Paralytic phycotoxin uptake by scallops (Pecten maximus) ArchiMer
Lassus, Patrick; Bardouil, Michele; Ledoux, Martial; Murail, Ingrid; Bohec, Madeleine; Truquet, Philippe; Frémy, Jean-marc; Rohmer, Véronique.
It is difficult to apply sanitary standards for mollusc contamination by paralytic shellfish poisoning to animals not consumed whole. This problem is illustrated by the 1990 embargo on a Japanese shipment of frozen scallop muscle and gonad: the sanitary threshold applied to digestive gland (in Japan) does not guarantee that muscle and especially gonad are toxin-free. Accordingly, we performed experimental contaminations of scallops (Pecten maximus) from Port-en-Bessin (Normandy, France) using a toxic Japanese strain of Alexandrium tamarense. During the contamination/decontamination experiment on different tissues (digestive gland, muscle, gonad), extracts were obtained using cold acetic acid 0.1 N to maintain the perfect integrity of the toxin profile....

Vigneau, Joel; Fifas, Spyros; Foucher, Eric.
The methodology used for obtaining absolute abundance indices for Eastern Channel scallop is described in this paper. For the survey (COMOR), stratified multi-stage random sampling is used. The number of sampled units is computed based on variance estimates from the last survey. Knowledge of gear (Brittany dredge with a diving plate) selectivity and efficiency parameters is required for estimating total exploitable biomass and absolute abundance indices by age and strata. Details of the mathematical development of all estimators are given. A non parametric bootstrap provides confidence intervals for all estimates.

Experimental shift in diet Delta13C: A potential tool for ecophysiological studies in marine bivalves ArchiMer
Paulet, Yves-marie; Lorrain, Anne; Richard, Joëlle; Pouvreau, Stephane.
To test the potential of diet switching experiments in ecophysiological studies of marine invertebrates, stable carbon isotope ratios were measured at different seasons in the gonad, adductor muscle, digestive gland and gills of scallops (Pecten maximus) and oysters (Crassostrea gigas) held for 15 days on a constant diet of phytoplankton depleted in 13C. The aim of this study was to determine if differences in carbon incorporation could be detected among species, seasons and organs, and if so, whether it was consistent with their known energy-allocation patterns. After offering the new diet, isotope values of the different organs gradually shifted and significant differences among organs, seasons and species were found. A carbon incorporation index (CII)...

Foucher, Eric; Quinquis, Jerome; Ton, Caroline.
The SELEDRAG project was conducted in French territorial waters in May 2019. It was a short project, lead by the CNPMEM (French National Committee for Marine Fisheries), with Ifremer as scientific partner, and funded by “France Filière Pêche”. The context of the project is the French King Scallop fishery in the Eastern Channel, where 720 fishing boats are directly concerned by this species. Despite its importance, there is very little management for this species at the European level, but several binding rules for French fishermen in order to protect the stock, are not applicable to other countries. One of the French specificity is the ring size used, 92mm for all Scallop fisheries in France vs 85mm (or less) for UK fisheries. Another sharp divergence is...

Modelling benthic invasion by the colonial gastropod Crepidula fornicata and its competition with the bivalve Pecten maximus . 1. A new 0D model for population dynamics of colony-forming species ArchiMer
Menesguen, Alain; Gregoris, Thomas.
The north-American marine gastropod Crepidula fornicata (Linné 1758), commonly called slipper limpet, has been introduced accidentally in Great Britain, along with American oysters (Crassostrea virginica) at the end of the 19th century. Its spreading, enhanced by oyster transport and international shipping, turned into invasion of a lot of benthic grounds along the North-European coasts. When local conditions are favorable to larval confinement and adult growth, the invasion by very dense populations may deeply change initially sandy or muddy benthic communities into similar “slipper limpet communities”. Competition for space eliminates several species of native infauna, and hampers the survival of some epibenthic bivalves such as the scallop Pecten...

