Shellfish viruses ArchiMer
Renault, Tristan.
Mortalities in a number of commercially important mollusc species have been associated with the detection of viruses belonging to several families. The first description of a virus was in adult eastern oyters, Crassostrea virginica, with the detection of virus particles resembling members of the family Herpesviridae. Subsequently, mass mortalities in French stocks of adult Portuguese oysters, C. angulata, were associated with irido-like virus infection. Other viruses observed in molluscs are described as members of the families Iridoviridae, Papovaviridae and Reoviridae. Little information is available on viral infections that affect molluscs due to primarily to the inadequacy of diagnostic methods that are employed when mass mortality events occur. Most...

Created in 1986, the REPAMO network (Network of mollusc pathology), is a national zoosanitary surveillance network of shellfish health status along the French coasts. Its activities are in keeping with two European Directives, 91 /67/EEC and 95/70/EC and are a patt of the institutional tasks ofIfremer. The aims of the network are to prevent the introduction and spread of exotic pathogens and to survey the evolution of notifiable pathogens already present in France. The network focused on the survey of listed diseases (bonamiosis and marteiliosis of flat oyster) in two areas under agreement process (Banc de Granville and zone X). In 2004, no sample couId be analysed to these two areas because of the difficulty to obtain flat oysters (sparse oyster beds,...

Lysozyme gene expression and hemocyte behaviour in the Mediterranean mussel, Mytilus galloprovincialis, after injection of various bacteria or temperature stresses ArchiMer
Li, H; Parisi, M; Toubiana, Mylene; Cammarata, M; Roch, Philippe.
The aim of the present study was to evaluate the expression of the Mytilus galloprovincialis lysozyme gene in different in vivo stress situations, including injection of bacteria Vibrio splendidus LGP32, Vibrio anguillarum or Micrococcus lysodeikticus, as well as heat shock at 30 C and cold stress at 5 C. Injection of V. splendidus LGP32 resulted in: (i) a general down-regulation of lysozyme gene expression, as quantified by Q-PCR; (ii) reduction in the number of circulating hemocytes; (iii) decrease in the percentage of circulating hemocytes expressing lysozyme mRNA which was now restricted to only small cells, as observed by ISH; and (iv) accumulation of hemocytes expressing lysozyme in the muscle sinus where injection took place. Injection of V....

Les peuplements benthiques du plateau insulaire de la Martinique et des zones bathyales adjacentes. Cas particulier de la malacofaune ArchiMer
Paulmier, Gerard; Durand, Francoise.
Several oceanographic surveys in Martinique waters with, as main objectives, either geological mapping and evaluating marine aggregate resources of research into deep water carcinologie resources, nafe produced interesting informtion on benthic pppulations situated from the infralittoral to the bathyal shelf, for the wole of faunistic groups and more specifically for the malacofauna. The distribution of the various faunistic group has been analysed in function to the bathymetric, edaphic and possibly, thermic factors. The infralittoral zones, up to the 20 metres isobath, and circalittoral, between 60  and 80 metres, are known to have the richest populations. Corals, sponges, polychaete annelida and molluscs are the best represented, numerically as well as...

Viruses infecting marine molluscs ArchiMer
Arzul, Isabelle; Corbeil, Serge; Morga, Benjamin; Renault, Tristan.
Although a wide range of viruses have been reported in marine molluscs, most of these reports rely on ultrastructural examination and few of these viruses have been fully characterized. The lack of marine mollusc cell lines restricts virus isolation capacities and subsequent characterization works. Our current knowledge is mostly restricted to viruses affecting farmed species such as oysters Crassostrea gigas, abalone Haliotis diversicolor supertexta or the scallop Chlamys farreri. Molecular approaches which are needed to identify virus affiliation have been carried out for a small number of viruses, most of them belonging to the Herpesviridae and birnaviridae families. These last years, the use of New Generation Sequencing approach has allowed increasing...

