En Yurécuaro, Michoacán, en el ciclo de cultivo verano otoño 2009 y 2011 se encontraron en cultivos de chile síntomas de variegado, nervaduras cloróticas, deformación de la lámina foliar y fruto, albinismo y escaldadura, síndrome al cual se le denominó “variegado del chile”. A simple vista, esta serie de síntomas indicaban que el problema era causado por virus fitopatógenos; sin embargo, en ensayos previos se detectó a Candidatus Liberibacter solanacearum, por lo que el objetivo del presente trabajo fue determinar si esta bacteria es el agente causal del variegado del chile. LaNe RAGE: a new tool for genomic DNA flanking sequence determination Electron. J. Biotechnol.

The determination of genomic DNA sequence flanking a known region is often problematic. Existing technologies depend on multiple, efficient enzyme-catalysed preparative processing steps and/or rely on relatively inefficient ‘one-sided' PCR mechanisms. I demonstrate the application of a simple ‘two-sided' PCR-based approach, lariat-dependent nested PCR for rapid amplification of genomic DNA ends (LaNe RAGE), applied to the mouse GAPDH and PGK1 gene flanking sequences. This demonstration offers great promise in applications such as genome walking, transposon mutagenesis mapping and DNA fingerprinting

We hereby present the complete sequence and annotation of pRG930cm, a spectinomycin/streptomycin/chloramphenicol-resistant cosmid vector. pRG930cm (17,256 bp; GenBank Accession No.: FM174471) has a broad host range, and is stably maintained by a number of Gram-negative bacteria including Pseudomonas spp, Escherichia coli, Agrobacterium tumefaciens and Azorhizobium caulinodans ORS571. pRG930cm is already widely used and its sequence will aid efficient construction and analysis of cosmid libraries.

Monocelis lineata consists of a complex of sibling species, widespread in the Mediterranean and Atlantic Ocean. Previous genetic analysis placed in evidence at least four sibling species. Nevertheless, this research was not conclusive enough to fully resolve the complex or to infer the phylogeny/phylogeography of the group. We designed specific primers aiming at obtaining partial sequences of the mtDNA gene Cytochrome c Oxidase subunit I (COI) of M. lineata, and have identified 25 different haplotypes in 32 analyzed individuals. Isolation of endophytic bacteria from arboreal species of the Amazon and identification by sequencing of the 16S rRNA encoding gene Genet. Mol. Biol.

Endophytic bacteria from three arboreal species native to the Amazon (Carapa guianenses, Ceiba pentandra, and Swietenia macrophylla), were isolated and identified, through partial sequencing of the 16S rRNA encoding gene. From these, 16 isolates were obtained, although, when compared to sequences deposited in GenBank, only seven had produced identifiable fragments. Bacillus, Pantoea and two non-culturable samples were identified. Results obtained through sequence analysis revealed low genetic diversity across the isolates, even when analyzing different species and plant structures. This is the first report concerning the isolation and identification of endophytic bacteria in these plant species.

Molecular cloning and sequence analysis of growth hormone cDNA of Neotropical freshwater fish Pacu (Piaractus mesopotamicus) Genet. Mol. Biol.

