An evaluation of the genotoxicity of domestic sewage was carried out in a municipal wastewater treatment plant. The study was carried out using Allium cepa root tip cells through cytological parameters such as aberrant cells in anaphase-telophase and the mitotic index, and the fish species Oreochromis niloticus and Tilapia rendalli, through the index of micronuclei in peripheral erythrocytes. In the Allium test, each of the four stages of the wasterwater treatment routine was analyzed, i.e., crude sewage, primary effluent, secondary effluent, and tertiary effluent. O. niloticus and T. rendalli specimens were maintained in an aquarium continuously receiving the final effluent, in an open system, before being discharged into Lake Paranoá. For micronuclei... |

Abstract Titanium dioxide nanoparticles (TiO2NPs) are widely used and may impact the environment. Thus, this study used a high concentration of TiO2NP (1000 mg/L) to verify the defense mechanisms triggered by a plant system – an indicator of toxicity. Furthermore, this study aimed at completely characterizing TiO2NP suspensions to elucidate their toxic behavior. TiO2NPs were taken up by meristematic cells of Allium cepa, leading to slight inhibition of seed germination and root growth. However, severe cellular and DNA damages were observed in a concentration-dependent manner (10, 100, and 1000 mg/L). For this reason, we used the highest tested concentration (1000 mg/L) to verify if the plant cells developed defense mechanisms against the TiO2NPs and... |

The aquatic environment receives many contaminants that can induce damages at the molecular, biochemical, cellular and physiological levels. Centropomus parallelus , an important food resource for local populations, is a predator fish that feeds on small fishes and benthic invertebrates, thus being vulnerable to the bioconcentration and biomagnification processes. This study aimed to evaluate cytogenotoxic responses in erythrocytes from C. parallelus juveniles collected in the Cananéia and São Vicente estuaries, both in winter and in summer. After anesthesia, blood samples were collected by caudal puncture. Blood smears were prepared on glass slides and stained with May-Grünwald-Giemsa dye. Two thousand cells were analyzed per slide (1000x), and nuclear... |

Agaricus blazei Murrill, popularly known as the sun mushroom, is a native mushroom in SP, Brazil, that has been widely used in the treatment of cancer and many other pathologies in different parts of the world. A water-soluble protein-polysaccharide complex (1 -> 6)beta-D-glucan has been isolated from its fruiting body that showed immune-modulation activity. From organic extracts, linoleic acid has been isolated and determined to be the main substance with antimutagenic activity. Using both the micronucleus (MN) and comet (single cell microgel electrophoresis) assays, this study determined the genotoxic and antigenotoxic potential of A. blazei (AB) obtained from commercial sources or the following strains: a) strains AB 97/29 (young and sporulated... |

Previous research has shown connections between the exposure to different agents and mutations in germinal and somatic cells. One of such agents, nitrate (NO3), can be potentially genotoxic for individuals who drink high nitrate well water, and may cause a subsequent impact on their health and on the whole ecosystem. The aim of this research was to conduct genotoxicity assays in somatic cells from people living in the northern area of Mar del Plata city, Argentina. The purpose of these experiments was, in turn, to establish the possible relationships between mutagenic effects and high nitrate water consumption. To this end, different diagnostic tests were carried out to detect potential genetic and/or chromosomal alterations. A non-exposed population equal... |

The effects of various cadmium (Cd) concentrations (10, 50 and 100 μM) on root growth, cell division and micronuclei formation in root tip cells of Allium cepa var. agrogarum L. were investigated to better understand the processes of Cd-induced apoptosis. The results indicated that 10 μM of Cd had an obviously passive influence on root growth during the 24 h treatment, and that the influence was even more serious with increasing Cd concentrations and duration of treatments. The mitotic index decreased with increasing Cd concentration and duration of treatments. Cadmium induced c-mitosis, chromosome bridges, chromosome stickiness and micronuclei. The frequency of cells with these disorders varied with prolonging the duration of treatments... |

ABSTRACT The skin secretion from toads of the Bufonidae family has great potential in the search for new active compounds to be used as drug candidates in treating some diseases, among them cancer. In this context, this study aimed to evaluate the cytotoxic and antimutagenic activity of the parotoid gland secretion extracts of Rhinella marina and Rhaebo guttatus, as well as biochemically analyze transaminases and serum creatinine for liver and renal damage, respectively. Cytotoxicity was performed by the colorimetric method based on MTT (3- [4, 5-dimethyl-2-thiazolyl]-2, 5-diphenyl-2H-tetrazolium bromide) with different concentrations of the extracts in Walker or splenic tumor cell cultures from rats and mice. The micronucleus test was performed with male... |

This study aimed to determine the variables leukocytary, erythrocytary, frequency of micronuclei and nuclear abnormalities in peripheral erythrocytes of Prochilodus argenteus, Pimelodus maculates and Myleus micans from São Francisco river basin, Minas Gerais State, during the summer and winter. Thrombocytes, hematocrit and leukocyte of P.argenteus series, was influenced by climatic conditions. In P. maculatus, not were significant differences for all leukocytes and thrombocytes. In M. micans, values are unprecedented in the literature, with MCV and absolute number of leukocytes found superior to other species of the family Characidae. The frequency of micronuclei and nuclear abnormalities in erythrocytes, the three species not showed seasonal differences... |

Solanum paniculatum L. is a plant species widespread throughout tropical America, especially in the Brazilian Cerrado region. It is used in Brazil for culinary purposes and in folk medicine to treat liver and gastric dysfunctions, as well as hangovers. Previous studies with S. paniculatum ethanolic leaf extract or ethanolic fruit extract demonstrated that they have no genotoxic activity neither in mice nor in bacterial strains, although their cytotoxicity and antigenotoxicity were demonstrated in higher doses. In order to assess the possible compounds responsible for the activities observed, we fractionated the ethanolic fruit extract of S. paniculatum, characterized by 1H and 13C NMR spectra, and evaluated two fractions containing steroidal alkaloids... |
