Elaboration and evaluation of a new screening medium for detection and presumptive identification of extended-spectrum beta-lactamase-producing organisms (ESBL) BJM

The new ß-lactamases have arisen largely as a consequence of heavy use of new expanded spectrum ß-lactam antibiotics. Health professionals need to be aware of the resistance problems caused by the new enzymes, and know the correct procedures to detect, prevent and control such problems. In this study, a new screening medium called Ceftazidime-Inositol-Vancomycin-Amphotericin B Agar (CIVA) was elaborated and the microbiological performance was evaluated for the detection and presumptive identification of ESBL-producing members of Enterobacteriaceae. Influence of initial L-asparagine and glycerol concentrations on the batch growth kinetics of Mycobacterium bovis BCG BJM

The influences of the L-asparagine and glycerol initial concentrations in Sauton medium on the productivities of biomass and colony forming units were studied. The submerged batch cultivations of Mycobacterium bovis were carried out in a 20 L bioreactor. The L-asparagine and glycerol initial concentrations of 4.54 g/L and 25 mL/L, respectively, corresponded to the best biomass productivity, namely 2.5 g/L.day. On the other hand, the concentrations of 2.27 g/L and 25 mL/L, respectively, led to the highest productivity in terms of colony forming units, namely 2.7·10(6) colonies/mg.day. Sweet potato (Ipomoea batatas L.) plants were cultivated in vitro in Murashige and Skoog (MS) medium with 20 and 40 g L-1 of sucrose under two different photon flux densities (21 and 60 µmol m-2 s-1). Leaves developed in vitro mostly showed high variable to maximum fluorescence ratio (F V/F M) , indicating good development of photosynthetic apparatus. This ratio decreased during leaf aging, especially in the plants cultivated in vitro on medium with higher sucrose concentration and higher photon flux. Leaves developed ex vitro showed high F V/F M ratio during whole experiment. The effective photochemical efficiency (F V'/F M') was maximum at 15th day after emergence of leaves. Effect of coconut water on growth of olive embryos cultured in vitro Ciência Rural

The experiment was carried out to determine the appropriate dose of coconut water as supplement for in vitro cultivation of zygotic embryos from 19 olive genotypes. The isolated embryos of the olive seeds were immersed on culture medium containing 0 (control), 25, 50, and 100mL L-1 of fresh and sterile coconut water and kept for 45 days under controlled environment. The percentage of germination, shoot length, number of roots, number of leaves and number of internodes were measured for all 19 olive genotypes. European apple canker: morphophysiological variability and pathogenicity in isolates of Neonectria ditissima in southern Brazil Ciência Rural

ABSTRACT: European apple canker (EC) is caused by Neonectria ditissima, a pathogen officially registered as a quarantine pest in 2012. Thirty-five isolates of N. ditissima of different geographical regions of southern Brazil from apple branches showing symptoms of EC were identified by the specific pair primers Ch1 and Ch2 and analyzed concerning the virulence on Gala apple cultivar and morphophysiological characteristics. The disease symptoms were characterized and the isolates compared based on average mycelium growth (AMG), mycelium growth index (MGI), colony color, conidia type, dimensions and growth on potato dextrose agar (PDA), malt agar (AM), and synthetic SNAY (SN) culture media. Calli induction in leaf explants of coffee elite genotypes Ciência Rural

Three experiments were carried out with the objective of achieving high effectiveness in calli induction from high heterozygosis leaf explants of Coffea arabica through indirect somatic embryogenesis. A randomized-block design in a 2x5 factorial arrangement made up of two media [BOXTEL & BERTHOULY (1996) and TEIXEIRA et al. (2004)] and five C. arabica genotypes were used in the first experiment.
