Zygomycotic Mediastinal Lymphadenitis in Beef Cattle with Ruminal Tympany OAK

A 9-month-old steer was autopsied due to recurrent ruminal tympany. A macroscopic examination found an enlarged caudal mediastinal lymph node, and a section of the lymph node revealed necrosis with marked calcification, similar to tuberculous lymphadenitis. Histopathologically, the lesion consisted of multiple coagulative necrotic foci and fibrosis with macrophage, lymphocyte, eosinophil and multinucleated giant cell infiltration. Non-uniform width hyphae were detected in the necrotic area and within the cytoplasm of the multinucleated giant cells, and they were found to be anti-Rhizopus arrhizus antibody positive in an immunohistochemical examination. Specific Molecular Detection and Characterization of Anaplasma marginale in Mongolian Cattle OAK

Anaplasma marginale is an etiologic agent of bovine anaplasmosis. This study aimed to molecularly detect and characterize A. marginale that is prevalent in Mongolian cattle populations. A highly specific and sensitive nested PCR (nPCR) method based on the Msp5 gene was developed to detect A. marginale (Msp5 nPCR). The method detected A. marginale from the positive DNA samples obtained from different countries, while no amplicons were observed from DNA samples of several other bovine blood pathogens tested. Anaplasma marginale has been detected in the Philippines only by peripheral blood smear examination and serological methods. This study generally aimed to molecularly detect and characterize A. marginale in cattle and ticks in Cebu, Philippines. A total of 12 bovine blood samples and 60 Rhipicephalus (Boophilus) microplus ticks were collected on the Cebu Island in 2011. 16S rRNA-based screening-PCR and DNA sequencing revealed 8 cattle (66.7%) and 8 ticks (13.3%) to be positive for A. marginale, and 1 tick (1.7%) to be positive for A. centrale. Babesia ovata: Taxonomy, phylogeny and epidemiology OAK

Babesia ovata, which is transmitted by Haemaphysalis longicornis, is an intraerythrocytic protozoan parasite of cattle. Based on its morphology, B. ovata is classified as a large-type Babesia. The developmental stages of B. ovata have been described both in cattle and the tick vector. In infected adult female ticks, the parasite is transovarially transmitted to the tick eggs. The sexual reproduction of B. ovata has been demonstrated in the tick midgut. 10 カ月齢のホルスタイン種雌育成牛が急性に斜頸,旋回,後弓反張,意識混濁の症状を呈した.大脳皮質壊死症を疑 い,ビタミンB1 製剤,コルチコステロイドおよび抗生物質による治療を実施したが反応せず,予後不良と判定された. 病理解剖により小脳に直径35. cm 大の腫瘤が確認され,左右側脳室および中脳水道の拡張がみられた.本症例は小脳腫 瘍による周辺部の圧迫および腫瘍の二次的圧迫による脳脊髄液流出障害により発生した内水頭症により,小脳徴候だけ でなく大脳および脳幹症状も出現したものと考えられた.病理組織学的検索の結果,本症例は上衣芽腫と診断された. Neospora caninum and Cryptosporidium parvum are intracellular protozoan parasites that are distributed worldwide and of major economical concern in cattle industry. N. caninum can cause abortion storms and high culling rates, whereas C. parvum has zoonotic implications and can cause diarrhea in calves. There are currently no data on the prevalence of neosporosis and cryptosporidiosis in humans or animals in southern Egypt. Epidemiology of bovine hemoprotozoa parasites in cattle and water buffalo in Vietnam OAK

A PCR-based survey of hemoprotozoa parasites detected Babesia bigemina, Theileria orientalis and Trypanosoma theileri among cattle and water buffalo in Vietnam, and a new Babesia sp. closely related to Babesia ovata was detected in cattle only. In addition, Theileria annulata and Trypanosoma evansi were not detected in both cattle and water buffalo. Phylogenetic analysis detected T. orientalis MPSP genotypes 3, 5, 7 and N3 in cattle and 5, 7, N1 and N2 in water buffalo. Additionally, water buffalo-derived T. theileri CATL sequences clustered together with a previously reported cattle-derived sequence from Vietnam. This is the first report of a new Babesia sp. in cattle, and T. orientalis MPSP genotype 7 and T. theileri in water buffalo in Vietnam.

