Marine fish spermatozoa: racing ephemeral swimmers ArchiMer

After along period of spermatogenesis (several weeks to months), marine fish spermatozoa are delivered at male spawning in seawater (SW) at the same time as ova. In some fish species, as the ova micropyle closes quickly after release, these minute unicells, the spermatozoa, have to accomplish their task of reaching the micropyle within a very brief period (several seconds to minutes), for delivery of the haploid male genetic information to the ova. To achieve this goal, their high-performance motile equipment, the flagellum, must fully activate immediately on contact with the SW and then propel the sperm cell at an unusually high initial velocity. The cost of such 'hyperactivity' is a very rapid consumption of intracellular ATP that outstrips the supply....

Induced spawning of red drum, Sciaenops ocellatus: use of multivariate and univariate analysis methods in the search for side effects of LH-RHa treatments and ovarian development state upon spawn quality ArchiMer

The present work examines the usefulness of the combination of multivariate analyses and further one-way analysis of variance in the search for links between causes and effects in heterologous stimulation of spawning in fish. Stimulations with both 20 and 50 mu g . kg(-1) doses of [D Trp(6)] luteinizing hormone-releasing hormone analogue (LH-RHa) were effective in inducing and synchronizing spawning of red drum, Sciaenops ocellatus. Seasonal changes of sperm quality which can affect sperm biological parameters throughout the breeding period, have been little studied in mollusc species. Controlling gamete quality would aid the management of gametes in hatcheries and the development of selection programs. The aim of the present study was to describe the changes in sperm quality of wild Pacific oysters through the spawning season by comparing sperm parameters at the beginning (May), middle (July) and end (October) of this period using a panel of bio-descriptors. Characterization of sperm motility in sea bass: the effect of heavy metals and physicochemical variables on sperm motility ArchiMer

Computer assisted sperm analysis (CASA) was used to characterize the motility of sea bass Dicentrarchus labrax spermatozoa and to study the effect of several physicochemical variables and heavy metals on sperm swimming performance. Duration of sperm motility in sea bass was very short (< 50 s). During the first 20 s all the motility variables measured remained approximately constant, the velocity and linearity of the movement being maximum during this period, while both variables decreased sharply later. While slight variations in pH did not significantly modify sperm swimming performance, changes in osmolality affected all the measured motility variables. The greater amberjack, Seriola dumerili (Risso, 1810), is a promising candidate for the diversification of European aquaculture production, but inconsistent reproduction in captivity prevents commercial production. Recent studies showed that greater amberjack confined in sea cages exhibited scarce gonad development and early interruption of gametogenic activity during the reproductive season. Slow-release GnRHa treatment prevented atresia during vitellogenesis and induced ovulation of captive wreckfish (Polyprion americanus) ArchiMer

The potential of wreckfish (Polyprion americanus) to breed in captivity was examined. Ovarian activity varied seasonally, with vitellogenesis beginning in November and ending in May. Slow-release implant with gonadotropin-releasing hormone agonist (GnRHa) prevented atresia, and both GnRHa injections or implants at the end of vitellogenesis triggered ovulation after 7 days. Spontaneous spawning was obtained only in one female, and the few eggs produced were of poor viability. Manual stripping and artificial fertilization produced viable gametes and embryos. Therefore, present captivity conditions inhibited final oocyte maturation (FOM) and spawning, but not vitellogenesis and spermatogenesis. GnRHa treatments stimulate FOM and ovulation of viable eggs.

Gamete quality and management for in vitro fertilisation in meagre (Argyrosomus regius) ArchiMer

The aquaculture of meagre (Argyrosomus regius) requires methods for the control of reproduction that enable the production of families from specific individuals for selective breeding programs. We experimentally determined the parameters required for an in vitro fertilisation protocol. A total of 14 females and 5 males (mean ± S.D. weights of 20.45 ± 6.22 and 15.94 ± 2.75 kg, respectively) were used. Selected females had vitellogenic oocytes >550 μm in diameter and males had fluid sperm upon application of abdominal pressure. Effect of water temperature on individual reproductive activity of pollack (Pollachius pollachius) ArchiMer

