Effect of PHA and LPS on the Activity of Nonspecific Cytotoxic Cells and Macrophages of Rainbow Trout (Oncorhynchus mykiss) Gayana

This work studied the effect of the lectin phytohaemagglutinin (PHA) and bacterial lipopolysaccharide (LPS) on the activity of nonspecific cytotoxic cells (NCCs) and macrophages of peripheral blood in the rainbow trout. NCCs are cells of the innate immunity, homologues to Natural Killer (NK) cells of mammals, capable of eliminating cells infected with intracellular pathogens and also tumoral cells. Their lytic activity is mediated by the perforin/granzyme system and the Fas receptor-Fas ligand (FasR-FasL) system. Activity of NCCs was evaluated by their capacity to lyse HL-60 tumor cells. The defence against infections is the essential function of the immune system of all Vertebrates. Fish present an efficient and developed immunologic response and in the case of Teleosts, with some similarities to the higher vertebrates. It can be divided into two types: innate or non specific and adquired or specific, both with a humoral and cellular compartments. Their relative importance can change according to the age of fish and is always under the influence of different factors. The most relevant components are mentioned here. Arnoglossus laterna is only infested by intestinal parasites. Each parasite distribution is independent of the distribution of other parasites and also independent of the sex of host. When a fish is infested by a great number of the two helminths Acanthocephaloides and Bothriocephalus clavibothrium , the proportion of lymphocytes tends to decrease and the proportion of neutrophils tends to increase (inflammatory response). Important infestation by Scolex pleuronectis induces an increase of the thrombocyte count for 100 counted leucocytes. After infestation by the nematode Thynnascaris sp. alone or with another parasite, even by small number, the proportion of lymphocytes decreases and the proportion of neutrophils increases.

Leukocyte-endothelial cell interactions have a pivotal role in immune responses. In our study Ten adult wistar albino rats weighing 180-200 g were used to obtain tissue samples. The animals were sacrificed by decapitation under ether anesthesia. The lymph nodes of the animals were then quickly removed. Specimens were immersed in the zinc iodide-osmium tetroxide (ZIO) solutation by Niebauer et al. (1969) and kept in the dark for 2 hours in this solution from temperature for fixation/staining. The Phrynops hilarii specie of turtle has its characterization not well defined in the literature, it was proposed in this study the leukocyte characterization of the blood, stained by Leishman and analyzed under light and transmission electron microscope. It was not observe any cellular type with similar characteristics to neutrophils in mammalian group. We believed, based on the data obtained in this study that the heterophils have a morphofuncional analogy with another neutrophils belonged to mammalian group. Purification of a lectin from Bothrops jararacussu venom (BjcuL) was carried out using agarose-D-galactose affinity gel. MALDI-TOF gave a major signal at m/z 32028, suggesting the presence of a dimmer composed of two identical subunits. Divalent cations were required for the lectin activity, as complete absence of such ions reduced hemagglutination. BjcuL was more effective at neutral pH and showed total loss of activity at pH values below 4.0 and above 9.0. Its agglutinating activity remained stable at 25°C until 60min, but increased when at 35°C for at least 15min. Among these species, tucunaré of the genus Cichla stand out, them being of high economical interest for sport fishing and Amazon's industry of ornamental fish. The present study describes the weight-length relationship (WLR), relative condition factor (Kn), red blood cell parameters, thrombocytes and leukocytes count of Cichla temensis Humboldt, 1821, farmed in central Amazon. The objective of this work was to characterize the hematological profiles of steers and bulls, according to residual feed intake (RFI), after feed deprivation. Twenty‑month‑old Nellore steers and bulls were fed feedlot diets for 70 and 56 days, respectively. RFI was calculated as the difference between actual feed intake and expected feed intake. More and less efficient steers and bulls, according to RFI, were subjected to 24 hours of food deprivation. Blood was sampled prior to and following the withdrawal period. The leukocytes parameters in approximately one-year-old freshwater fish Colossoma macropomum Cuvier, 1818 (tambaqui) kept in an intensive monobreeding system as well as the correlation among these parameters and the biometric data (total weight and standard length) were investigated. The mean value of the white blood cell count (WBC) in the peripheral blood of tambaqui was 2663.3±1288µl and in the differential count, the following means were observed: neutrophils (1566.2±754µl); lymphocytes (973.6±447µl); monocytes (86.7±123µl) and special granulocitic cells (7.8±144µl). Methodological limitations of counting total leukocytes and thrombocytes in reptiles (Amazon turtle, Podocnemis expansa): an analysis and discussion Acta Amazonica

