Inicialmente... |

This paper describes an efficient bacterial transformation system that was established for the preparation of competent cells, plasmid preparation, and for the storage in bacterial stocks in our laboratory. Using this method, a number of different plasmids have been amplified for further experiments. Competent cells for bacterial transformation were prepared by the calcium chloride method with an optimum concentration of 75 mM. Three different strains of Escherichia coli that were tested are DH5α, TG1 and XL1 blue, and the most efficient strain being XL1 blue. The optimal optical density (OD600) range for competent cell preparation varied for each of the strains investigated, and for XL1 blue it was 0.15-0.45; for TG1 it was 0.2-0.5; and for... |

ABSTRACT This paper aims at evaluating the shelf life of mini tomatoes (Lycopersicum esculentum Mill.) cultivar ‘Sweet Grape’, grown in hydroponics, and stored under environmental and refrigerated conditions inside different packages. We adopted a completely randomized design, in which treatments were combinations of storage conditions: environment (e) and refrigerated (r) with packaging: polyvinyl chloride film (PVC); low-density polyethylene (LDPE); biofilm of tomato fruit of Solanum lycocarpum A.St.-Hil (lobeira) (TFB); cassava starch biofilm (CSB); carnauba wax (Copernicia prunifera) (CW), and without packaging - control (C). Physicochemical and sensory tests were carried out at the beginning (day zero), and at 8, 19, and 33 days of storage (DS). Fruit... |

ABSTRACT Peach palm heart (Bactris gasipaes) is a highly perishable product and, therefore, studies on extending its shelf life are quite necessary. Considering the high demand of this vegetable, this study aimed to evaluate the influence of using different plastic packages in order to extend the shelf life of the peach palm heart in natura. Stipes were collected from plants grown on family farms located in the municipality of Seropédica-RJ. Stipes were peeled, sanitized and packed at the Post-harvest Laboratory in Universidade Federal Rural do Rio de Janeiro (UFRRJ). In this experiment, the following treatments were applied: a control without package (C), unperforated polyethylene bag (UPB), perforated polyethylene bag (PPB), microperforated polyethylene... |

ABSTRACT The aim of this work was to evaluate effects of different gas concentrations (O2 and CO2) on conservation of minimally processed eggplants. The eggplants were sanitized, processed and packed in nylon + transparent polyethylene. The vegetables were submitted to modified atmospheres (T1= atmospheric air; T2= vacuum; T3= 4% O2 and 5% CO2; T4= 4% O2 and 6% CO2; T5= 4% O2 and 7% CO2; T6= 4% O2 and 8% CO2). The minimally processed eggplants were kept at 5±1°C and 90±1% RH, for 10 days. After 0, 2, 4, 6, 8 and 10 days of storage, fresh weight loss, respiratory rate, color, soluble solids (SS), titratable acidity (TA), pH, SS/TA ratio, visual analysis and purchase intent were determined. We used completely randomized experimental design, in a factorial... |

ABSTRACT Industrial and organic residues cause environmental impacts at various levels. However, these residues present potential to correct soil acidity and fertility. Considering the possibility of using these alternative sources, the objective of this study was to analyze the quality of scarlet eggplant seeds produced with the use of industrial waste. The experiment was carried out in a greenhouse. We used scarlet eggplant seeds (Solanum gilo), cv. ‘Verde-Claro’, and the soil was Typic Hapludox. The experimental design was randomized blocks, in a factorial distribution (3x4x5), being three soil variations [(pure soil (S), soil + cattle manure (SE) and, soil + sewage sludge (SL)], four corrective treatments [without correction (C1), limestone (C2), steel... |
