Diagnosis of herpes-like virus infections in oysters using molecular techniques ArchiMer
Renault, Tristan; Lipart, Cecile.
Several herpes-like virus infections have been reported since 1991 among different bivalve species around the world (Nicolas et al., 1992; Comps and Cochennec, 1993; Renault et al., 1994; Hine and Thome, 1997; Hine 1997). To date, detection of these infections has been strongly correlated with severe losses of oyster larvae and spat. The virus infections are currently diagnosed by laborious histologicai and transmission electron microscopical examinations. These techniques, however, facilitate neither rapid diagnosis nor the ability to analyse large sample numbers.

Characterization of 27 microsatellite loci in the European flat oyster Ostrea edulis ArchiMer
Lallias, Delphine; Stockdale, Ruth; Boudry, Pierre; Beaumont, Andy R.; Lapegue, Sylvie.
The flat oyster Ostrea edulis is native to Europe and populations have been severely depleted by the parasite Bonamia ostreae since the 1980s. Additional genetic markers are required to improve population genetics study and linkage map development for selection for B. ostrea-resistance in this species. Here, we characterized 27 novel microsatellite loci for O. edulis. Number of alleles per locus ranged from 6 to 25 and observed heterozygosity between 0.375 and 1. Null alleles were suggested at a few loci but most loci were in Hardy-Weinberg agreement enabling their reliable use in further population and mapping genetics approaches.

Cellular and molecular responses of haemocytes from Ostrea edulis during in vitro infection by the parasite Bonamia ostreae ArchiMer
Morga, Benjamin; Renault, Tristan; Faury, Nicole; Chollet, Bruno; Arzul, Isabelle.
Bonamia ostreae is a protozoan, affiliated to the order Haplosporidia and to the phylum Cercozoa. This parasite is intracellular and infects haemocytes, cells notably involved in oyster defence mechanisms. Bonamiosis due to the parasite B. ostreae is a disease affecting the flat oyster, Ostrea edulis. The strategies used by protozoan parasites to circumvent host defence mechanisms remain largely unknown in marine bivalve molluscs. In the present work, in vitro experiments were carried out in order to study the interactions between haemocytes from O. edulis and purified parasite. B. ostreae. We monitored cellular and molecular responses of oyster haemocytes by light microscopy, flow cytometry and real-time PCR 1, 2, 4 and 8 h p.i. Light microscopy was used...

Bonamia ostreae, an intrahemocytic protozoan parasite infecting the oyster Ostrea edulis, is responsible for an endemic disease affecting all intensive breeding areas of European countries. Epidemiological studies showed variable prevalences, 43% in Galicia (Spain), from 6% to 32% in France and 45% in the most important area of Ireland. The impact of oyster transfers, handlings, high densities have been implicated in the disease development but environmental stressors could be more involved than zootechnical methods. The effect of water temperature, salinity and tidal exposure were particulary examined to assess stress induced changes and determine physiological, biochemical and immunological state indices of oyster populations, which will contribute to...

Can the protozoan parasite Bonamia ostreae infect larvae of flat oysters Ostrea edulis? ArchiMer
Arzul, Isabelle; Langlade, Aime; Chollet, Bruno; Robert, Maeva; Ferrand, Sylvie; Omnes, Emmanuelle; Lerond, Sophie; Couraleau, Yann; Joly, Jean-pierre; Francois, Cyrille; Garcia, Celine.
Bonamia ostreae is an intracellular protistan parasite affecting flat oysters Ostrea edulis. It can be detected in juveniles but mortalities mainly affect oysters which are more than 2 years old. The parasite is usually observed inside haemocytes and sometimes free, notably in gill epithelia suggesting a parasite release through this organ. However, the infective form and ways of entry and release remain undetermined. Flat oysters incubate their larvae in their pallial cavity for 8–10 days before releasing them into the water column. Flat oysters in Bay of Quiberon in South Brittany (France) are known to be infected with B. ostreae since 1979 and is the most important area in France for O. edulis spat collection. Flat oysters incubating larvae were sampled...

