Bovine papillomavirus type 2 in reproductive tract and gametes of slaughtered bovine females BJM
Carvalho,Claudemir de; Freitas,Antonio Carlos de; Brunner,Olga; Góes,Luiz Gustavo Bentim; Cavalcante,Andréa Yaguiu; Beçak,Willy; Santos,Rita de Cassia Stocco dos.
Papillomaviruses are described selectively infecting epithelial tissues and are associated with many forms of cancer in different species. Considering the widespread dissemination of papillomatosis in livestock, interest is being centred on possible forms of viral transmission and respective mechanisms. In the present study, we report the detection of bovine papillomavirus (BPV) DNA sequences in female reproductive tract tissues, fluids and oocytes from slaughtered bovines not afflicted by cutaneous papillomatosis. BPV-2 DNA sequences were found in ovarian and uterine tissues as well as in oocytes, cumulus cells and uterine flushings. The presence of papillomavirus sequences in reproductive organ tissues and fluids shows that viral infection in organisms...

The dinoflagellate Alexandrium minutum affects development of the oyster Crassostrea gigas, through parental or direct exposure ArchiMer
Castrec, Justine; Hegaret, Helene; Alunno-bruscia, Marianne; Picard, Mailys; Soudant, Philippe; Petton, Bruno; Boulais, Myrina; Suquet, Marc; Queau, Isabelle; Ratiskol, Dominique; Foulon, Valentin; Le Goic, Nelly; Fabioux, Caroline.
Harmful algal blooms are a threat to aquatic organisms and coastal ecosystems. Among harmful species, the widespread distributed genus Alexandrium is of global importance. This genus is well-known for the synthesis of paralytic shellfish toxins which are toxic for humans through the consumption of contaminated shellfish. While the effects of Alexandrium species upon the physiology of bivalves are now well documented, consequences on reproduction remain poorly studied. In France, Alexandrium minutum blooms have been recurrent for the last decades, generally appearing during the reproduction season of most bivalves including the oyster Crassostrea gigas. These blooms could not only affect gametogenesis but also spawning, larval development or juvenile...

La Reproduction naturelle et contrôlée des bivalves cultivés en France. -Rapport de Groupe de Travail- IFREMER/Nantes/France, 14 et 15 novembre 1995 ArchiMer
Devauchelle, Nicole; Barret, Jean; Salaun, Gilles.
This report compiles 17 short and illustrated syntheses concerning the natural strategies of reproduction and the artificial control of the reproductive process for the flat oyster, Ostrea edulis, the japanese oyster, Crassostrea gigas, the mussels, Mytilus edulis and My ti/us galloprovincialis, the scallop, Pecten maximus. An additional syntheses is presented, as a comparison within molluscs, which are the main research topics at IFREMER concerning the artificial control of the reproductive processes in Marine Fish. Three main subjects are documented : 1. the ecophysiology of the natural period of reproduction in the wild, 2. the methods developed in the hatcheries for the artificial control of the reproductive periods, the gametes collection and the...

Cellular responses of Pacific oyster ( Crassostrea gigas ) gametes exposed in vitro to polystyrene nanoparticles ArchiMer
Gonzalez-fernandez, Carmen; Tallec, Kevin; Le Goic, Nelly; Lambert, Christophe; Soudant, Philippe; Huvet, Arnaud; Suquet, Marc; Berchel, Mathieu; Paul-pont, Ika.
While the detection and quantification of nano-sized plastic in the environment remains a challenge, the growing number of polymer applications mean that we can expect an increase in the release of nanoplastics into the environment by indirect outputs. Today, very little is known about the impact of nano-sized plastics on marine organisms. Thus, the objective of this study was to investigate the toxicity of polystyrene nanoplastics (NPs) on oyster (Crassostrea gigas) gametes. Spermatozoa and oocytes were exposed to four NPs concentrations ranging from 0.1 to 100 mg L−1 for 1, 3 and 5 h. NPs coated with carboxylic (PS-COOH) and amine groups (PS-NH2) were used to determine how surface properties influence the effects of nanoplastics. Results demonstrated the...

Nanoplastics impaired oyster free living stages, gametes and embryos ArchiMer
Tallec, Kevin; Huvet, Arnaud; Di Poi, Carole; Gonzalez-fernandez, Carmen; Lambert, Christophe; Petton, Bruno; Le Goic, Nelly; Berchel, Mathieu; Soudant, Philippe; Paul-pont, Ika.
In the marine environment, most bivalve species base their reproduction on external fertilization. Hence, gametes and young stages face many threats, including exposure to plastic wastes which represent more than 80% of the debris in the oceans. Recently, evidence has been produced on the presence of nanoplastics in oceans, thus motivating new studies of their impacts on marine life. Because no information is available about their environmental concentrations, we performed dose-response exposure experiments with polystyrene particles to assess the extent of micro/nanoplastic toxicity. Effects of polystyrene with different sizes and functionalization (plain 2-μm, 500-nm and 50-nm; COOH-50 nm and NH2-50 nm) were assessed on three key reproductive steps...

Genotoxicity of diuron and glyphosate in oyster spermatozoa and embryos ArchiMer
Akcha, Farida; Spagnol, Charlene; Rouxel, Julien.
We investigated the effects of genotoxicant exposure in gametes and embryos to find a possible link between genotoxicity and reproduction/developmental impairment, and explore the impact of chemical genotoxicity on population dynamics. Our study focused on the genotoxic effects of two herbicides on oyster gametes and embryos: glyphosate (both as an active substance and in the Roundup formulation) and diuron. France is Europe's leading consumer of agrochemical substances and as such, contamination of France's coastal waters by pesticides is a major concern. Glyphosate and diuron are among the most frequently detected herbicides in oyster production areas; as oyster is a specie with external reproduction, its gametes and embryos are in direct contact with...

Artificial fertilization of oocytes and sperm activation in pacu: effects of the spermatozoa:oocyte ratio, water volume, and in natura semen preservation R. Bras. Zootec.
Sanches,Eduardo Antônio; Baggio,Diego Mendes; Piana,Pitágoras Augusto; Souza,Bruno Estevão de; Bombardelli,Robie Allan.
The objective of this work was to investigate artificial fertilization and the duration of sperm motility in pacu with different insemination doses, water volume, and in natura semen preservation. It was carried out four experimentsfor evaluation of insemination doses (7x10³, 7x10(4), 7x10(5), 7x10(6), and 7x10(7) spermatozoa oocytes-1) on the artificial fertilization of oocytes; the effect of water volume (0.5, 15.0, 30.0, 45.0, and 60.0 mL water mL-1 of oocyte) with insemination doses of 105,481 and 210,963 spermatozoa oocytes-1; the effect of semen dilutions (0.005, 0.05, 0.5, and 5.0 µL semen mL-1 of water) on sperm motility duration; and the effect of storage at 15ºC for 9h on sperm motility duration and sperm survival ratio. The highest results...