Babesia ovata is a tick-transmitted hemoprotozoan parasite that infects cattle. In our study, bovine blood samples (n=2,034) were collected from 10 different countries (Brazil, China, Ghana, Japan, Mongolia, the Philippines, South Africa, Sri Lanka, Thailand and Vietnam) and DNA extracted. The DNA samples were screened using an established and specific polymerase chain reaction (PCR) assay targeting the Apical membrane antigen 1 (AMA-1) gene. Parasite DNA was detected among samples collected from Japan, Mongolia and Thailand. Loop-mediated isothermal amplification (LAMP) assays for detection of Theileria parva infections targeting the PIM and p150 genes OAK

We have developed two loop-mediated isothermal amplification (LAMP) assays for the detection of Theileria parva, the causative agent of East Coast fever (ECF), an economically important cattle disease in eastern, central and southern Africa. These assays target the polymorphic immunodominant molecule (PIM) and p150 LAMP genes. The primer set for each gene target consists of six primers, and each set recognises eight distinct regions on the target gene to give highly specific detection of T. parva. Theileria orientalis is a benign protozoan species that is widely distributed in Japan, yet sometimes causes serious economic losses in the livestock industry. In this study, we conducted a molecular survey based on genes encoding the major piroplasm surface protein (MPSP) and p23 for T. orientalis detected in cattle grazing in southern areas of Japan, consisting of 2 farms in Kumamoto prefecture (Aso and Kuma districts) and 3 farms in Okinawa prefecture (Ishigaki, Iriomote, and Yonaguni Islands). Conservation and improvement strategies should be based on the association between genetic and phenotypic characteristics. The objective of this work was to characterize five native Brazilian cattle breeds (Caracu, Crioulo Lageano, Curraleiro, National Polled and Pantaneiro) and two commercial breeds (Holstein and Nellore) using RAPD technique to estimate genetic distances and variability between and within breeds. Genetic relationships were investigated using 22 primers which generated 122 polymorphic bands. Abstract: The objective of this work was to compare two types of ovulation inducers - estradiol benzoate (EB group) or prostaglandin F2α (PG group) -, in postpartum dairy cows subjected to timed artificial insemination (TAI). Lactating Holstein-Gir crossbred cows (n=118) with 60 to 120 days of milk production were used in this study. All cows were treated with an intravaginal progesterone-releasing device (CIDR) plus 2 mg of estradiol benzoate (EB) on day 0. The CIDR inserts were removed on day 9, and all cows received 500 µg sodium cloprostenol. One objective of the NAHMS Dairy '96 Study was to assess dairy producer awareness of Johne's disease, estimate national and regional herd-level prevalence of Mycobacterium paratuberculosis infection, estimate economic losses due to Johne's disease on dairy operations, and describe use of recommended preventive measures on U.S. dairy operations. The beef industry in the United States consists of several distinct production levels ranging from the cow-calf producer at the lowest level to the final consumer. These sectors face varying levels of profitability, degrees of market power, conflicting goals, and price signals. Environmental regulations involve questions of what costs are involved, who is in a position to pay these costs, and whether market prices are capable of signaling different environmental practices. Understanding the relationships within the beef industry may allow researchers to fine-tune analyses of environmental issues in the beef industry.

The NAHMS Dairy '96 Study was designed to provide both participants and the industry with information on the nation's dairy animal population for education and research. The USDA's National Agricultural Statistics Service (NASS) collaborated with NAHMS to select a statistically valid sample yielding 2,542 producers. Included in the study were 20 states that represented 83.1 percent of the U.S. milk cows as of January 1, 1996. Data on digital dermatitis were collected during the second phase as Federal or state Veterinary Medical Officers (VMO's) or Animal Health Technicians (AHT's) contacted producers from February 20 through May 24, 1996 to complete a questionnaire. The National Dairy Heifer Evaluation Project ( NDHEP ) was conducted by the National Animal Health Monitoring System (NAHMS ) from April 1991 through July 1992. A total of 921 producers monitored calves on a daily basis from the first through the eighth week of life; results were extrapolated to the U.S. dairy population. Mortality of heifer calves was examined according to region ( west, midwest, northeast, southeast ). Mortality was highest in the western region ( 9.1 percent ) and lowest in the midwestern region ( 5.6 percent ).