Création d'un banc d'Ostrea edulis ArchiMer
Marteil, Louis.

Using a satellite DNA family for species identification of commercial oysters ArchiMer
Lopez-flores, I.; De La Herran, R.; Garrido-ramos, M. A.; Boudry, Pierre; Ruiz-rejon, C.; Ruiz-rejon, M..
The cultivation of oyster presents some problems related with the identification and differentiation of species as Ostrea edulis (European flat oyster) with respect to Ostrea stentina (Provence oyster) or Crassostrea angulata (portuguese oyster). Crossostrea gigas (Pacific oyster). We have characterized a satellite DNA family and used it as a molecular marker for genetic differentiation between commercial and non-commercial oyster species present in Europe. This marker clearly supports a high degree of differentiation between O. edulis and O. stentina and conversely upholds the contention that C. gigas and C. angulata are the same species.

Monoclonal antibodies against Bonamia ostreae (Protozoa: Ascetospera), an intrahaemocytic parasite of flat oyster Ostrea edulis (Mollusca : Bivlavia) ArchiMer
Rogier, Hervé; Hervio, Dominique; Boulo, Viviane; Clavies, Christine; Hervaud, Eliane; Bachere, Evelyne; Mialhe, Eric; Grizel, Henri; Pau, Bernard; Paolucci, Francis.
The protozoan Bonamia ostreae (Ascetospora), a paasite of the flat oyster Ostrea edulis, was purified by differential and isopycnic centrifugations. Mice of the strain Balb/c were immunized with purified parasites and hybridomas were prepared by fusion of immunized mouse splenocytes with the mouse myeloma cell line P3-X63-Ag8-653. From the fusion, 12 clones were isolated and saved. The resulting antibodies were characterized with a solid phase radioimmunoassay (RIA). Antibodies from 7 clones reacted only with B. ostreae and not with normal host tissue. Although some of the antibiodies identified. One of these antibodies (20B2-1B12) has an apparent association constant of ca 3 X 10(8) M-1 and bound to a number of different sites on the parasite. These...

Xue, Quiggang.
The mechanism that bivalve molluscs react to infectious agents are important research subjects in the fIeld of disease control studies for these animals. While the selection of disease resistant populations have made decisive progression in some species, the mechanism behinds these resistant phenomena is not yet clear so far. However, hemocytes must represent one of the key factors in the reaction process. Based on this hypothesis, the structure and function of European flat oyster, Ostrea edulis, hemocytes have been investigated in aid of different techniques in the present study. ln addition, Pacific oyster, Crassostrea gigas, hemocytes have been analysed in certain aspects so as to provide some results for comparison. European flat oyster and Pacific...

The European flat oyster Ostrea edulis has been considered as the traditional French oyster for a very long time and has now nearly disappeared from our farms, as a consequence of the two successive epizootic of the Marteilia refringens parasite and Bonamia ostreae. The actual quasi monoculture of Crassostrea gigas has made necessary the search for a diversification in oyster culture, including the search for a new start of the indigenous flat oyster culture. The Ifremer has been conducting parallel works since 1985 in order to better understand the biology of the parasite as well as to try and select different lines of flat oyster resistant to Bonamia ostreae.

Flat oyster follows the apoptosis pathway to defend against the protozoan parasite Bonamia ostreae ArchiMer
Gervais, Ophelie; Chollet, Bruno; Renault, Tristan; Arzul, Isabelle.
The in vitro model Ostrea edulis hemocyte - Bonamia ostreae is interesting to investigate host-parasite interactions at the cellular level. Indeed, this unicellular parasite infects the flat oyster Ostrea edulis and multiplies within hemocytes, the central effectors of oyster defenses. Apoptosis is a mechanism used by many organisms to eliminate infected cells. In order to study the potential involvement of this mechanism in the oyster response to B. ostreae, in vitro experiments were carried out by exposing hemocytes from the naturally susceptible oyster O. edulis and a resistant oyster species Crassostrea gigas to live and heat-inactivated parasites. Hemocyte apoptotic response was measured using a combination of flow cytometry and microscopy analyses....