Contrasted survival under field or controlled conditions displays associations between mRNA levels of candidate genes and response to OsHV-1 infection in the Pacific oyster Crassostrea gigas ArchiMer
Normand, Julien; Li, Ronghua; Quillien, Virgile; Nicolas, Jean-louis; Boudry, Pierre; Pernet, Fabrice; Huvet, Arnaud.
Pacific oyster Crassostrea gigas suffers from chronic or sporadic mortality outbreaks worldwide, resulting from infectious diseases and/or physiological disorders triggered by environmental factors. Since 2008, ostreid herpesvirus OsHV-1 μVar has been identified as the main agent responsible for mass mortality of juvenile oysters in Europe. Previous studies of genome-wide expression profiling have provided candidate genes that potentially contribute to genetically-based resistance to summer mortality. To assess their value in determining resistance to the juvenile mass mortality that has occurred in France since 2008, we analyzed the expression of 17 candidate genes in an experimental infection by OsHV-1 μVar, and in an in vivo field experiment. Individual...

In vitro culture of mollusc hemocytes. Functional study of burst respiratory activity and analysis of interactions with protozoan and procaryotic pathogens ArchiMer
Boulo, Viviane; Hervio, Dominique; Morvan, Annie; Bachere, Evelyne; Mialhe, Eric.
The chief importance of hemocytes as immune effectors in molluscs has led to develop researches for in vitro culture. The chemiluminescence technique has been used to estimate the phagocytosis capacity of Crassostrea gigas hemocytes according to different culture media and times. Sea water was used as a basic medium. Additives, such as antibiotics or glucose and buffers, such as Tris or Hepes, were tested as weIl as artificial sea water. Hemocyte chemiluminescence activities were recorded at t=o in relation to the medium composition. The highest values were observed when hemocytes were kept in sea water without any modification. Whichever medium, chemiluminescence activity was substantially decreased when hemocytes were previously kept in vitro for a few...

The role of microalgae in aquaculture: situation and trends ArchiMer
Muller-feuga, Arnaud.
Algae are utilized diversely in aquaculture, but their main applications are related to nutrition. They are used in toto, as a sole component or as a food additive to supply basic nutrients, color the flesh of salmonids or for other biological activities. The need for nutritional sources safer than traditional animal products has renewed interest in plants in general and algae in particular. This report deals principally with the nutritional role of microalgae in aquaculture. The larvae of molluscs, echinoderms and crustaceans as well as the live prey of some fish larvae feed on microalgae. Though attempts have been made to substitute inert particles for these micro-organisms which are difficult to produce, concentrate and store, only shrimp and live prey...

First evidence for a Vibrio strain pathogenic to Mytilus edulis altering hemocyte immune capacities ArchiMer
Ben Cheikh, Yosra; Travers, Marie-agnes; Morga, Benjamin; Godfrin, Yoann; Rioult, Damien; Le Foll, Franck.
Bacterial isolates were obtained from mortality events affecting Mytilus edulis and reported by professionals in 2010-2013 or from mussel microflora. Experimental infections allowed the selection of two isolates affiliated to V. splendidus/V. hemicentroti type strains: a virulent 10/068 1T1 (76,6% and 90% mortalities in 24h and 96h) and an innocuous 12/056 M24T1 (0% and 23,3% in 24h and 96h). These two strains were GFP-tagged and validated for their growth characteristics and virulence as genuine models for exposure. Then, host cellular immune responses to the microbial invaders were assessed. In the presence of the virulent strain, hemocyte motility was instantaneously enhanced but markedly slowed down after 2h exposure. By contrast, hemocyte velocity...

Scientific Opinion on an update on the present knowledge on the occurrence and control of foodborne viruses ArchiMer
A review of the biology, epidemiology, diagnosis and public health importance of foodborne viruses was performed. Data needs to support a risk assessment were also identified. In addition possible control options and their anticipated impact to prevent or reduce the number of foodborne viral human infections were identified, including the scientific reasons for and against the establishment of food safety criteria and process hygiene criteria for viruses for certain food categories. Food may be contaminated by virus during all stages of the food supply chain, and transmission can occur by consumption of food contaminated during the production process (primary production, or during further processing), or contaminated by infected food handlers. Transmission...

