Environmental quality of an estuary in Amazon delta using immunohistochemical and morphological analyses of gill as biomarkers Biological Sciences
Montes, Caroline da Silva; Ferreira, Maria Auxiliadora Pantoja; Santos, Simone do Socorro Damasceno dos; Rocha, Rossineide Martins.
In this study, histological, ultrastructural, and immunohistochemical observations of the gill tissue of two fish species (Plagioscion squamosissimus and Lithodoras dorsalis) were used as biomarkers to assess the ecological conditions of an estuary in the Amazon delta, by relating the health status of the fishes to environmental quality. The gill tissue was analyzed both qualitatively and semi-quantitatively. Marked differences in the type and severity of lesions were verified between species and among study sites. A variety of severe damages were confirmed histologically in specimens from the two impacted areas (B and C), including aneurysms, epithelial lifting, and intense hyperplasia. Electron microscopy and immunohistochemical analyses also revealed...

Oocyte surface in four teleost fish species postspawning and fertilization BABT
Rizzo,Elizete; Moura,Thais F.C.; Sato,Yoshimi; Bazzoli,Nilo.
Cytological and cytochemical studies were carried out to investigate the surface characteristics of oocytes of four teleost species from the São Francisco river. The fishes were submitted to hypophysation at the Três Marias Hybrobiology and Fishculture Station, Minas Gerais, Brazil, in January 1996. Postspawning, oocytes of the curimatãs Prochilodus affinis, Prochilodus marggravii and dourado Salminus brasiliensis were surrounded by a thick, three-layered zona pellucida with radial striae. The surface of spawned oocytes of the surubim, Pseudoplatystoma coruscans, was comprised of mucous coat located externally to a thin, two-layered and striated zona pellucida. Oocyte activation during fertilization, lead to cortical reaction, formation of a perivitelline...

Epithelial gill cells in the armored catfish, Hypostomus cf. plecostomus (Loricariidae) BJB
FERNANDES,M. N.; PERNA-MARTINS,S. A..
Epithelial gill cell morphology and distribution were investigated in the armored catfish, Hypostomus cf. plecostomus, which lives in soft ion-poor Brazilian freshwaters. Pavement cells are the most abundant type of cell on both filament and lamellar epithelia and there are a great number of mucous and chloride cells between them. Mucous cells are almost covered by adjacent pavement cells and have large packed granules showing electrondense differences. No mucous cells were found on the lamellar epithelium. Chloride cell were distributed throughout both epithelia and usually have large apical surface facing the external medium and may exhibit short and sparsely distributed microvilli. The presence of chloride cells on the lamellar epithelium may be an...

Annual changes in serum calcium and inorganic phosphate levels and correlation with gonadal status of a freshwater murrel, Channa punctatus (Bloch) BJMBR
Srivastav,S.K.; Srivastav,A.K..
Adult Channa punctatus murrels of both sexes (60-80 g) were collected locally from Ramgarh Lake during the second week of every month (10 individuals of each sex/month) throughout the year. Blood samples were collected and analyzed for serum calcium and phosphate levels by the methods of Trinder (1960) and Fiske and Subbarow (1925), respectively. Gonads were fixed to judge the state of maturation of the fish. Males exhibited no change in serum calcium levels throughout the year in correlation with testicular maturation. However, serum phosphate levels exhibited a rise in correlation with the increased gonadosomatic index. Females showed marked seasonal changes in serum calcium and phosphate levels which were associated with ovarian maturation...

Chlorpheniramine facilitates inhibitory avoidance in teleosts submitted to telencephalic ablation BJMBR
Faganello,F.R.; Mattioli,R..
The present study investigated the involvement of H(1) histaminegic receptor on the acquisition of inhibitory avoidance in Carassius auratus submitted to telencephalic ablation. The fish were submitted to telencephalic ablation 5 days before the experiment. The inhibitory avoidance procedure included 1 day for habituation, 3 days for training composed of 3 trials each (1st day: T1, T2, T3; 2nd day: 2T1, 2T2, 2T3; 3rd day: 3T1, 3T2, 3T3) and 1 day for test. On training days, the fish were placed in a white compartment, after 30 s the door was opened. When the fish crossed to a black compartment, a weight was dropped (aversive stimuli). Immediately after the third trial, on training days, the fish received, intraperitoneally, one of the pharmacological...

