Sodium azide mutagenesis resulted in a peanut plant with elevated oleate content Electron. J. Biotechnol.
Wang,Chuan Tang; Tang,Yue Yi; Wang,Xiu Zhen; Zhang,Shu Wei; Li,Gui Jie; Zhang,Jian Cheng; Yu,Shan Lin.
Screening of peanut seeds resulting from 0.39% sodium azide treatment with NIRS calibration equation for bulk seed samples identified a plant with more than 60% oleate. Oleate content in individual seeds of the plant, as predicted by NIRS calibration equation for intact single peanut seeds, ranged from 50.05% ~ 68.69%. Three seeds with >60% oleate thus identified were further confirmed by gas chromatography. Multiple sequence alignments of the FAD2B gene from Huayu 22 (wild type) and peanut seeds with elevated oleate (mutant type) revealed a C281T transition in the coding region causing an I94T substitution in the oleoyl-PC desaturase, which may be responsible for reduction in the enzyme activity.

Analysis of genomic sequences from peanut (Arachis hypogaea) Electron. J. Biotechnol.
B,Jayashree; Ferguson,Morag; Ilut,Dan; Doyle,Jeff; Crouch,Jonathan H.
Peanut is an important legume crop across the world. However, in contrast to most legume crops, groundnut lacks taxonomic proximity to any major model genome. A relatively large number of genomic sequences were generated from groundnut as part of a microsatellite marker development project. In the current study, a total of 1312 sequences were analyzed of which 448 contained microsatellite motifs. All sequences (GenBank Accessions: BZ999351-CC000573) were analyzed after clustering for possible similarity with publicly available sequences from Arabidopsis, Lotus, soybean and Medicago. At least 39% of the sequences analyzed had significant BLAST similarities with sequences from the four databases searched, of which nearly half (47%) found significant...

Novel protocol to identify true hybrids in normal oleate x high oleate crosses in peanut Electron. J. Biotechnol.
Wang,Chuan Tang; Yu,Shan Lin; Zhang,Shu Wei; Wang,Xiu Zhen; Tang,Yue Yi; Zhang,Jian Cheng; Chen,Dian Xu.
A novel hybrid identification protocol was developed for F0:1 peanut seeds resulting from crosses between normal oleate cultivars with wild type FAD2B gene and high oleate genotypes with an A insertion in FAD2B gene. Presence of a series of overlapped peaks in trace file of the PCR product amplified with bF19/R1 primers was an indication of hybridity. This protocol may facilitate high oleate breeding and genetic studies in peanut.

Identification of chilling-responsive transcripts in peanut (Arachis hypogaea L.) Electron. J. Biotechnol.
Tang,Yue Yi; Wang,Chuan Tang; Yang,Guan Pin; Feng,Tong; Gao,Hua Yuan; Wang,Xiu Zhen; Chi,Xiao Yuan; Xu,Ya Long; Wu,Qi; Chen,Dian Xu.
To isolate differentially expressed peanut genes responsive to chilling, a suppression subtractive hybridization (SSH) cDNA library was constructed for a chilling tolerant peanut cultivar A4 with mRNAs extracted from the seeds imbibed at 2ºC and 15ºC, respectively, for 24 hrs. A total of 466 cDNA clones were sequenced, from which 193 unique transcripts (73 contigs and 120 singlets) were assembled. Of these unique transcripts, 132 (68.4%) were significantly similar to the sequences in GenBank non-redundant (nr) protein database, which belonged to diverse functional categories including metabolism, signal transduction, stress response, cell defense and transcriptional regulation. The remaining 61 (31.6%) showed no similarity to either hypothetical or known...

Simple method to prepare DNA templates from a slice of peanut cotyledonary tissue for Polymerase Chain Reaction Electron. J. Biotechnol.
Yu,Shu Tao; Wang,Chuan Tang; Yu,Shan Lin; Wang,Xiu Zhen; Tang,Yue Yi; Chen,Dian Xu; Zhang,Jian Cheng.
An efficient DNA extraction method was developed for peanut seed, where only 3-5 mg cotyledonary tissue was enough for more than 50 PCR reactions with a reaction volume of 15 μl. Both low copy number and high copy number DNA sequences were successfully amplified. Processing one seed sample only took about half an hour. Sampling had no significant effects on germination and development. The DNA extraction method makes it possible to identify transformants and conduct molecular marker studies prior to sowing, and thus may greatly hasten research progress.

Cloning and analysis of a NBS-LRR disease resistance gene candidate PnAG1 from peanut (Arachis hypogaea L.) Electron. J. Biotechnol.
Shan,Shi-hua; Zhang,Ting-ting; Li,Chun-juan; Yang,Chen; Yan,Cai-xia; Wan,Shu-bo.
Background: Based on the conserved sequences of a known NBS resistance gene, a pair of degenerate primers was designed to amplify the NBS-LRR resistance gene from peanut using PCR and RACE methods. Results: Analyzing the amino acid sequence by BLAST on NCBI, which was deduced from the 1088bp-long gene named PnAG1-2, showed that it had a certain homology with some resistance proteins, among which Arachis cardenasii resistance protein gene had the highest homology (66%). Relative quantification PCR analysis indicated that PnAG1-2 gene expresses more in J11 (an A. flavus-resistant variety) than in JH1012 (an A. flavus-susceptible variety) when the harvest time was coming. Conclusions: In this study, the NBS-LRR resistance sequence was successfully cloned from...

