The historical background of bivalve cell culture and its potential uses in molluscan pathology are reviewed. Then, primary cultures for the study of some host-pathogen interactions at the cellular level are considered. Next, alternative methods to classical cell cultures are examined that may lead to the production of bivalve celllines.

Infectious pathology in mollusc and shrimp hatcheries ArchiMer

Development of aquaculture has been linked to zootechnical mastering of larval production in hatcheries. Regular and excessive use of antibiotics (antifungal or antibacterial) may thus also be reduced, permitting to limit release of these products in sea-water and, consequently, avoiding progressive selection of resistant-strains.

Localization and characterization of acid phosphatase activity in Bonamia ostreae (Ascetospora), an intrahemocytic protozoan parasite of the fIat oyster Ostrea edulis (Bivalvia) ArchiMer

Acid phosphatase activity was cytochemically detected at the ultrastructural level in the protozoan Bonamia ostreae, an intrahemocytic parasite of the flat oyster Ostrea edulis. Lead nitrate was used as capture agent for phosphate liberated during the hydrolysis of the substrate (cytidine 5' monophosphate). Lead phosphate precipitation, indicative of enzyme activity, was located in membrane-bound organelles corresponding to the cytoplasmic inclusions known as 'dense bodies'. Acid phosphatase activity level and sodium L-tartrate inhibition were measured spectrophotometrically from a purified parasite suspension. The possible involvement of this enzyme in intracellular survival of B. ostreae is discussed.

Preparation and characterization of monoclonal antibodies against neoplastic hemocytes of Mytilus edulis (Bivalvia) ArchiMer

Mytilus edulis develop progressive and fatal hemocytic neoplasia. We have generated monoclonal antibodies using neoplastic hemocytes from highly infected individuals collected from Puget Sound in Washington State (USA). Twenty-seven monoclonal antibodies against neoplatic cell antigens were obtained. Specificity of 2 purified antibodies, 14F1 and 16G10, was demonstrated by immunofluorescence when incubated with neoplastic hemocytes, whereas these anbidodies did not react with normal cells. This specificity was confirmed by immunogold labelling and electron microscopic visualization. Monoclonal antibodies : a tool for molluscan pathology ArchiMer

Hybridoma technology is reviewed, and the characteristics of monoclonal antibodies are compared with those of polyclonal antibodies. The contribution of monoclonal antibodies to molluscan pathology is developed with special emphasis on their use as diagnostic tools. The results of studies with monoclonal antibodies prepared against the protozoan oyster pathogen Bonamia ostreae are briefly described.

The protozoan Bonamia ostreae (Ascetospora), a paasite of the flat oyster Ostrea edulis, was purified by differential and isopycnic centrifugations. Mice of the strain Balb/c were immunized with purified parasites and hybridomas were prepared by fusion of immunized mouse splenocytes with the mouse myeloma cell line P3-X63-Ag8-653. From the fusion, 12 clones were isolated and saved. The resulting antibodies were characterized with a solid phase radioimmunoassay (RIA). Antibodies from 7 clones reacted only with B. ostreae and not with normal host tissue. Although some of the antibiodies identified. Les productions d'invertébrés sont prépondérantes en aquaculture marine qui est économiquement très importate pour un grand nombre de pays, notamment de la zone Sud. Cette activité est confrontée à de nombreux problèmes des pathologie qui remettent en cause sa périnnité et son développment. Jusque dans les années quatre-vingt, les travaux relatifs à la pathologie des mollusques et des crevettes ont essentiellement porté sur la description des agents pathogènes. La création en 1985 par l'ISTPM/IFREMER d'un laboratoire pourvu de tous les équipements nécessaires aux techniques de pathologie a permis de mettre en place un programme de recherches orienté vers le contrôle des maladies.
