Seeds are the most important plant storage organ and play a central role in the life cycle of plants. Since little is known about the protein composition of rice (Oryza sativa) seeds, in this work we used proteomic methods to obtain a reference map of rice seed proteins and identify important molecules. Overall, 480 reproducible protein spots were detected by two-dimensional electrophoresis on pH 4-7 gels and 302 proteins were identified by MALDI-TOF MS and database searches. Together, these proteins represented 252 gene products and were classified into 12 functional categories, most of which were involved in metabolic pathways. Resumo - Este trabalho teve como objetivo realizar testes de campo e de laboratório, utilizando os princípios das boas práticas de laboratório (BPL) e boas práticas agrícolas (BPA). Esses princípios foram criados para harmonizar procedimentos e garantir a aceitação dos resultados em diferentes países e regiões da Organização para Cooperação e Desenvolvimento Econômico (OCDE), além de serem obrigatórios para registro de agrotóxicos. It is essential to have simple rapid methods for the determination of fatty acid structures. Traditionally, fatty acids are analysed by gas chromatography using their methyl ester derivatives (FAME). However, their corresponding mass spectra exhibit molecular ions but are usually devoid of ions indicative of structural features and, notably, the position of double bounds on the aliphatic chains [1]. Recent work has shown that variations in zinc (Zn) isotope ratios enable us to identify contamination sources in the terrestrial environment and uptake processes in higher plants. Here in this study, we demonstrate that this also holds true for mangrove forests, which play an important role in the biogeochemical cycling of metals in tropical coastal ecosystems and that are seriously threatened by anthropogenic pollution. Although Glycosyl-Inositol-Phospho-Ceramides (GIPCs) are the main sphingolipids of plant tissues, they remain poorly characterized in term of structures. This lack of information, notably with regard to polar heads, currently hampers the understanding of GIPC functions in biological systems. The lipids of a deep-sea hydrothermal vent arch ae on, Thermococcus S 557, were isolated, purified and structurally determined. Based on acid methanolysis and spectroscopic studies, the polar lipids were shown to comprise diphytanyl glycerol and dibiphytanyl diglycerol, typical membrane lipids of Arch aea. From the neutral lipids, 2,3-di-O-dihydro-14,15-geranylgeranyl glycerol was isolated. The green microalgae Dunaliella genus is known for the production of high added value molecules. In this study, strain AL-1 was isolated from the Sebkha of Sidi El Hani (Sousse, Tunisia). This isolate was identified both morphologically and genetically via 18S rRNA gene sequence as a member of the genus Dunaliella. Strain AL-1 was found to be closely related to Dunaliella salina, Dunaliella quartolecta and Dunaliella polymorpha with more than 97% similarity.
