Delta de l'Ebre is a natural bay model for Marteilia spp. (Paramyxea) dynamics and life-cycle studies ArchiMer

Marteilia spp. are paramyxean parasites that affect several bivalve species of economic interest, such as Ostrea edulis and Mytilus galloprovincialis. Certain aspects of Marteilia spp., such as their life cycle and host affinity and infection dynamics, still remain unknown. The 'Delta de l'Ebre' constitutes a natural model for the study of the life cycle of the parasite Marteilia, since uninfected mussels and flat oysters immersed in the bays can become infected. Of the 75 laboratories contacted through the survey, 54 answered, 14 of which are partially or totally devoted to mollusc diseases. In fact, only 4 laboratories are devoted full-time to this work. Of the 14 laboratories that declared to work on the diagnosis of mollusc diseases, 12 stated that they worked with mussels, 12 with oysters, 9 with clams and 8 with other mollusc species. This is coherent with available production information. According to production data in 2001 (FAO source), the main mollusc species in the region are the blue mussel, Mytilus edulis (297,485 tonnes), the Mediterranean mussel, Mytilus galloprovincialis (131,014 tonnes), the Pacific cupped oyster, Crassostrea gigas (127,323 tonnes, an important part of which is produced on the...

Over the last thirty years, the French production of flat oyster Ostrea edulis dropped from15,000 tons/year to 1,500 tons. Marteilia refringens, a parasite of the flat oyster's digestive gland, is partly responsible for this drop. The fight against this pathogen agent requires a better knowledge of its parasitic cycle. The main hypothesis is the existence of a heteroxenous cycle, that is to say a cycle involving more than one host. However, for thirty years this or these hosts have not been found, and the cycle of Marteilia refringens remains unknown.

Development of a TaqMan PCR assay for the detection of Bonamia species ArchiMer

The development of molecular diagnostic assays with increased sensitivity compared with conventional histological techniques is highly desirable for effective management of bonamiosis in cultured oyster stocks and wild populations. A real-time TaqMan PCR assay was developed for the specific detection of Bonamia species in infected oyster tissues. The TaqMan assay was shown to be significantly more sensitive than histopathology. Although a real-time TaqMan PCR assay is comparable with conventional PCR in terms of sensitivity, it offers the advantages that it is a rapid test and has a very low risk of sample cross-contamination. Some regulatory issues related to perkinsosis ArchiMer

Perkinsosis is one of the OIE listed diseases of molluscs caused by Perkinsus marinus and P. olseni. Consequently, molluscs originating from endemic areas should not be transferred into areas free of the disease. Impediments to the implementation of international zoosanitary regulation will be reviewed.

Le réseau REPAMO (Réseau de Pathologie des Mollusques) est un réseau de santé animale, qui s'occupe des coquillages marins du littoral français métropolitain. Infectious diseases are currently recognised as a major limiting factor to development and sustainability of aquaculture of molluscs. The study is focused on the European flat oyster, Ostrea edulis and its paramyxean parasite Marteilia refringens. Results are contributing to the prevention of risk associated to infected stocks imports from endemic zones, by developing molecular diagnostic tools that are specific, sensible and reliable. Phylogeny and taxonomy of Marteilia refringens, previously impeded by microscopy limits, were clarified by sequencing the rDNA SSU, small sub-unit in the rRNA gene cluster. Diseases in mollusc hatcheries and their paraedox in health management ArchiMer

Global molluscan aquaculture production is continuously increasing, dominated by five species, among which the Pacific cupped oyster, Crassostrea gigas, predominates. Hatchery production is accompanying this increase to assist consistent availability of juveniles for restocking, fishery enhancement, genetic improvements as well as for species diversification. Hatchery development contributes significantly to the demand for international transfers of live molluscs; a consequence of which pathogen transfer via transfer of live molluscs is currently recognised as a major cause of epizootic disease outbreaks. Molecular detection of the oyster parasite Mikrocytos mackini, and a preliminary phylogenetic analysis ArchiMer

The protistan parasite Mikrocytos mackini, the causative agent of Denman Island disease in the oyster Crassostrea gigas in British Columbia, Canada, is of wide concern because it can infect other oyster species and because its life cycle, mode of transmission, and origins are unknown. PCR and fluorescent in situ hybridization (FISH) assays were developed for M mackini, the PCR assay was validated against standard histopathological diagnosis, and a preliminary phylogenetic analysis of the M mackini small-subunit ribosomal RNA gene (SSU rDNA) was undertaken. A PCR designed specifically not to amplify host DNA generated a 544 bp SSU rDNA fragment from M mackini-infected oysters and enriched M. mackini cell isolates, but not from uninfected control oysters....

Bonamia sp. is a pathogenic parasite that occurs in the haemocytes of dredge oysters Ostrea chilensis Philippi in New Zealand, Ultrastructurally it resembles other haplosporidians in the possession of haplosporosomes, haplosporogenesis, persistence of mitotic microtubules during interphase and of the nuclear envelope during mitosis, and occurrence of a diplokaryotic or multi-nucleate plasmodial stage. Molecular characterisation of an Australian isolate of Bonamia exitiosa ArchiMer

An Australian (New South Wales) isolate of Bonamia was characterised at the molecular level by sequencing the 18S-ITS-1 region of the small subunit rRNA operon obtained from flat oysters Ostrea angasi shown to be infected by histological examination. Sequence data alignment with homologous genes from 2 other isolates of Bonamia (New Zealand and France) revealed high levels of nucleotide identity with both isolates. However, the Australian Bonamia is shown to be more closely related to the New Zealand isolate, suggesting the existence of an oceanic subgroup of Bonamia.

An ultrastructural study was carried out on Mikrocytos mackini, the cause of Denman Island disease in Pacific oysters Crassostrea gigas in western Canada. Three forms were identified, quiescent cells (QC), vesicular cells (VC) and endosomal cells (EC), QC occurred in the vesicular connective tissue (VCT), haemocytes (hyalinocytes), adductor and heart myocytes, and extracellularly, They had a central round to ovoid nucleus, <7 cisternae of inactive nuclear membrane-bound Golgi, few vesicles and lysosome-like bodies.