Growth anomalies in Pecten maximus from coastal waters (Bay of Brest, France): relationship with diatom blooms ArchiMer
Lorrain, Anne; Paulet, Yves-marie; Chauvaud, Laurent; Savoye, Nicolas; Nezan, Elisabeth; Guerin, Laurent.
The aim of this study was to characterize the daily shell growth of Pecten maximus from early 1998 to late spring 1999 in the Bay of Brest with a careful qualitative and quantitative description of the pelagic primary production. Our results, in accordance with previous studies, demonstrate that there are episodic declines in the growth rate. Concurrent events of low growth rate and large bottom-concentrations of algae following diatom blooms (Cerataulina pelagica or Rhizosolenia delicatula), suggests that this high concentration of phytoplankton on the bottom layer may affect food intake or respiratory activity of the scallops by gill clogging or oxygen depletion. In this study, silicic acid or phosphorus are inferred to be limiting factors responsible...

We combined the use of pigments, fatty acids and sterols as biomarkers of the seasonal variation in food sources of the great scallop Pecten maximus. From March to October 2011, on a biweekly to twice-weekly basis, scallops and seawater from the water column and the water-sediment interface were collected in the Bay of Brest (Brittany, France). Pigment compositions in the seawater and in the stomach and rectum content of the scallops were analyzed by HPLC. Fatty acids and sterols from digestive gland (DG) tissue were analyzed by gas chromatography. Potential relationships between the temporal proportion of each marker in the environment and in the digestive tract were tested using multivariate analysis. Proportions of diatoms and Dinophyceae biomarkers...

Optimizing sperm cryopreservation in great scallop (Pecten maximus) ArchiMer
Suquet, Marc; Malo, Florent; Gourtay, Clemence; Ratiskol, Dominique; Riobe, Marion; Le Grand, Jacqueline; Queau, Isabelle; Mingant, Christian.
A basic cryopreservation protocol for great scallop sperm was recently published, with which thawed sperm survival remained limited to 25% of control values (Suquet et al. 2014). We designed here a set of five experiments to test ways of improving thawed sperm survival and storage by studying the effects of (1) cryoprotectant concentration; (2) sugar incorporation in the extender; (3) egg yolk addition to the extender; (4) sperm dilution rate; and (5) storage capacity of thawed sperm at 4 degrees C. A sixth experiment was then performed to compare rearing performances of larvae produced using thawed sperm from the optimal protocol defined in the present study with those produced using fresh sperm. During the first experiment, a significantly higher...

Robert, Rene; Miner, Philippe; Nicolas, Jean-louis; Mazuret, Michel; Connan, Jean-paul.
Larval mortalities occurring in molluscan hatcheries have often been associated with bacterial contamination. Although batches of oyster and clam larvae have been routinely reared in the hatchery of Argenton without antibiotics, high larval mortalities have been recorded with Pecten maxirnus under similar conditions. For this species, an addition of chloramphenicol was found necessary. However, this chemical has now been banned in Europe. Thus either substitition products or an improvement in the rearing procedures is essential. Studies carried out have shown that neither a decrease in larval density (1 larvae. ml"l) nor sea water change frequency (1 per day) had any positive effects. Furthermore, elective substances such as sugars were not suitable and...

Effect of sampling location, release technique and time after activation on the movement characteristics of scallop (Pecten maximus) sperm ArchiMer
Suquet, Marc; Quere, Claudie; Mingant, Christian; Lebrun, Luc; Ratiskol, Dominique; Miner, Philippe; Cosson, Jacky.
Sperm characteristics of scallops have not been well described in the scientific literature. The effects of sperm release technique (thermal shock versus serotonin injection), of sperm collection technique (testis sampling versus serotonin injection), of sperm sampling location along the genital tract, of in vitro sperm maturation, and of time post activation on scallop sperm characteristics were assessed in the present work. Whatever sperm release technique used, no significant differences were observed regarding the percentage of motile spermatozoa and the velocity of the average path (YAP). Compared to testicular sperm, a higher percentage of motile spermatozoa, YAP and intracellular adenosine triphosphate (ATP) content were observed for sperm shed...

