The Amplification Fragment Length Polymorphism (AFLP) technique was employed to study genetic variations which can be induced in vines by the stress occurring during different aspects of viticulture (in vitro cultivation, in vitro thermotherapy and virus infection). Analysis of AFLP banding patterns, generated by using 15 primer combinations, pointed to negligible genetic variation among plants exposed to individual stress. The average of similarity coefficients between differently stressed plants of the cultivars Müller Thurgau and Riesling were 0.984 and 0.991, respectively, as revealed by AFLP analysis. The low incidence of observed polymorphism demonstrates the high level of genome uniformity in plants reproduced by in vitro micropropagation via nodes,... |

Originating from the north eastern Asia, Crassostrea gigas has been introduced and translocated, mainly for aquaculture purpose, into several European countries (from Norway to Portugal and in the Mediterranean Sea) (1). Although highly variable, the invasiveness pattern of C. gigas has been demonstrated in several countries and therefore considered as a pest or a noxious species in those areas (2 and 3). Our project aims at identifying the characteristics of such a flourishing species: can its success be explained by chance and/or global warming only or does it exhibit a more important potential of adaptation than other species? Therefore we developed a population genomics approach, known as "genome scan". It corresponds to the study of numerous loci... |

This study presents the first genetic linkage map for the European flat oyster Ostrea edulis. AFLP and 20 microsatellite markers were genotyped in a three-generation pedigree comprised of 2 grand-parents, 2 parents and 92 progeny. Chi-square goodness of fit tests revealed high segregation distortion, which was significant for 32.8% of markers. Sixteen microsatellites and 235 AFLPs (170 AFLPs type 1:1 and 65 AFLPs type 3:1) were used to build sex-specific linkage maps using CriMap software. The first parental map (P1) consisted of 104 markers grouped in nine linkage groups, spanning 471.2 cM with an average spacing of 4.86 cM. The second parental map (P2) consisted of 117 markers grouped in ten linkage groups (which equals the haploid chromosome number),... |

We report the construction of the first genetic linkage map in the blue mussel, Mytilus edulis. AFLP markers were used in 86 full-sib progeny from a controlled pair mating, applying a double pseudo-test cross strategy. Thirty-six primer pairs generated 2354 peaks, of which 791 (33.6%) were polymorphic in the mapping family. Among those, 341 segregated through the female parent, 296 through the male parent (type 1:1) and 154 through both parents (type 3:1). Chi-square goodness-of-fit tests revealed that 71% and 73% of type 1:1 and 3:1 markers respectively segregated according to Mendelian inheritance. Sex-specific linkage maps were built with mapmaker 3.0 software. The female framework map consisted of 121 markers ordered into 14 linkage groups, spanning... |

An accurate amplified fragment length polymorphism (AFLP) method, including three primer sets for the selective amplification step, was developed to display the phylogenetic position of Photobacterium isolates collected from salmon products. This method was efficient for discriminating the three species Photobacterium phosphoreum, Photobacterium iliopiscarium and Photobacterium kishitanii, until now indistinctly gathered in the Photobacterium phosphoreum species group known to be strongly responsible for seafood spoilage. The AFLP fingerprints enabled the isolates to be separated into two main clusters that, according to the type strains, were assigned to the two species P. phosphoreum and P. iliopiscarium. P. kishitanii was not found in the collection.... |

The blue mussel, Mytilus edulis, is widely distributed in the northern hemisphere, from the White Sea to the Atlantic coast of southern France, and is an economically important species (1 million tons per year, FAO, 2002). At present however, little domestication has been achieved with bivalves, in contrast to agricultural crops, livestock and horticultural species like tomatoes, where selection and production is entirely controlled by man. For bivalves, despite their economic importance, no lines with selected characteristics have been taken to the production scale. There is therefore a need to make up for lost time. Our interest in genetic mapping sprang from some results showing a genetic component to some economically important traits like growth and... |

Genetic linkage maps consist of ordering molecular markers across the genome and require a high number of markers for a good coverage of the genome. Such maps represent a framework which enables the identification and localisation of Quantitative Trait Loci (QTLs) for traits of interest, such as growth or disease resistance, with the final aim of achieving genetic improvement through marker-assisted selection (MAS). Data on bivalves are scarce. No genetic map has yet been constructed in any mussel or flat oyster species. We report construction of a preliminary genetic linkage map in the blue mussel, Mytilus edulis (n=14). Amplified Fragment Length Polymorphism (AFLP) markers were used in a mapping family containing 86 full-sibs issued from a controlled... |

Late Quaternary (Eemian) deposits of the Netherlands contain shells that resemble those of living Mytilus galloprovincialis. Similar broad-shelled mytilids also occur in estuaries of the southwestern Netherlands together with slender individuals typical of M. edulis. We sampled living mussels along a depth gradient in the Oosterschelde to a) investigate whether a relation exists between shell shape and depth, b) test if the broadshelled specimens might represent M. galloprovincialis (or a hybrid with M. edulis) and c) assess by inference if the Quaternary specimens might be attributed to M. galloprovincialis as well. In order to do so, we compared genetic (length polymorphism of Me 15/16, COIII sequences and AFLPs) and shellmorphological characteristics... |

Abstract: The objective of this work was to compare wild poinsettia (Euphorbia heterophylla) biotypes as to their morphological characteristics, as well as to determine their genetic similarity in the state of Rio Grande do Sul, Brazil, and to confront the light compensation point of susceptible biotypes and of biotypes with low-resistance level to glyphosate. Besides the morphology analysis, the study included the genetic characterization, by AFLP, of susceptible (GR50= 58.65 g ha-1 a.e.) biotypes and of biotypes with low resistance (GR50= 310.36 g ha-1 a.e.) to glyphosate. The competitive ability of the biotypes was inferred according to their light compensation point. The observed morphological characteristics do not allow to differentiate biotypes as... |

Weeds in pastures can intoxicate animals, and Arrabidaea bilabiata is the most important species for herbivores in floodplain areas in the Amazon Basin. Genetic diversity studies in natural populations may contribute to the better understanding of the range of toxicity and the genetic variability organization in this species. The objective of this study was to assess the variability and genetic structure in six populations of A. bilabiata sampled in floodplain areas in three municipalities of the Amazonas State, from the AFLP markers analysis. AFLP markers were efficient to characterize the genetic variability of the 65 individuals analyzed. From four combinations of oligonucleotides, a total of 309 AFLP fragments was obtained, where 304 (98.38%) were... |
