Genetic analysis of species in the Genus Catasetum (ORCHIDACEAE) using RAPD Markers BABT

In this work, RAPD molecular markers were used to access the genetic variability and to study the inter and intraespecifc relationship in a group of 37 species, including 56 individuals. A total of 15 RAPD primers were selected for DNA amplification. From a total of 221 bands analyzed, 209 (95%) were polymorphics. The level of interespecifc genetic similarity ranged from 37% between Catasetum complanatum and Catasetum laminatum to 83% between Catasetum triodon and Catasetum uncatum. The intraspecifc genetic similarity varied 88% for the individuals of Catasetum triodon to 93% between the individuals of Catasetum atratum and Catasetum macrocarpum. Use of RAPD molecular markers on differentiation of brazilian and chinese Ganoderma lucidum strains BABT

The aim of this work was to analyze the Brazilian and Chinese strains of Ganoderma lucidum with molecular RAPD markers. A similarity matrix was elaborated and the RAPD profiles of G. lucidum strains were also compared to two other Ganoderma spp: G. applanatum and G. lipsiense in order to produce genetic similarity among the species. Based on the primers used, it was possible to determine that the Brazilian strains and Chinese strain CC-22 are alike. The method and the primers selection showed to be appropriate for the genetic identification of G. lucidum strains, enabling them to be improved and used in research, as well as in the world market.

Random amplified polymorphisms between two South American subspecies of rattlesnakes (Crotalus durissus collilineatus and Crotalus durissus terrificus) BABT

The present work was made to determine the suitability of RAPD analysis for the identification of specimens of two subspecies of South American rattlesnakes (Crotalus durissus terrificus and Crotalus durissus collilineatus). The 11 arbitrary primer sequence tested amplified a total of 161 bands, of which 31% were polymorphics when compared with Crotalus specimens. The combining results from different primers allowed the identification of all the specimens under analysis. Several characteristic bands of each subspecies were identified. Dendrogram, based on RAPD markers, show three groups that corresponded to the subspecies of Crotalus, and to Bothrops jararaca specimen.

Randomly Amplified Polymorphic DNA of Trichoderma isolates and antagonism against Rhizoctonia solani BABT

Random Amplified Polymorphic DNA (RAPD) procedure was used to examine the genetic variability among fourteen isolates of Trichoderma and their ability to antagonize Rhizoctonia solani using a dual-culture assay for correlation among RAPD products and their hardness to R. solani. Seven oligodeoxynucleotide primers were selected for the RAPD assays which resulted in 197 bands for 14 isolates of Trichoderma. Efficiency of RAPD and ISSR markers in assessing genetic variation in Arthrocnemum macrostachyum (Chenopodiaceae) BABT

The goal of this study was to investigate the molecular characterization of A. macrostachyum genotypes grown in the Western coast of Syria using RAPD and ISSR techniques. PCR amplifications with 20 RAPD primers gave an average of 9.25 selected markers/ primer, with a maximum of 17 (OPG05) and a minimum of four (OPA02). Percentage of polymorphic bands ranged from 40 to 100% according to RAPD primers tested. Among the 185 selected bands, 160 (84.96%) were polymorphic. Genetic variability of pre and post-fragmentation cohorts of Aspidosperma polyneuron Muell. Arg. (Apocynaceae) BABT

RAPD was used to access the genetic variability in Aspisdosperma polyneuron, a long-lived, late-reproducing tropical tree, and highly important for the Atlantic Forest. RAPD profiles from adults (pre-fragmentation, >300 years old) and seedlings (post-fragmentation, <<50 years old) were analyzed. Results showed a decrease of genetic polymorphism of post-fragmentation cohorts in small fragments and higher genetic diversity within population. The genetic diversity distribution suggested the establishment of fragments as protected reserves, and the transference of seedlings among fragments for conservation of A. polyneuron.

Temporal genotypic diversity of Schizaphis graminum (Rondani 1852) (Hemiptera: Aphididae) in a black oats (Avena strigosa) field BABT

The aim of this work was to analyze the clonal diversity variation in Schizaphis graminum during a complete phenological cycle of black oats (Avena strigosa). RAPD markers were used for detection of DNA polymorphisms of each clonal lineage, derived from a single clone collected weekly during a period of four months, in a crop field of black oats, Londrina, Paraná, Brazil. The monthly genotypic diversity was estimated by Shannon Information Index (H). Characterization of Nomuraea rileyi strains using polymorphic DNA, virulence and enzyme activity BABT

The characterization of entomopathogenic microorganisms is important for the selection of more effective strains for use in integrated pest-control programs. Five Nomuraea rileyi strains (SA86101, GU87401, SR86151, CG128 and VA9101) were characterized using random amplified polymorphic DNA (RAPD) analysis, virulence studies and assessment of chitinolytic and proteolytic activity. Genetic, morphological and spatial characterization of two populations of Mabea fistulifera Mart. (Euphorbiaceae), in different successional stages BABT