A mathematical model for growth in weight of silver catfish (Rhamdia quelen) (Heptapteridae, Siluriformes, Teleostei) Ciência Rural
Benaduce,Ana Paula da Silva; Rodrigues,Luiz Alberto Díaz; Mistro,Diomar Cristina; Baldisserotto,Bernardo.
The use of a mathematical model applied to biological science helps to predict the specific data. Based on biological data (weight and age) of silver catfish, Rhamdia quelen, a mathematical model was elaborated based on a nonlinear difference equation to demonstrate the relationship between age and growth in weight. Silver catfish growth was described following the Beverton-Holt model Pt+1 = (r Pt) / (1+ a Pt ), where r > 0 is the maximum growth rate and a > 0 is a constant of growth inhibition. The solution of this equation is Pt= 1 /{[1/P0 - a / (r-1)] 1/r t + a/ (r-1)}, were P0 is the initial weight of the fish. Through this model it was observed that the female reaches the theoretical maximum weight approximately at the age of 18 years and the...

Simultaneous overexpression of GH and STAT5b genes inhibits the STAT5 signalling pathway in tilapia (Oreochromis niloticus) embryos Genet. Mol. Biol.
Marins,Luis Fernando; Iyengar,Arati; Maclean,Norman; Levy,Jose A.; Sohm,Frédéric.
In this study, we describe the use of a STAT5 responsive element (LHRE) reporter gene to monitor the activity of the growth hormone (GH) transduction pathway following expression of heterologous fish GH and rat STAT5b in tilapia embryos and fish fibroblast cells. Our results indicate that both GH and STAT5b are able to activate the LHRE at high levels when transferred separately, demonstrating the substantial level of conservation of the GH signal transduction pathways between fish and mammals. Unexpectedly, co-expression experiments show a strong inhibition of the GH-dependent activation, suggesting that simultaneous GH and STAT5b overexpression can counteract effects of GH expression in tilapia embryos.

Morphological development of Corydoras aff. paleatus (Siluriformes, Callichthyidae) and correlation with the emergence of motor and social behaviors Iheringia, Sér. Zool.
Rodríguez-Ithurralde,Daniel; del Puerto,Gabriela; Fernández-Bornia,Fernando.
Here we examine major anatomical characteristics of Corydoras aff. paleatus (Jenyns, 1842) post-hatching development, in parallel with its neurobehavioral evolution. Eleutheroembryonic phase, 4.3-8.8 days post-fertilization (dpf); 4.3-6.4 mm standard length (SL) encompasses from hatching to transition to exogenous feeding. Protopterygiolarval phase (8.9-10.9 dpf; 6.5-6.7 mm SL) goes from feeding transition to the commencement of unpaired fin differentiation, which marks the start of pterygiolarval phase (11-33 dpf; 6.8-10.7 mm SL) defined by appearance of lepidotrichia in the dorsal part of the median finfold. This phase ends with the full detachment and differentiation of unpaired fins, events signaling the commencement of the juvenile period (34-60 dpf;...

Maternally Inherited npm2 mRNA Is Crucial for Egg Developmental Competence in Zebrafish ArchiMer
Bouleau, Aurelien; Desvignes, Thomas; Traverso, Juan Martin; Nguyen, Thaovi; Chesnel, Franck; Fauvel, Christian; Bobe, Julien.
The molecular mechanisms underlying and determining egg developmental competence remain poorly understood in vertebrates. Nucleoplasmin (Npm2) is one of the few known maternal effect genes in mammals, but this maternal effect has never been demonstrated in nonmammalian species. A link between developmental competence and the abundance of npm2 maternal mRNA in the egg was previously established using a teleost fish model for egg quality. The importance of maternal npm2 mRNA for egg developmental competence remains unknown in any vertebrate species. In the present study, we aimed to characterize the contribution of npm2 maternal mRNA to early developmental success in zebrafish using a knockdown strategy. We report here the oocyte- specific expression of npm2...

