Genetic changes in grapevine genomes after stress induced by in vitro cultivation, thermotherapy and virus infection, as revealed by AFLP Genet. Mol. Biol.

The Amplification Fragment Length Polymorphism (AFLP) technique was employed to study genetic variations which can be induced in vines by the stress occurring during different aspects of viticulture (in vitro cultivation, in vitro thermotherapy and virus infection). Analysis of AFLP banding patterns, generated by using 15 primer combinations, pointed to negligible genetic variation among plants exposed to individual stress. The average of similarity coefficients between differently stressed plants of the cultivars Müller Thurgau and Riesling were 0.984 and 0.991, respectively, as revealed by AFLP analysis. A first generation genetic linkage map of the European flat oyster Ostrea edulis (L.) based on AFLP and microsatellite markers ArchiMer

This study presents the first genetic linkage map for the European flat oyster Ostrea edulis. AFLP and 20 microsatellite markers were genotyped in a three-generation pedigree comprised of 2 grand-parents, 2 parents and 92 progeny. Chi-square goodness of fit tests revealed high segregation distortion, which was significant for 32.8% of markers. Sixteen microsatellites and 235 AFLPs (170 AFLPs type 1:1 and 65 AFLPs type 3:1) were used to build sex-specific linkage maps using CriMap software. AFLP-based genetic linkage maps of the blue mussel (Mytilus edulis) ArchiMer

We report the construction of the first genetic linkage map in the blue mussel, Mytilus edulis. AFLP markers were used in 86 full-sib progeny from a controlled pair mating, applying a double pseudo-test cross strategy. Thirty-six primer pairs generated 2354 peaks, of which 791 (33.6%) were polymorphic in the mapping family. Among those, 341 segregated through the female parent, 296 through the male parent (type 1:1) and 154 through both parents (type 3:1). Chi-square goodness-of-fit tests revealed that 71% and 73% of type 1:1 and 3:1 markers respectively segregated according to Mendelian inheritance. Genetic diversity analysis of isolates belonging to the Photobacterium phosphoreum species group collected from salmon products using AFLP fingerprinting ArchiMer

An accurate amplified fragment length polymorphism (AFLP) method, including three primer sets for the selective amplification step, was developed to display the phylogenetic position of Photobacterium isolates collected from salmon products. This method was efficient for discriminating the three species Photobacterium phosphoreum, Photobacterium iliopiscarium and Photobacterium kishitanii, until now indistinctly gathered in the Photobacterium phosphoreum species group known to be strongly responsible for seafood spoilage. At present however, little domestication has been achieved with bivalves, in contrast to agricultural crops, livestock and horticultural species like tomatoes, where selection and production is entirely controlled by man. O objetivo do trabalho foi analisar a estrutura e a diversidade molecular de quatro populações de Spodoptera frugiperda (J.E. Smith) associadas às culturas de milho e arroz no Rio Grande do Sul. Foram coletadas lagartas de quatro populações em áreas isoladas (distanciadas em mais de 300 km) nos municípios de Santa Rosa (milho) e Uruguaiana (arroz irrigado) e em áreas adjacentes, no município de Pelotas (milho e arroz irrigado). A análise de 40 lagartas (10 de cada população), com cinco combinações de oligonucleotídios iniciadores geraram o total de 241 locos, dentre os quais 229 (95%) foram polimórficos. Correlação da heterose de híbridos de milho com divergência genética entre linhagens PAB

O objetivo deste trabalho foi avaliar a produtividade de híbridos simples de milho obtidos de cruzamentos dialélicos entre linhagens divergentes, estimar a capacidade de combinação das linhagens e confirmar se a divergência genética entre as linhagens, obtida por marcadores moleculares, é correlacionada com a heterose dos híbridos simples no campo. Potato cultivar identification using molecular markers PAB

The objective of this work was to evaluate a set of microsatellite markers for varietal identification and characterization of the most widespread potato cultivars in Brazil. The DNA from 14 potato cultivars was genotyped using microsatellite markers and the alleles were scored in silver-stained polyacrylamide gel. Twenty-four microsatellite markers were evaluated, and only one locus was monomorphic. Based on band patterns, a set of two microsatellites that were able to identify and differentiate all examined cultivars was obtained.

Para contribuir al conocimiento de la diversidad genética del nopal se analizaron 85 genotipos representativos de Opuntia (silvestres y cultivados) por medio de la técnica de AFLPs y características del fruto. El análisis de AFLPs con los iniciadores Mse ICAC/Eco RI-AAG generaron más fragmentos polimórficos (57,5 en promedio) por genotipo; se obtuvo un dendrograma que no mostró una clara relación de acuerdo a la taxonomía conocida. AFLP polymorphism in restored provenances of Ceiba aesculifolia within an urban heat island Phyton

To quantify the effect on genetic diversity of restoring tree populations in an area under the influence of an urban heat island, we evaluated the genetic diversity of 72 Ceiba aesculifolia individuals, from a restoration experiment established from 2170 to 2260 m a.s.l. Reintroduced individuals were compared with the provenance from which the seeds were obtained, and two external provenances, for a total of 123 individuals. Estimación de la diversidad genética del Nopal, usando marcadores moleculares tipo AFLP Phyton

El presente estudio tiene por objetivo aplicar la técnica de los marcadores moleculares tipo AFLP para la estimación de la diversidad genética en nopal dentro del Banco de Germoplasma de la FAUANL, donde se han reportado 12 accesiones como posibles duplicados por medio de marcadores moleculares tipo RAPD. El método utilizado para la extracción de ADN fue el del ruptor celular. El ADN fue digerido y ligado, para posteriormente realizar una preamplificación y después la amplificación selectiva. Los fragmentos amplificados fueron luego separados y analizados. Se concluyó que ninguna de las accesiones estaba duplicada.
