Agrobacterium rhizogenes infection at the base of microcuttings in vitro can improve the rooting of some fruit species. A study was carried out comparing rooting of almond, apple, plum, Pyrus pyraster and two hybrid rootstocks, when infected with A. rhizogenes strain 1855, with and without the addition of hormones. Three responses occurred: genotypes rooted without auxins; genotypes rooted only with auxins; genotypes rooted only after infection. All genotypes rooted after bacterial infection. Background: Cultivation of algae for conversion to biofuels has gained global interest. Outdoor raceway cultivation is preferred because of its lower capital and operating costs. A major disadvantage of outdoor cultivation is susceptibility of algal crops to attack by predatory rotifers. In order to quantify the impact of rotifer attack on different species of algae, we evaluated the growth of eleven microalgal species over a 21-d period after being infected by the predatory rotifer Brachionus rubens. Analysis of genetic variability by ISSR markers in Calibrachoa caesia Electron. J. Biotechnol.

Background: Calibrachoa Cerv. (ex La Llave & Lexarza) is a genus of the Solanaceae family (La Llave and Lexarza, 1825). This genus has a high ornamental and economic value due to its intrinsic variability and multiplicity of flower colours. In Argentina there are eight native species, and one of them is Calibrachoa caesia. The genetic diversity among 35 accessions of C. caesia, from five departments in the province of Misiones, was analyzed using ISSR markers. Results: Thirteen ISSR primers yielded a reproducible banding pattern, with 701 amplified loci and 98% of polymorphism. Cloning, characterization and expression of Peking duck fatty acid synthase during adipocyte differentiation Electron. J. Biotechnol.

Background Fatty acid synthase (FAS) is a key enzyme of de novo lipogenesis (DNL), which has been cloned from several species: Gallus gallus, Mus musculus, Homo sapiens, but not from Anas platyrhynchos. The current study was conducted to obtain the full-length coding sequence of Peking duck FAS and investigate its expression during adipocyte differentiation. Results We have isolated a 7654 bp fragment from Peking duck adipocytes that corresponds to the FAS gene. The cloned fragment contains an open reading frame of 7545 bp, encodes a 2515 amino acid protein, and displays high nucleotide and amino acid homology to avian FAS orthologs. Production of lignocellulolytic enzymes by Aspergillus niger biofilms at variable water activities Electron. J. Biotechnol.

Lignocellulolytic enzyme production by Aspergillus niger was compared both in submerged fermentation (SF) and biofilm fermentation (BF) at varying water activities. Maximal filter paper activity, endoglucanase and xylanase activities were much higher in BF (2.96, 4.7 and 4.61 IU ml-1, respectively) than in SF cultures (1.71, 1.31 and 2.3 IU ml-1, respectively) but biomass yields were lower in BF than in SF (0.338 g g-1 and 0.431 g g-1, respectively). In the presence of 20% ethylene glycol (a w = 0.942) the enzyme activities decreased in both systems but BF still had higher levels (1.0, 1.0 and 2.6 IU ml-1, respectively) than SF cultures (0.6, 0.7 and 1.5 IU ml-1, respectively). For promoter studies the cloning, subcloning and transfer to different plasmid vectors usually requires use of restriction enzymes and ligation reactions. One obstacle is the nucleotide polymorphisms of eukaryotic genomic DNA, which has the consequence that a sequence often differs from published sequences. Therefore sequencing, rigorous restriction enzyme analysis or introduction of suitable sites has to be performed prior to cloning and subcloning. Minibioreactor-gas collector for determining bacteria-produced hydrogen sulfide Electron. J. Biotechnol.

A simple and economical minibioreactor-gas collector system for determination of hydrogen sulfide produced by a microorganism was designed. The detection of hydrogen sulfide was based on the reaction between the hydrogen sulfide in the gas stream from the culture, and a lead acetate solution 0.090 mol/L, contained in a tube gas collector; the conductimetric back titration of lead excess was made, and hydrogen sulfide was quantified indirectly, being the detection limit 0.5 µmol. The developed system was applied using Tsukamurella paurometabola DSM 20162 as a model, being the amount of hydrogen sulfide produced, 1.2 µmol in 24 hrs.

Gene expression and characterization of 2-keto-3-deoxy-gluconate kinase, a key enzyme in the modified Entner-Doudoroff pathway of Serratia marcescens KCTC 2172 Electron. J. Biotechnol.

We cloned 2-keto-3-deoxy-gluconate kinase (KDGK), which catalyzes the phosphorylation of 2-keto-3-deoxygluconate (KDG) to 2-keto-3-deoxy-6-phophogluconate (KDPG) from Serratia marcescens KCTC 2172. The nucleotide sequence revealed a single open reading frame containing 1,208 bp and encoding for 309 amino acids, with a molecular weight of 33,993 Da. The enzyme was purified via GST affinity chromatography. The putative KdgT binding site was detected upstream of the initial codon. The KDG kinase utilized 2-ketogluconate (KG) and KDG as substrates. The optimal temperature and pH for KDGK activity were 50ºC and 8.0, respectively.

Performance of biofilm carriers in anaerobic digestion of sisal leaf waste leachate Electron. J. Biotechnol.

Three methanogenic biofilm bioreactors were studied to evaluate the performance of three types of carriers. The carrier material were consisted of sisal fibre waste, pumice stone and porous glass beads, and the bioprocess evaluated was the methanogenesis anaerobic digestion of sisal leaf waste leachate. Process performance was investigated by increasing the organic loading rate (OLR) step-wise. The best results were obtained from the bioreactor packed with sisal fibre waste. It had the highest chemical oxygen demand (COD) removal efficiencies in the range of 80-93% at OLRs in the range of 2.4-25 g COD L-1d-1. High-density cultures of mammalian neurons offer a model system for studies of brain development, but the morphological features of individual neurons is difficult to ascertain. We show that a herpes virus vector expressing a bioluminescent protein allows detailed morphometric analyses of living neurons in complex culture environments. The use of dried spent yeast as a low-cost nitrogen supplement in ethanol fermentation from sweet sorghum juice under very high gravity conditions Electron. J. Biotechnol.

Dried spent yeast (DSY) was used as a low-cost nitrogen supplement for ethanol fermentation from sweet sorghum juice under very high gravity (VHG) conditions by Saccharomyces cerevisiae NP 01. The fermentation was carried out at 30ºC in a 5-litre bioreactor. The results showed that DSY promoted ethanol production efficiencies. The ethanol concentration (P), productivity (Qp) and yield (Yp/s) of the sterile juice (total sugar of 280 g l-1) supplemented with 8 g l-1 of DSY were not different from those supplemented with yeast extract and/or peptone at the same amount. Characterization of novel genic SSR markers in Linum usitatissimum (L.) and their transferability across eleven Linum species Electron. J. Biotechnol.

Little is known about the evolutionary relationships among Linum species, basically because of the lack of transferable molecular markers. Currently, expressed sequence tags available in public databases provide an opportunity for the rapid and inexpensive development of simple sequence repeat (SSR) markers in wild flax species. In this regard, fifty expressed sequence tag-derived microsatellite markers (EST-SSRs) were evaluated for polymorphism and transferability in 50 Linum usitatissimum cultivars/accessions and 11 Linum species. RNA isolation from hard and woody internodal bamboo (Bambusa balcooa) tissue is very difficult due to the presence of secondary metabolites, polysaccharides, and polyphenolics.
