Qualification zoo-sanitaire du naissain de captage 2013 de l'huître creuse Crassostrea gigas (QUALIF) - Détection précoce des maladies en épreuve thermique de laboratoire ArchiMer

Depuis 2008, des mortalités massives d’huîtres creuses âgées de moins d’un an sont relevées sur le littoral français dès que la température de l’eau de mer atteint le seuil de 16°C. Ces mortalités de naissains sont associées à la détection du virus OsHV-1 μVar. Des travaux de qualification zoo-sanitaire menés depuis 2010 ont montré qu’un lot de naissains issus du captage naturel sur deux était infecté par le virus bien avant l’apparition des mortalités dans le milieu naturel, mais qu’a contrario les naissains d’écloserie expertisés présentaient un moindre risque de contamination. Review of the reproductive physiology of the scallop, Pecten maximus, applicable to intensive aquaculture ArchiMer

This paper presents a summary of information on reproduction biology in the scallop, Pecten maximus, which is required for the successful reseeding and intensive aquaculture programs developed in France since 1982. Data are presented on the following subjects: environmental factors that sustain gametogenic activity and the time of year this activity occurs; biochemical changes that are associated with gametogenesis; the consistency of egg quality; and development of a simple test to predict egg quality.

Plastics are persistent synthetic polymers that accumulate as waste in the marine environment. Microplastic (MP) particles are derived from the breakdown of larger debris or can enter the environment as microscopic fragments. Because filter-feeder organisms ingest MP while feeding, they are likely to be impacted by MP pollution. To assess the impact of polystyrene microspheres (micro-PS) on the physiology of the Pacific oyster, adult oysters were experimentally exposed to virgin micro-PS (2 and 6 µm in diameter; 0.023 mg·L−1) for 2 mo during a reproductive cycle. An amylase gene polymorphism is associated with growth differences in the Pacific cupped oyster Crassostrea gigas ArchiMer

This study investigated the non-neutrality of genetic polymorphism in two alpha-amylase genes (AMYA and AMYB) in the oyster Crassostrea gigas. Bi-parental oyster families, bred to be polymorphic for markers in these genes, were monitored for growth and survival for 1 year under standard culture conditions in two French production sites. Within-family genotype frequencies indicated that the two amylase genes were closely linked (c. 1.7 cM). Within two of three families, significant differences in growth were observed between genotypes at one of the two production sites, suggesting that this polymorphism is not neutral and might be under selection because of its role in digestive function. Survival, Growth and Reproduction of Cryopreserved Larvae from a Marine Invertebrate, the Pacific Oyster (Crassostrea gigas) ArchiMer

This study is the first demonstration of successful post-thawing development to reproduction stage of diploid cryopreserved larvae in an aquatic invertebrate. Survival, growth and reproductive performances were studied in juvenile and adult Pacific oysters grown from cryopreserved embryos. Cryopreservation was performed at three early stages: trochophore (13 +/- 2 hours post fertilization: hpf), early D-larvae (24 +/- 2 hpf) and late D-larvae (43 +/- 2 hpf). From the beginning (88 days) at the end of the ongrowing phase (195 days), no mortality was recorded and mean body weights did not differ between the thawed oysters and the control. Setting of a procedure for experimental incubation of Pacific oyster (Crassostrea gigas) embryos ArchiMer

Standardized experimental protocols designed to study Pacific oyster (Crassostrea gigas) gamete quality have not been previously published. Gamete sampling variations and confounding effect of interacting factors result in large variations between replicates, decreasing the effects of the studied factors. This work aims at defining a standardized procedure for incubation of oyster embryos designed for experimental purposes. In a first phase, four experiments were developed to improve embryo sampling and handling. They showed that a minimum of 50 embryos must be counted to decrease the variation between counting replicates. The oceanographic cruise Califa 2000, as the previous cruise, which concerned the "Parc National de Port-Cros", had for its principal objective to furnish some elements susceptible to help in the control of Caulerpa taxifolia expansion and in the protection of particularly sensitive areas (national parks, patrimonial sectors, regions of socio-economic importance). The cruise lasted from the 4th to the 21st of September 2000 in the rade d'Hyeres, Porquerolles island included, and the rade of Toulon.