POSITION ERRORS IN SOWING IN CURVED AND RECTILINEAR ROUTES USING AUTOPILOT REA
Santos,Adão F. dos; Correa,Lígia N.; Gírio,Lucas A. S.; Paixão,Carla S. S.; Silva,Rouverson P. da.
ABSTRACT The use of automatic routing significantly increases the yield and quality of agricultural operations and reduces the overlap rates between the mechanized set passes during the operation. The objective of this study was to analyse the quality of peanut sowing using automatic routing in two paths, curved and rectilinear, based on the error of parallelism and execution error, through statistical process control. The maps used came from an area of peanut seed production, in the city of Luzitânia, São Paulo, Brazil. The sowing was carried out by the execution of a project elaborated in CAD software that, for its elaboration, was taken into consideration the working width of the seeder-fertilizer, as well as the spacing of the crop. The CenterPoint RTX...

Genetic diversity in section Rhizomatosae of the genus Arachis (Fabaceae) based on microsatellite markers Genet. Mol. Biol.
Angelici,Carla M.L.C.D.; Hoshino,Andrea Akemi; Nóbile,Paula Macedo; Palmieri,Dario Abel; Valls,José F. Montenegro; Gimenes,Marcos A.; Lopes,Catalina Romero.
The genus Arachis (Fabaceae) native to South America, contains 80 species divided into nine sections, three of which contain species of special economic importance such as the cultivated peanut (Arachis hypogaea), belonging to the section Arachis, and some perennial forage species from sections Caulorrhizae and Rhizomatosae. We used microsatellite markers to assay genetic variability among 77 accessions of four species from section Rhizomatosae, the diploid Arachis burkartii (2n = 2x = 20) and the tetraploid Arachis glabrata, Arachis pseudovillosa and Arachis nitida (2n = 4x = 40). A total of 249 alleles were found in the fifteen loci analyzed and a high degree of intra and interspecific polymorphism was detected. The lowest intraspecific variation...

Successful crosses between fungal-resistant wild species of Arachis (section Arachis) and Arachis hypogaea Genet. Mol. Biol.
Fávero,Alessandra Pereira; Santos,Rodrigo Furtado dos; Simpson,Charles E.; Valls,José Francisco Montenegro; Vello,Natal Antonio.
Peanut (Arachis hypogaea) is the fifth most produced oil crop worldwide. Besides lack of water, fungal diseases are the most limiting factors for the crop. Several species of Arachis are resistant to certain pests and diseases. This study aimed to successfully cross the A-genome with B-K-A genome wild species previously selected for fungal disease resistance, but that are still untested. We also aimed to polyplodize the amphihaploid chromosomes; cross the synthetic amphidiploids and A. hypogaea to introgress disease resistance genes into the cultivated peanut; and analyze pollen viability and morphological descriptors for all progenies and their parents. We selected 12 A-genome accessions as male parents and three B-genome species, one K-genome species,...

Heterologous microsatellite primer pairs informative for the whole genus Arachis Genet. Mol. Biol.
Hoshino,Andrea Akemi; Bravo,Juliana Pereira; Angelici,Carla M.L.C.D.; Barbosa,Andréa V. Gobbi; Lopes,Catalina Romero; Gimenes,Marcos A..
The genus Arachis currently comprises 69 described species, some of which have potential and real value as human and animal foods. These Arachis species have been collected and maintained in germplasm banks to provide material that can be used as sources of genes in breeding programs and for the selection of new cultivars. One of the principal objectives of germplasm conservation is the evaluation of genetic variability, which is best conducted using molecular markers. We investigated the use of heterologous primers to amplify microsatellite loci that could be used to evaluate genetic variability in Arachis germplasm. Fifteen microsatellite primer pairs were tested in 76 accessions of 34 species from the nine Arachis sections. The data indicated that...

Genetic relationships among Arachis species based on AFLP Genet. Mol. Biol.
Gimenes,Marcos A.; Lopes,Catalina R.; Valls,Jose F.M..
Amplified Fragment Length Polymorphism (AFLP) was used to establish the genetic relationships among 20 species from seven of the nine sections of genus Arachis. The level of polymorphism among nine accessions of the cultivated peanut, A. hypogaea L., was also evaluated. Three combinations of primers were used to amplify the AFLPs. The fragments were separated in 6% denaturing acrylamide gels. A total of 408 fragments were analyzed. An average of 135.3 fragments per primer combination were scored, and the largest number of fragments was 169 using primer combination Eco RI - ACC / Mse I - CTG, while the lowest was 108, with Eco RI - ACT / Mse I - CTT. In general, the genetic relationships established using AFLPs agreed with the classification established...

Transferability and use of microsatellite markers for the genetic analysis of the germplasm of some Arachis section species of the genus Arachis Genet. Mol. Biol.
Bravo,Juliana Pereira; Hoshino,Andrea Akemi; Angelici,Carla Maria Lara C.D.; Lopes,Catalina Romero; Gimenes,Marcos Aparecido.
The Arachis section is the most important of the nine sections of the genus Arachis because it includes the cultivated peanut, Arachis hypogaea. The genetic improvement of A. hypogaea using wild relatives is at an early stage of development in spite of their potential as sources of genes, including those for disease and pests resistance, that are not found in the A. hypogaea primary gene pool. Section Arachis species germplasm has been collected and maintained in gene banks and its use and effective conservation depends on our knowledge of the genetic variability contained in this material. Microsatellites are routinely used for the analysis of genetic variability because they are highly polymorphic and codominant. The objective of this study was to...
