. Gene expressions known as biomarkers of hematopoietic neoplasia in human were also examined in the present case. Increased messenger RNA expression...

Heat shock proteins on beef quality PAB
Moncau,Cristina Tschorny; Rosa,Alessandra Fernandes; Eler,Joanir Pereira; Balieiro,Júlio César de Carvalho.
Abstract: The objective of this work was to quantify heat shock proteins (HSP) 27 and 70 in the Longissimus dorsi muscle of cattle during aging and to check their potential as biomarkers for meat quality. A total of 191 steers ½ South African Simmental x ½ Nellore (16-18 months, 391.7±99.7 kg), castrated, and feedlot finished were used. Meat quality was measured by pH, color, cooking loss, and shear force (SF) at 1 and 14 days of aging time. HSP27 and HSP70 were quantified according to the SF values in the more and less tender meat groups, with 20 samples each, for each aging time. HSP27 concentrations in more and less tender meat decrease from 1 to 14 days of aging, and do not differ when evaluated at the same period. HSP70 concentrations in more tender...

Use of clearance rate in Choromytilus chorus (Bivalvia: Mytilidae) as a non-destructive biomarker of aquatic pollution RChHN
TORO,BEATRIZ; NAVARRO,JORGE M.; PALMA-FLEMING,HERNÁN.
Reduction in clearance rate was observed in the mussel Choromytilus chorus in relation to the degree of environmental pollution. Three sampling sites included the polluted San Vicente Bay, middling polluted Corral Bay, and weakly polluted Yaldad Bay in southern Chile. Pollution levels were estimated by quantitative analysis of polynuclear aromatic hydrocarbons (PAHs) in mussel tissue from the three sites. Mussels from San Vicente showed lowest clearance rates and highest PAHs levels in both spring 1998 and summer 1999. Populations from Corral and Yaldad showed generally low levels of PAHs, so the mussels from Yaldad showed the highest rates of clearance in comparison to Corral and San Vicente populations. The significant negative relationship (P <...

Abstract Circulating miRNAs presenting in plasma in a stable manner have been demonstrated their potential role as a promising biomarkers in many human diseases, such as Alzheimer’s disease, melanoma and ovarian carcinoma. However, few circulating miRNAs could be used for breast ductal cancer diagnosis. Here, we identified miR-1273g-3p as a biomarker for detecting breast ductal cancer. We detected miR-1273g-3p levels in the plasma of 39 sporadic breast ductal cancer patients and 40 healthy donors by Stem-loop Quantitative Real-time PCR (qRT-PCR). The results showed the plasma miR-1273g-3p level were significantly up-regulated in breast ductal cancer patients compared with healthy donors (p=0.0139). The objective of this work was to estimate the accuracy of mesothelin as a biomarker for ovarian cancer. A quantitative systematic review was performed. A comprehensive search of the Medline, LILACS, SCOPUS, Embase, Cochrane Central Register of Controlled Trials, Biomed Central, and ISI Web of Science databases was conducted from January 1990 to June 2015. For inclusion in this systematic review, the papers must have measured mesothelin levels in at least two histological diagnoses; ovarian cancer (borderline or ovarian tumor) vs. benign or normal ovarian tissue. For each study, 2 x 2 contingency tables were constructed. We calculated the sensitivity, specificity and diagnostic odds ratio. The verification bias was performed according to QUADAS-2....