Two populations of Mabea fistulifera (Euphorbiaceae) located at Rio Doce State Park (Minas Gerais, Brazil) in places characterized as in different successional stages, were investigated through genetic and ecological traits. Twenty randomly chosen individuals from each population had its genetic data assessed by 43 RAPD markers and were also evaluated through five morphologic traits and its spatial distribution. The genetic variability of 40 accessions of_C. arabica was evaluated using a combination of the random amplified polymorphic DNA (RAPD) technique and restriction digestion of genomic DNA. The genetic variability and the relatedness among all accessions were initially evaluated using 195 RAPD primers which revealed a very low level of genetic variation. To improve the efficiency in the detection of polymorphism, the genomic DNA of all accessions were submitted to digestion with restriction endonucleases prior to PCR amplification. Genetic relationships among species of the genus Bothrops based on RAPD markers BABT

The genetic relationships among 11 species of Bothrops found in Brazil were determined using random amplified fragment (RAPD) information. A total of 239 amplified bands were scored using 20 aleatory primers. Most of the bands (88.7%) were polymorphic. Phylogenetic analysis of the data determined three similarity groups within Bothrops: Group I- B. alternatus, B. neuwiedi, B. cotiara, and B. jararacusu; Group II- B. insularis, B. jararaca, and B. erythromelas; and Group III- B. moojeni, B. leucurus, and B. atrox. These groups coincided with those obtained by other methods, indicating that RAPD's could be a useful tool for the evaluation of genetic relationships at the interspecific level.

RAPD markers were used to investigate the distribution of genetic variability among a group of Guignardia citricarpa, G. mangiferae, and Phyllosticta spinarum isolates obtained from several hosts in Brazil, Argentina, Mexico, Costa Rica, Thailand, Japan, United States and South Africa. Direct RAPD evaluation of bacteria without conventional DNA extraction BABT

The present work reports successful DNA amplification of Pantoea agglomerans and Bacillus pumilus through Random Amplified Polymorphic DNA (RAPD). For this, template DNA was obtained without conventional DNA extraction. The procedure was as follows: cultures grown for 20 hours in 5 mL LB medium were centrifuged and the resulting preparation was suspended in TE buffer. After boiling, the cell suspension was diluted and 2.0 µl were used in reactions of 15 µl. The results showed no significant differences among the RAPD profile of the PCR reactions derived from the boiling and phenol extraction methods, suggesting the utilization of this method for genetic population analysis.

The best shoot regeneration was observed on MS basal supplemented with 1.0 mg/L Benzyl amino purine (BAP) in combination with Indole-3-acetic acid (IAA) (0.5 mg/L) that resulted in the increase by multiplication rate (7.65) with an average of 33.33 numbers of shoots and average shoot length of 2.70 cm. Genetic variability of three natural populations of Maytenus aquifolium (Celesteraceae) from Telêmaco Borba, Paraná, Brazil BABT

Three populations of Maytenus aquifolium from Monte Alegre farm, Telemaco Borba county, Paraná, Brazil were analyzed by RAPD markers. A total of 13 primers were applied wich yielded 283 well amplified markers in all the studied populations (Mortandade, Vila Preta and Trinita), producing different values of gene diversity and polymorphic loci. The analysis of molecular variance (AMOVA) indicated that 21.77% of the genetic variation was among the population. Molecular investigation of a fungemia outbreak due to Candida parapsilosis in an intensive care unit BJID

We investigated a nosocomial cluster of four Candida parapsilosis fungemia episodes that occurred in a neurological intensive care unit over a two-week period. The four infected patients had received parenteral nutrition through central lines, and all four had catheter-related candidemia. All of the isolates were susceptible to all of the antifungals tested, including amphotericin B, fluconazole, voriconazole, and caspofungin. Methicillin-resistant Staphylococcus aureus (MRSA) infections frequently complicate the post-operative course of transplant recipients, and despite nasal carriage and endemic colonization, MRSA outbreaks are not commonly described. DNA polymorphism and total protein in mutants of Metarhizium anisopliae var. Anisopliae (Metsch.) Sorokin strain E9 BJM

Five mutants (MaE10, MaE27, MaE24, MaE41 e MaE49) of Metarhizium anisopliae wild strain E9 were analysed for DNA profile through the RAPD technique and for changes in total protein content by spectrophotometry, polyacrylamide gel electrophoresis and densitometry. The pattern of RAPD markers showed genetic polymorphism among the strains: out of twenty primers seven were selected, producing 113 bands.