The effects of water temperature on pollack (Pollachius pollachius) spawning features were individually assessed. During the spawning period, nine fish sets, each composed of one female and two males (mean body weight: 2.5+/-1.0 kg), were placed in small-volume tanks (2 m(3)). Fish sets were held at 8, 10, or 12 degreesC (n=3 for each temperature). Three females released eggs at 8 and 10 degreesC, while only two spawned at 12 degreesC. One hundred thirty-one individual spawns were collected from the eight spawning fish sets. The number of spawns collected per female was significantly lower at 12 degreesC (3.0+/-3.6), compared to 10 degreesC (17.3+/-10.1) and 8 degreesC (23.3+/-2.5). Changes in Atlantic cod (Gadus morhua L.) sperm quality during the spawning season ArchiMer

The biology of cod reproduction is well described in the scientific literature. However, sperm biology and spermatozoa management are poorly studied in this species. Because of its recent farming expansion, a better knowledge of cod gametes is becoming especially useful. This work aimed at establishing tools to study sperm biology in cod, and also investigated the existence of changes in cod sperm quality during the spawning period. We showed that sperm concentration could be assessed using spectrophotometry at 260 nm. Sperm motility significantly decreased after a 168-h storage at 4 degrees C. Cryopreservation of sea bass (Dicentrarchus labrax) spermatozoa in experimental and production simulating conditions ArchiMer

A sperm cryopreservation protocol adapted from turbot, was tested on sea bass using either 250-mu L straws or 1.5-mL cryovials. A dilution to 1/3 in Mounib's extender and a cooling rate of 65 degrees C.min(-1) allowed frozen sperm to recover an initial motility similar to that of fresh sperm at thawing; however, significant differences in motility (P < 0.001, n = 10 fish semen) were observed at further post-activation times, the motility decrease being faster in thawed sperm. At the experimental scale, triplicate inseminations of 2-mL aliquots (approximately 2 000 eggs) showed a significant fertility decay of thawed sperm compared to that of fresh sperm (P < 0.01, n = 12 fish semen) when a discriminating 35.10(3) spermatozoa to egg ratio was applied....

Motility characteristics of spermatozoa in cod (Gadus morhua) and hake (Merluccius merluccius) ArchiMer

As in many marine fish, motility is triggered by osmotic pressure (OP) in cod and hake spermatozoa and motility ranges several minutes with a high velocity (above 50 mu m/s) period restricted to 100 s. A decrease in the flagellar beat frequency (BF) is partly responsible of this briefness. Detailed observations of the flagella during the motility period showed: 1) sea water provokes OP damages (blebs) which impair the correct wave propagation 2) waves become restricted to the proximal flagellum and tip becomes devoid of wave 3) the wave amplitude decreases. The combination of these factors drastically limits to the earliest period of motility the ability for spermatozoa to efficiently progress towards egg for fertilization.

Marathon vs sprint racers: an adaptation of sperm characteristics to the reproductive strategy of Pacific oyster, turbot and seabass ArchiMer

This work assesses the present knowledge on Pacific oyster sperm biology in comparison to two marine fish species (turbot and seabass) whose sperm characteristics are well described. Sperm morphology mainly differs by the presence of an acrosome in Pacific oyster which is absent in both fish species. In turbot as in Pacific oyster, a sperm maturation process along the genital tract is observed. Sperm motility is triggered by changes in osmolality for seabass and turbot and in pH for Pacific oyster. However, complementary factors are involved to maintain sperm immotile in the genital tract. Marine fish culture of "new species" farmed in Europe ArchiMer

Optimizing the exploitation of wild and reared bluefin tunas under the constraint of sustainability: the Ifremer R&D project ArchiMer

A brief account is given of a research project commenced in 2000 by IFREMER concerning aspects of wild bluefin farming in offshore cages and reproduction control in land-based facilities. Three inter-related sub­ projects have been suggested: (i) wild fish ongrowing and/or fattening; (ii) reproduction; and (Hi) fingerling production and improvement of the understanding of early stage recruitment limitations. The Integration of this project in other EU community activities is discussed.
