High concentrations of xenobiotics from urban and industrial wastes have contributed to the contamination of many aquatic environments. We used the comet assay to evaluate the genotoxic potential of water collected from the River Paraná, which receives a great deal of waste, at three points (Puerto Piray, Eldorado and Montecarlo) in the Misiones Province of Argentina. The aim of this study was to monitor changes in genetic size of a small-closed population of Iranian Zandi sheep, by using pedigree information from animals born between 1991 and 2005. The genetic size was assessed by using measures based on the probability of identity-by-descend of genes (coancestry, f, and effective population size, Ne), as well as measures based on probability of gene origin (effective number of founders, f e, effective number of founder genomes, f g, and effective number of non-founder genomes, f ne). Prevalence of codon 72 P53 polymorphism in Brazilian women with cervix cancer Genet. Mol. Biol.

The p53 codon 72 polymorphism seems to be associated with HPV-carcinogenesis, although controversial data have been reported. A series of Brazilian women with cervix carcinomas were analyzed. Ninety-nine (67%) of 148 women were found to be homozygous (arg/arg) for the arginine polymorphism, and 49 (33%) were heterozygous (arg/pro). This polymorphism may be an important determinant of the risk for cervix cancer, but does not seem to be sufficient for carcinogenesis.

Spinal muscular atrophy (SMA) is an autosomal recessive inherited disorder caused by alterations in the survival motor neuron I (SMN1) gene. SMA patients are classified as type I-IV based on severity of symptoms and age of onset. About 95% of SMA cases are caused by the homozygous absence of SMN1 due to gene deletion or conversion into SMN2. PCR-based methods have been widely used in genetic testing for SMA. In this work, we introduce a new approach based on TaqMan® real-time PCR for research and diagnostic settings. DNA samples from 100 individuals with clinical signs and symptoms suggestive of SMA were analyzed. Mutant DNA samples as well as controls were confirmed by DNA sequencing. The use of self-incompatibility in the production of hybrid eucalyptus seed by `Aracruz Celulose' in Brazil Genet. Mol. Biol.

Self-incompatibility found in a Eucalyptus grandis clone was used to promote interspecific hybridization between E. grandis and E. urophylla clones. The interspecific hybridization achieved in an open-pollinated commercial seed orchard planted in Espírito Santo, Brazil was evaluated by the multi-locus outcrossing rate (tm) of the seed producing clone, E. grandis. The percentage of outcrossed seeds reached 95.9%. The outcrossing rate of individual trees was quite variable, but was always above 70.0%. In this work we describe the establishment of mesenchymal stem cells (MSCs) derived from embryonic stem cells (ESCs) and the role of bFGF in adipocyte differentiation. The totipotency of ESCs and MSCs was assessed by immunofluorescence staining and RT-PCR of totipotency factors. MSCs were successfully used to induce osteoblasts, chondrocytes and adipocytes. MSCs that differentiated into adipocytes were stimulated with and without bFGF. The OD/DNA (optical density/content of total DNA) and expression levels of the specific adipocyte genes PPARγ2 (peroxisome proliferator activated receptor γ2) and C/EBPs were higher in bFGF cells. Previous cytochrome B (CytB) mtDNA studies have suggested four species for the opossum genus Philander (four-eyed opossums), three (P. mcilhennyi, P. andersoni and P. opossum) from the Amazon and one (P. frenata) from the Brazilian Atlantic forest. During a faunal survey nine specimens of Philander sp. and four of Didelphis marsupialis were collected in the Mamirauá Sustainable Reserve, Amazonas State, Brazil. In this paper an extension of the biometric model of Mather and Jinks for the analysis of variation with digenic epistasis is presented. Epistatic effects can contribute favorably to the determination of the genotypic values of selected individuals or families and of superior hybrids. The genetic basis of cultivated soybean is rather narrow. This observation has been confirmed by analysis of agronomic traits among different genotypes, and more recently by the use of molecular markers. During the construction of an RFLP soybean map (Glycine soja x Glycine max) the two progenitors were analyzed with over 2,000 probes, of which 25% were polymorphic. Chicken embryos were incubated with BrdU, embedded in plastic resin, sectioned and screened immunohistochemically to identify proliferating cells in the neural tube and somites. Fixation in 4% paraformaldehyde for 1 h was essential for detecting specific colorimetric signals of BrdU incorporation into cells during the S phase of the cell cycle. Transverse sections of the neural tube showed that the nuclei of proliferating cells (BrdU positive) had a uniform and centralized distribution, whereas unstained nuclei were found only along the extremities of the neural tube. All Manihot species so far examined, including cassava (Manihot esculenta), have 2n = 36. Interspecific hybrids between cassava and its wild relatives show fair regular meiosis, and backcrossed generations exhibit high fertility. Electrophoresis shows affinity between species of different sections, as well as between some of them and cassava itself. Polyploidy has apparently contributed to the rapid speciation of this genus, while apomixis has offered a means of perpetuating new hybrid types adapted to different environments. It is assumed that cassava originated by hybridization between two wild Manihot species followed by vegetative reproduction of the hybrid.

A fragment of 466 base pairs from a highly variable peripheral region of the mitochondrial D-loop of horses was amplified and analyzed by single stranded conformational polymorphism (SSCP). Fourteen distinct SSCP variants were detected in 100 horses belonging to four breeds (Arabian, ARB; Thoroughbred, TB; Argentinian Creole, ARC; and Peruvian Paso from Argentina, PPA). Abstract Genes related to the response to drought stress in leaf and root tissue of drought-susceptible (DS) and tolerant (DT) genotypes were characterized by RNA-Seq. In total, 54,750 transcripts, representative of 28,590 genes, were identified; of these, 1,648 were of high-fidelity (merge of 12 libraries) and described for the first time in the Andean germplasm. From the 1,239 differentially expressed genes (DEGs), 458 were identified in DT, with a predominance of genes in categories of oxidative stress, response to stimulus and kinase activity. Intra-individual numerical chromosomal polymorphism in Trichomycterus davisi (Siluriformes, Trichomycteridae) from the Iguaçu River basin in Brazil Genet. Mol. Biol.

Cytogenetic analysis of Trichomycterus davisi, collected from the Iguaçu River basin, has shown a diploid number of 2n = 54 chromosomes. However, we observed intra-individual numerical polymorphism in a T. davisi specimen. There were three cell populations with diploid number 2n = 54 (40M + 12SM + 2ST), 2n = 55 (40M + 12SM + 2ST + 1M) and 2n = 56 (40M + 12SM + 2ST + 2A) chromosomes. This variation was attributed to a probable post-zygotic non-disjunction of a metacentric chromosome of small/middle size, followed by centric fission, which originated in this individual.