Contribution à l'étude de mollusques marins : Iridovirus-like et Herpèsvirus-like. Description, caractérisation biochimique, cycle de multiplication viral, diagnostic et étude épidémiologique ArchiMer
Renault, Tristan; Le Deuff, Rose-marie; Cochennec, Nathalie; Chollet, Bruno; Maffart, Patricia.
Among the marine molluscs, Pacifie oyster, Crassostrea gigas, is actually the most reared specie in the world. Noticely in France, where it was imported between 1960 and 1970, and replaced by Portuguese oyster, Crassostrea angulata, which specie was decimated during this period because of iridoviruses. Uncontroled introductions of new species may however be hazardous. Indeed, they might be responsible for the appearance and the dissemination of new diseases of bivalve molluscs in importing country. In addition to these observations, the quasi monobreeding of Pacific oyster in France enhances the necessity to develop research concerning the diagnosis of pathologies affecting this specie, namely the diagnosis of viral diseases. Several families of viruses...

Evaluation of the carrying capacity of molluscan shellfish ecosystems ArchiMer
Heral, Maurice.
Growth of molluscs raised in coastal zones, where the water exchange rate is low, appears to be limited, once a certain biomass level is attained, by the availability of food, making it necessary to determine as accurately as possible the trophic capacity of an ecosystem. This paper analyses the various sources of food for molluscs. It would appear that dissolved organic substances play a major role, providing up to half the energy necessary for their metabolism. Particulate material is filtered by molluscs in proportion to their size, with optimum filtration rates varying with species. The negative effect on meat production of high mineraI seston rates is demonstrated, and an analysis provided of the respective role of each component of organic matter....

Innovations in the nursery culture and maturation of bivalves ArchiMer
Flassch, Jean-pierre.
Innovations in the nursery culture and maturation of bivalves are being worked on at aIl levels of research and production, but transfer to the development stage must be effected differently in each case. Over the past twenty years, there have been a great many innovations in bivalve rearing, most of which may be described in terms of rearing conditions: higher and lower densities; confined or open environemnt; overflow; upwelling; floating, suspended, raised or ground systems, etc. The quality of an innovation is characterized by its influence on the efficient operation of the specific culture program and by how easily it may be transferred.

Le réseau de pathologie des mollusques. REPAMO ArchiMer
Grizel, Henri.
The creation of a mollusc pathology network was in effect and made official during 1992. The subject of this document is to present this network, which has not yet achieved its definitive size. Why this network? The creation of this network responds to several institutional and scientific needs. Indeed, the setting up of control measures aimed at regulating mollusc exchanges notably via European Directive 91/67 and its transcription into French law has, as a corollary, caused the recognition of IFREMER as the agency charged with performing the analyses necessary for responding to the needs of this Directive, as far as marine invertebrates are concerned. Moreover, France being among the primary producers of shellfish in the world, the setting up of a...

Viruses infecting bivalve molluscs ArchiMer
Renault, Tristan; Novoa, Beatriz.
Bivalve molluscs are filter feeders and as a consequence they may bioaccumulate in their tissues viruses that infect humans and higher vertebrates. However, there have also been described mortalities of bivalve molluscs associated with viruses belonging to different families. Mass mortalities of adult Portuguese oysters, Crassostrea angulata, among French livestocks (between 1967 and 1973) were associated with irido-like virus infections. Herpesviruses were reported in the eastern oyster, Pacific oyster and European flat oyster and lately in scallops and clams. Disseminated neoplasia, a proliferative cell disorder of the circulatory system of bivalves, although of uncertain aetiology, has been suggested to be caused by retroviral infections. Other viruses...