Bivalve hatchery technology: The current situation for the Pacific oyster Crassostrea gigas and the scallop Pecten maximus in France ArchiMer
Robert, Rene; Gerard, Andre.
The French oyster farming industry relies almost exclusively on juveniles collected from the natural environment; the supply of spat produced by hatcheries is low, about 10% of the industry's requirements. Development through selective breeding of oyster stocks which are better suited for aquaculture purposes, is likely to reverse this tendency since only hatcheries will be able to supply such animals. Scallop farming, which at present is poorly developed in France, relies exclusively on hatchery produced spat. Although hatchery technology is constantly being improved, significant production problems remain which must be solved before hatcheries become a major supplier of juveniles for the industry. This paper describes the present state of hatchery...

Buestel, Dominique; Cochard, Jean-claude; Gerard, Andre.

Foucher, Eric.
The assessment of the King scallop stock of the Bay of Seine (Pecten maximus) was carried out in July 2018, in the Bay of Seine sensu stricto, located in French territorial waters (from the Normandy coast to the south to the limit of 12 nautical miles to the North), as well as the bordering zone lying north of this 12-mile limit and up to a latitude of 49°48N to the North, called "Extérieur Baie de Seine". For this, a scientific survey, based on a stratified random sampling plan, was conducted aboard the F/R Thalia, a coastal research vessel of the French Oceanographic Fleet. Nearly 170 sampling points were completed in the whole area. Analysis of these data leads to an overall increase in exploitable biomass over the entire area. In the area “Exterieur...

Holbach, Marine.
In many countries, aquaculture of pectinids depends on the success of artificial spat production in hatchery. This production is always unpredictable due to the variability of hatching rate and larval survival. Flow-through larval rearing systems were developed in Norway for the King scallop Pecten maximus and showed promising results. Unfortunately the system needs to be optimized since the larval yields and the densities used are still relatively low. In France, a small-scale (5 L) and high-density (≤ 300 larva mL-1) flow-through larval rearing system was successfully developed for oysters. First trials in such system and in similar conditions with P. maximus failed as we registered slower growth and high mortality rate in only a few days. It is known...

Genetic structure of a commercially exploited bivalve, the great scallop Pecten maximus, along the European coasts ArchiMer
Morvezen, Romain; Charrier, Gregory; Boudry, Pierre; Chauvaud, Laurent; Breton, Florian; Strand, Oivind; Laroche, Jean.
The great scallop Pecten maximus is harvested in several European countries and fisheries targeting this species are severely regulated by fishing quotas. In addition, hatchery-based population enhancement has been developed in some countries to provide alternative or complementary production. The genetic structure of wild populations of P. maximus and the potential impact of aquaculture on the genetic diversity of this species remains poorly documented. In this study, we explored the genetic structure of P. maximus using 12 microsatellite markers, considering 14 populations sampled from Galicia (Spain) to the North of Norway, and one population of Pecten jacobaeus (L., 1758) from the Lion Gulf (Mediterranean Sea). Results indicated a clear differentiation...

Effects of bioactive extracellular compounds and paralytic shellfish toxins produced by Alexandrium minutum on growth and behaviour of juvenile great scallops Pecten maximus ArchiMer
Borcier, Elodie; Morvezen, Romain; Boudry, Pierre; Miner, Philippe; Charrier, Gregory; Laroche, Jean; Hegaret, Helene.
Dinoflagellates of the genus Alexandrium are a major cause of harmful algal blooms (HABs) that have increasingly disrupted coastal ecosystems for the last several decades. Microalgae from the genus Alexandrium are known to produce paralytic shellfish toxins (PST) but also bioactive extracellular compounds (BEC) that can display cytotoxic, allelopathic, ichtyotoxic or haemolytic effects upon marine organisms. The objective of this experimental study was to assess the effects of PST and BEC produced by A. minutum upon juvenile great scallops Pecten maximus. Scallops were exposed for one week to two different strains of A. minutum, the first producing both PST and BEC and the second producing only BEC. Escape response to starfish, daily shell growth,...

Lepennec, M; Dorange, G; Beninger, P; Donval, A; Widowati, I.
Histological, histochemical and ultrastructural studies were performed on the region of the gonad traversed by the intestine in Pecten maximus . The results indicate a digestive and absorptive function of intestine epithelial cells and a transfer of metabolites to the surrounding gonad tissue. Several possible mechanisms of translocation are presented, including diffusion into the haemolymph and transfer via vacuolated cells of the gonad interstitial connective tissue.