RT-PCR was used for amplifying Piaractus mesopotamicus growth hormone (GH) cDNA obtained from mRNA extracted from pituitary cells. The amplified fragment was cloned and the complete cDNA sequence was determined. The cloned cDNA encompassed a sequence of 543 nucleotides that encoded a polypeptide of 178 amino acids corresponding to mature P. mesopotamicus GH. Comparison with other GH sequences showed a gap of 10 amino acids localized in the N terminus of the putative polypeptide of P. mesopotamicus. This same gap was also observed in other members of the family. Among 1280 cultured and wild adult Mediterranean mussels, Mytilus galloprovincialis, collected over a 1-year surveillance period from the Slovene Adriatic Sea, 0.3% were histologically positive for the presence of Marteilia spp. The infection was concentrated in winter. Employing the molecular methods of PCR, cloning, DNA restriction and sequencing, only Marteilia refringens type M was detected in all the infected mussels. To explore the microbial diversity at deep sea vents, molecular techniques based on the analysis of the gene coding the 16S ribosomal RNA permitted to highlight a large archaeal and bacterial diversity. Nevertheless, the diversity of the hydrothermal microorganisms cultivated to date remains much lower than the diversity described with molecular tools. Innovative methods should therefore be used in order to cultivate new microorganisms, and also determine their metabolic properties and their potential role in the ecosystem. Genetic differentiation within the salmon louse Lepeophtheirus salmonis (Krøyer, 1837), was investigated by the sequencing of specific nucleotide regions. Partial sequences of the 18S ribosomal RNA gene and the ribosomal internal transcribed spacer (ITS-1) region from single sea lice were amplified by the polymerase chain reaction (PCR). None of the previous genetic analysis found significant differences within the Mediterranean. However, a more exhaustive analysis is needed inside this sea. In this sense, we have analyzed the mitochondrial control region sequence variability of three Mediterranean locations: Libya (n = 22), Tunisia (n = 23) and Malta (n = 12). Additionally a sample from the Gulf of Cadiz (n = 24) in the East Atlantic Ocean was also included in the study. This study aimed to genetically characterize four new Rhizobium strains, and to evaluate their nodulation and fixation capacity compared to commercial strains and to native rhizobia population of a Brazilian Rhodic Hapludox. Two experiments were carried out in randomized blocks design, under greenhouse conditions, in 2007. In the first experiment, the nodulation and nitrogen fixation capacity of new strains were evaluated, in comparison to the commercial strains CIAT-899 and PRF-81 and to native soil population. It was carried out in plastic tubes filled with vermiculite. Avian Infectious Laryngotracheitis, caused by Infectious Laryngotracheitis Virus (ILTV), has been reported for decades in Brazilian laying and broiler flocks. More recently, outbreaks have occurred in São Paulo State. This study reports the application of PCR and DNA sequencing targeted to the p32 gene of ILTV using laying chicken samples from Bastos, São Paulo, Brazil. Three out of four field samples were positive by PCR. DNA sequencing of two samples evidenced homology of the amplified fragments with the p32 gene of ILTV. The results definitely confirmed the presence of ILTV in the birds during the outbreak. The objective of this study was to partially characterize some genes involved in the desiccation tolerance of the embryonic axis of Melanoxylon brauna seeds subjected, or not, to oven fast-drying. Seeds were initially dried rapidly in an oven at 40 ºC, 50 ºC, 60 ºC, 70 ºC, and 80 °C, for 24, 48 and 72 h and then subjected to germination tests and moisture content determination. Degenerate primers were designed for 19 genes. The CDNA was used as a template for PCR amplifications using the degenerate primers, and the PCR products obtained were purified, cloned and sequenced. The objective of the present study was to report the molecular cloning and determination of the sequences N-methyl-D-aspartate subunit-1 receptor (NMDAR-1) in the hypothalamus of sheep. The re-amplified DNA template for NMDA -1 from the hypothalamus of male sheep was cloned using pBluescript-Sk-plasmids (pBSK, 2958 bp). Purified plasmids containing the NMDA receptor c-DNA were sequenced using the dye-terminator chemistry at the University of Nebraska-Lincoln, Nebraska, USA. Bemisia tabaci (Genn.) is one of the most important pests in cultivated areas of vegetables and ornamental crops around the world. Based on the mitochondrial cytochrome oxidize I (mtCOI) sequence, there is evidence that B. tabaci should be considered a cryptic species complex of 11 groups containing 24 species. Two of the groups, Middle East-Asia Minor 1 and Mediterranean include biotypes B and Q, respectively. Drought stress could induce expressions of various genes that are involved in stress tolerance and responses. Recent studies were emphasized on a gene encoded for the 9-cis epoxycarotenoid dioxygenase (NCED) enzyme which catalyzes the biosynthesis of abscisic acid (ABA) in plants. However, little is known about drought tolerance genes in maize. We designed primers from the VP14 sequence in the NCBI gene bank for the study of expression of drought tolerance gene in SW 2301 drought tolerant and KSX 4605 drought sensitive lines. Tomato chlorotic spot virus in hydroponically-grown lettuce in São Paulo State, Brazil Trop. Plant Pathol.

In the regions of Campinas and Sumaré, São Paulo, Brazil, hidroponically grown crops of Lettuce (Lactuca sativa) cv. Verônica, which showed virus-like symptoms were examined by electron microscope, biological, serological and molecular tests. Pleomorphic, enveloped particles (80-100 nm in diameter) were always detected in these samples. Experimentally inoculated host plants, including lettuce, reacted with tospoviruses-induced symptoms. Some differences were observed in Gomphrena globosa, which reacted by showing local lesions and systemic mosaic. Abstract The main open-field and greenhouse producer regions of tomato in Panama (Los Santos, Herrera, Veraguas and Chiriquí provinces) were surveyed to determine the occurrence and distribution of Bemisia tabaci and Trialeurodes vaporariorum during the growing seasons of 2017 and 2018. Twenty-two plots were surveyed, and DNA extracts of 112 adult whiteflies were analyzed by multiplex PCR, using a set of specific primers of B. tabaci MEAM1, B. tabaci NW, and T. vaporariorum. Digitaria species are sugar cane crop weeds in Brazil and are being controlled with herbicides, although there are some reports of control failure, notably to the triazine group. Molecular techniques are recommended to analyze the genetic variability in weeds. Genetic and taxonomic relationships of five species of Rallidae (Aves: Gruiformes) based on mitochondrial cytochrome oxidase subunit I sequences Animal Sciences

This is the first study to detection the genetic relationship between Porphyrio alleni and four Rallidae species: Fulica atra, Gallinulla angulata, Gallinulla chloropus and Porphyrio porphyrio. Mitochondrial cytochrome oxidase subunit I (COI) sequences were used as an effective marker in this study. DNA of Rallidae species were extracted, amplified using polymerase chain reaction (PCR) and then sequenced. The results obtained from information based on COI sequences revealed that Gallinulla angulata and Gallinulla chloropus fall into two separate clades and they are not monophyletic.