Daily variations of the saccular endolymph and plasma compositions in the turbot Psetta maxima: relationship with the diurnal rhythm in otolith formation ArchiMer
Edeyer, Anaick; De Pontual, Helene; Payan, P; Troadec, Herve; Severe, Armelle; Mayer-gostan, N.
We studied the circadian variations of the ionic composition ([Na+], [K+], [Cl-]), total CO2 ([tot CO2]), and protein concentration ([protein]) in the plasma and endolymph in the distal and proximal sides of the sacculus in the turbot Psetta maxima. Daily variations in total plasma calcium ([tot Ga]) were also recorded. Significant diurnal fluctuations occurred in most parameters and the spatial heterogeneity of the endolymph composition was maintained throughout a 24 h period. The main parameters implicated in otolith calcification, endolymph [tot CO2] and endolymph [protein], fluctuated diurnally, [tot CO2] was highest and [protein] lowest at night; the relative concentrations were reversed during the day. The inverse, alternating relationship of maximal...

The nme gene family in zebrafish oogenesis and early development ArchiMer
Desvignes, T.; Fauvel, Christian; Bobe, J..
After the recent report of the expression of several nme genes in the zebrafish gonads, the present study aimed at further analyzing the expression of nme genes in the ovary with special attention for the nme transcripts that are maternally inherited and could thus participate in the determination of oocyte developmental competence. The expression levels of all groups I and II nme genes were characterized by QPCR in a panel of zebrafish tissues. The nme genes exhibiting an ovarian expression were subsequently monitored throughout oogenesis and early development, and their expression sites characterized using in situ hybridization. Here, we show that nme2b1, nme3, nme4, and nme6 are highly expressed in the ovary and present in the zebrafish oocyte...

Ion uptake pathways in European sea bass Dicentrarchus labrax ArchiMer
Blondeau-bidet, Eva; Hiroi, Junya; Lorin-nebel, Catherine.
Ion uptake mechanisms are diverse in fish species, certainly linked to duplication events that have led to the presence of a multitude of paralogous genes. In fish, Na+ uptake involves several ion transporters expressed in different ionocyte subtypes. In the European sea bass Dicentrarchus labrax, several key transporters potentially involved in Na+ uptake have been investigated in seawater (SW) and following a 2 weeks freshwater (FW) acclimation. Using gel electrophoresis, we have shown that the Na+/H+-exchanger 3 (nhe3, slc9a3) is expressed in gills and kidney at both salinities. Quantitative realtime PCR analysis showed a significantly higher nhe3 expression in fresh water (FW) compared to SW. Its apical localization in a subset of gill ionocytes in...

Description of the unusual digestive tract of Platax orbicularis and the potential impact of Tenacibaculum maritimum infection ArchiMer
Alix, Maud; Gasset, Eric; Bardon-albaret, Agnes; Noel, Jean; Pirot, Nelly; Perez, Valérie; Coves, Denis; Saulnier, Denis; Lignot, Jehan-hervé; Cucchi, Patricia N..
Background Ephippidae fish are characterized by a discoid shape with a very small visceral cavity. Among them Platax orbicularis has a high economic potential due to its flesh quality and flesh to carcass ratio. Nonetheless, the development of its aquaculture is limited by high mortality rates, especially due to Tenacibaculum maritimum infection, occurring one to three weeks after the transfer of fishes from bio-secure land-based aquaculture system to the lagoon cages for growth. Among the lines of defense against this microbial infection, the gastrointestinal tract (GIT) is less studied. The knowledge about the morphofunctional anatomy of this organ in P. orbicularis is still scarce. Therefore, the aims of this study are to characterize the GIT in...