The aim of this paper is to compare three different methods for counting white blood cells [WBC] (Natt and Herrick method, estimation with 1,000 and 2,000 erythrocytes) and three methods for counting total thrombocytes [TT] (Wojtaszek method, estimation with 1,000 and 2,000 erythrocytes) in a South American freshwater turtle species, Podocnemis expansa, Schweigger 1812 (Reptilia, Pelomedusidae). Direct WBC counts using the Natt and Herrick method showed limitations, which are discussed here. ABSTRACT The hematological and biochemical responses of pirarucu fingerlings (Arapaima gigas) fed with diets containing different concentrations of a glucomannan product derived from yeast and algae were evaluated in order to ascertain the effect of these diets on fish physiology. Four treatments were conducted, with three replications, with 12 fish in each tank. The product evaluated (MycosorbA+(r)) was incorporated into the commercial diet, at four concentrations: 0, 1, 2 and 4 g.kg-1, called M0%, M0.1%, M0.2% and M0.4%, respectively. Haematological assessment of four amazonian ornamental armoured catfish (Teleostei, Loricariidae) Biological Sciences

The aim of this study was to characterize the haematological profile of four Amazonian ornamental freshwater armoured catfish: acari-bola (Peckoltia oligospila - L06), acari-pleco (Cochliodon sp. - L145), acari-canoa (Lasiancistrus saetiger - L323) and acari-assacu (Pseudocanthicus spinosus - L160). The highest blood glucose levels (72.47 ± 28.7 mg dL-1) and erythrocyte counts (0.51 ± 0.2 x 106 cel. μL-1) were recorded for acari-canoa. Natural insecticides derived from plant extracts have been used as an alternative to synthetic products in order to reduce environmental contamination. The present study aimed to examine the effects of Fumydro®, a natural insecticide based in the tobacco plant Nicotiana tabacum, on the Nile tilapia (Oreochromis niloticus) by determining the 48-h LC50 and evaluating their effects on hematological variables. Adult specimens of O. niloticus were exposed to four Fumydro® concentrations (200, 300, 400 and 500 μL L-1). The 48-h LC50 of Fumydro® was determined as 370 ± 50 μL L-1. ABSTRACT This study describes the hematological and biometric characteristics of male and female Gymnotus species from the Pantanal, Mato Grosso do Sul state, Brazil. Fifty adult specimens of Gymnotus inaequilabiatus were weighed, measured, and then euthanized. The action of the "Stress Factor Ostrich (Arenales - Fauna and Flora)" was tested in the release of superoxide anion by cells in the peripheral blood of rhea (Rhea americana). Sixteen samples of 0.5mL of venous blood were collected through the jugular vein in the morning and placed in heparinized tubes. The leukocytes were separated into polymorphonuclear (PMN) and mononuclear (MN). The production of superoxide anion by phagocytes of peripheral blood was determined using the chromogen ferricytochrome C. There was a reduction of superoxide by MN cells in the presence of "Stress Factor Ostrich" indicating a positive influence of product against oxidative stress. Hepatocytes, rather than leukocytes reverse DNA damage in vivo induced by whole body y-irradiation of mice, as shown by the alkaline comet assay Biol. Res.

DNA damage repair was assessed in quiescent (G0) leukocytes and in hepatocytes of mice, after 1 and 2 hours recovery from a single whole body y-irradiation with 0.5, 1 or 2 Gy. Evaluation of single-strand breaks (SSB) and alkali-labile sites together were carried out by a single-cell electrophoresis at pH>13.0 (alkaline comet assay). In non-irradiated (control) mice, the constitutive, endogenous DNA damage (basal) was around 1.5 times higher in leukocytes than in hepatocytes. Irradiation immediately increased SSB frequency in both cell types, in a dose-dependent manner. Two sequential phases took place during the in vivo repair of the radio-induced DNA lesions.
