Contenido y caracterización reológica de los polisacáridos estructurales de nopalitos (Opuntia spp.) en un gradiente de domesticación. Colegio de Postgraduados

Evidencias diversas indican que la domesticación del género Opuntia comenzó con la selección de variantes de O. streptacantha, seguida de O. hyptiacantha y O. megacantha y se centró en O. albicarpa y O. ficus indica. Las causas por las cuales se domesticó Opuntia fueron por sus frutos y nopalitos. Los polisacáridos más abundantes en los nopalitos son del tipo estructural. La hipótesis que se plantea es que los polisacáridos muestran un gradiente de concentración y sus propiedades reológicas han sido modificadas en función del grado de domesticación de Opuntia. Efficient expression and characterization of a cold-active endo-1, 4-β-glucanase from Citrobacter farmeri by co-expression of Myxococcus xanthus protein S Electron. J. Biotechnol.

Background: Cold-active endo-1, 4-β-glucanase (EglC) can decrease energy costs and prevent product denaturation in biotechnological processes. However, the nature EglC from C. farmeri A1 showed very low activity (800 U/L). In an attempt to increase its expression level, C. farmeri EglC was expressed in Escherichia coli as an N-terminal fusion to protein S (ProS) from Myxococcus xanthus. Results: A novel expression vector, pET(ProS-EglC), was successfully constructed for the expression of C. farmeri EglC in E. coli. SDS-PAGE showed that the recombinant protein (ProS-EglC) was approximately 60 kDa. The activity of ProS-EglC was 12,400 U/L, which was considerably higher than that of the nature EglC (800 U/L). Generation and analysis of an Eucalyptus globulus cDNA library constructed from seedlings subjected to low temperature conditions Electron. J. Biotechnol.

Eucalyptus globulus is the most important commercial temperate hardwood in the world because of its wood properties and due to its characteristics for biofuel production. However, only a very low number of expressed sequence tags (ESTs) are publicly available for this tree species. We constructed a cDNA from E. globulus seedlings subjected to low temperature and sequenced 9,913 randomly selected clones, generating 8,737 curated ESTs. The assembly produced 1,062 contigs and 3,879 singletons forming a Eucalyptus unigene set. Based on BLASTX analysis, 89.3% of the contigs and 88.5% of the singletons had significant similarity to known genes in the non-redundant database of GenBank. At present, reforestation has focused on native forests with anthropogenic intervention and eroded soils. There is interest in producing Nothofagus seedlings which can overcome adverse conditions encountered on reforestation sites. It is necessary to find new fungi that can be utilized as mycorrhizal inoculants and that enable the seedlings to increase their tolerance to adverse conditions. Two ectomycorrhizal strains of the fungus Descolea antarctica (D1 and D2) were cultured at different temperatures, pH levels and the activities of amylases, cellulases, and phosphatases were determined. Production of cold-adapted cellulase by Verticillium sp. isolated from Antarctic soils Electron. J. Biotechnol.

Background: Cellulose can be converted to ethanol by simultaneous saccharification and fermentation (SSF). The difference between the optimal temperature of cellulase and microbial fermentation, however, has been identified as the critical problem with SSF. In this study, one fungal strain (AnsX1) with high cellulase activity at low temperature was isolated from Antarctic soils and identified as Verticillium sp. by morphological and molecular analyses. Results: The biochemical properties of crude AnsX1 cellulase samples were studied by filter paper cellulase assay. Reunindo excelentes características, a madeira de Ucuúba (Virola surínamensis (Rol) Warb) tem sido extensivamente empregada na fabricação de laminados. Como consequêncía do processo produtivo, a madeira que não pode ser integralmente aproveitada, deixa um apreciável resíduo que, a falta de melhor emprêgo, não tem tido outra utilizaçãc senão como combustível na própria Usina. The mineralization of cellulose was examined in mono and cocultures of marine anaerobic bacteria (cellulolytic bacteria, fermentative non-cellulolytic bacteria and methane-producing bacteria). These studies displayed interactions between these groups of microorganisms: inhibition relationships (inhibition of methanogens during active growth of fermentative bacteria) and syntrophic relationships (growth of fermentative non-cellulolytic bacteria depended on metabolites produced by cellulolytic bacteria; increased in rate of cellulose hydrolysis in presence of fermentative non-cellulolytic bacteria; interspecies H sub(2) transfer reaction between fermentative bacteria and methane-producing bacteria).

Abstract: The objective of this work was to evaluate the diet components - crude fiber, neutral detergent fiber, acid detergent fiber, hemicellulose, cellulose, and lignin - as internal indicators in the determination of the apparent digestibility coefficients of dry matter, gross energy, and of the nutrients - crude protein, phosphorus, and amino acids - in Nile tilapia (Oreochromis niloticus). Groups of ten juveniles (80.3±1 g) were randomly distributed in six tanks of 250 L and fed two practical diets, either of a plant-origin diet or of a plant- and animal-origin diet. The objective of this study was to evaluate the effects of supplementation with different sources of energy and nitrogenous compounds on the in vitro growth and production of bacteriocin of lactic acid bacteria. Incubations were performed by using ruminal fluid from a rumen-fistulated Holstein-Zebu steer. The animal was kept on a Brachiaria decumbens pasture receiving 200 g/day of supplemental crude protein. The objective was to evaluate the differences between distinct types of litter material and their combinations in the dynamics of degradation on the organic matter fractions and the quality of the final compound. The treatments were established according to material used as substrate for broiler litter: treatment 1 - rice husks; 2 - sugar cane bagasse; 3 - wood shavings; 4 - wood shavings + sugar cane bagasse; 5 - rice husks + sugar cane bagasse; and 6 - Napier grass. The growth kinetics of Acetivibrio cellulolyticus grown in medium containing different carbon sources (cellobiose, amorphous or crystalline cellulose) was investigated. The specific growth rate was higher in cellobiose fed cultures than in the presence of the other two substrates. Endoglucanase production was greater in cultures grown on amorphous cellulose; enzyme activity increased during the stationary phase in cultures grown on crystalline cellulose.

Optimized acid hydrolysis of the polysaccharides from the seaweed Solieria filiformis (Kützing) P.W. Gabrielson for bioethanol production Biological Sciences

 The seaweeds are bio-resource rich in sulfated and neutral polysaccharides. The tropical seaweed species used in this study (Solieria filiformis), after dried, shows 65.8% (w/w) carbohydrate, 9.6% (w/w) protein, 1.7% (w/w) lipid, 7.0% (w/w) moisture and 15.9% (w/w) ash. The dried seaweed was easily hydrolyzed under mild conditions (0.5 M sulfuric acid, 20 min.), generating fermentable monosaccharides with a maximum hydrolysis efficiency of 63.21%. Galactose and glucose present in the hydrolyzed were simultaneously fermented by Saccharomyces cerevisiae when the yeast was acclimated to galactose and cultivated in broth containing only galactose.