Primers and polymerase chain reaction conditions for DNA barcoding teleost fish based on the mitochondrial cytochrome b and nuclear rhodopsin genes ArchiMer
Sevilla, Rafael G.; Diez, Amalia; Noren, Michael; Mouchel, Olivier; Jerome, Marc; Verrez-bagnis, Veronique; Van Pelt, Hilde; Favre Krey, Laurence; Krey, Grigorios; Bautista, José M..
This report describes a set of 21 polymerase chain reaction primers and amplification conditions developed to barcode practically any teleost fish species according to their mitochondrial cytochrome b and nuclear rhodopsin gene sequences. The method was successfully tested in more than 200 marine fish species comprising the main Actinopterygii family groups. When used in phylogenetic analyses, its combination of two genes with different evolutionary rates serves to identify fish at the species level. We provide a flow diagram indicating our validated polymerase chain reaction amplification conditions for barcoding and species identification applications as well as population structure or haplotyping analyses, adaptable to high-throughput analyses.

First links between self-feeding behaviour and personality traits in European seabass, Dicentrarchus labrax ArchiMer
Ferrari, Sebastien; Benhaim, David; Colchen, Tatiana; Chatain, Beatrice; Begout, Marie-laure.
Most studies carried out with seabass under self-feeding conditions report an intriguing social structure that is built around the device and the food dispenser with three coexisting triggering categories: high-triggering (HT), low-triggering (LT) and zero-triggering (ZT) fish. However, neither sex nor feeding motivation or hierarchy can explain the establishment of this specialization. We characterised the personality of seabass with the commonly used restraint and open field tests and assessed the link between personality traits and individual triggering activity towards the self-feeder apparatus. We found no differences between triggering categories during the restraint test but high triggering fish were characterised as shyer than low- and...

Ionic composition of endolymph in teleosts: Origin and importance of endolymph alkalinity ArchiMer
Payan, P; Kossmann, Hans; Watrin, A; Mayer-gostan, N; Boeuf, Gilles.
Ionic (Na+, K+, Cl-, PO43-, pH), total CO2, total calcium and protein concentrations in the plasma and endolymph of the inner ear were compared in trout Oncorhynchus mykiss and turbot Scophthalmus maximus. In both species, saccular endolymph was characterized by high levels of K+ and total CO2 and in trout by an alkaline pH, The kinetic characteristics of proton secretion across the saccular epithelium of trout were investigated using a titration technique in which isolated saccules were mounted as closed sacs. The rate of proton secretion depends strongly on the pH of the Ringer's solution and secretion stops at a pH below 7.2, Proton secretion is driven by an energy-dependent mechanism involving basolateral ouabain-sensitive Na+/K+ exchangers. Background: The surface of marine animals is covered by abundant and diversified microbial communities, which have major roles for the health of their host While such microbiomes have been deeply examined in marine invertebrates such as corals and sponges, the microbiomes living on marine vertebrates have received less attention. Specifically, the diversity of these microbiomes, their variability among species, and their drivers are still mostly unknown, especially among the fish species living on coral reefs that contribute to key ecosystem services while they are increasingly affected by human activities. Here, we investigated these knowledge gaps analyzing the skin microbiome of 138 fish individuals belonging to 44 coral reef fish species living in the...

The Nme gene family in fish ArchiMer
Desvignes, T.; Fostier, A.; Fauvel, Christian; Bobe, J..
The Nme gene family, also known as Nm23 or NDPK, is a very ancient gene family that can be found in all kingdoms of life. In the late eighties, a gene of the Nme family, NME1, was identified as the first metastatic suppressor gene, resulting in a major interest for this family. Due to the complexity of the family, the need for a unified and evolutionary-supported gene nomenclature was recently stressed by the scientific community. Based on a complete evolutionary history study of the gene family in metazoans and vertebrates, a unified nomenclature was recently proposed and accepted by gene nomenclature consortia. In addition to its well-documented role in tumor metastasis, members of the Nme family are also involved in a wide variety of cellular and...
