Pressure and cold shock induction of meiotic gynogenesis and triploidy in the European sea bass, Dicentrarchus labrax L.: relative efficiency of methods and parental variability ArchiMer
Peruzzi, Stefano; Chatain, Beatrice.
The optimal conditions for the retention of the second polar body in sea bass eggs were investigated by altering the timing, intensity and duration of application of pressure and cold shocks. Treatment optima for cold shocks were 0-1 degrees C for 15-20 min at 5 min after fertilisation (a.f.) and 8500 psi for 2 min at 6 min a.f. for pressure shocks. Meiogenesis was obtained by fertilising eggs with UV-irradiated homologous sperm (32,000 erg mm(-2)) and pressure or cold shocking eggs as above. 100% triploidy was induced following definition of liable periods for the disruption of the meiotic spindle obtained in gynogenesis. Ploidy investigations were performed on experimental groups by flow-cytometry. Verification of uniparental transmission in meiogens was...

Effect of salinity and temperature on the expression of genes involved in branchial ion transport processes in European sea bass ArchiMer
Masroor, Waliullah; Farcy, Emilie; Blondeau-bidet, Eva; Venn, Alexander; Tambutté, Eric; Lorin-nebel, Catherine.
The responses of European sea bass to temperature increase and salinity decrease were investigated measuring mRNA expression levels of main genes involved in ion transport. Juvenile fish were pre-acclimated to seawater (SW) at 18 °C (temperate) or 24 °C (warm) for two weeks and then transferred for two weeks to either fresh water (FW) or SW at the respective temperature. Unlike temperate conditions, there is no change in Na+/K+-ATPase α1a (nka α1a) and Na+/H+ exchanger 3 (nhe3) mRNA expression following FW transfer in warm conditions. This is linked to the high expression of these genes in warm SW compared to temperate SW. Na+/Cl−-cotransporter (ncc2a) expression however is increased following FW transfer in temperate and warm conditions. Main transporters...

Characterising Essential Fish Habitat using spatio‐temporal analysis of fishery data: A case study of the European seabass spawning areas ArchiMer
Dambrine, Chloe; Woillez, Mathieu; Huret, Martin; De Pontual, Helene.
Fish habitats sustain essential functions for fish to complete their life cycle, such as feeding, growing and spawning. Conservation is crucial to maintain fish populations and their exploitation. Since 2013, the spawning stock biomass of the northern stock of European seabass (Dicentrarchus labrax) has been in a worrying state. A series of low recruitments with a persistently high level of fishing has been blamed, raising concerns about the processes involved in seabass reproduction and settlement in nurseries. Here, we characterise seabass spawning areas along the French Atlantic coast using vessel monitoring system (VMS) data. A non‐linear geostatistical approach was applied, from 2008 to 2014, to detect locations where seabass aggregate for spawning....

Genetics of European seabass (Dicentrarchus labrax L.) ArchiMer
Chatain, Beatrice; Chavanne, Herve.
The production of sea bass, a major species in Mediterranean fish farming, still relies in many cases on the production of fingerlings issued from wild breeders. Still, genetic improvement is a major key to sustainable development for all optimized forms of animal production. Three types of genetic improvement can be acheived in fish: i) the production of monosex populations. For seabass, monosex female populations would be the objective as females grow 20-30% faster than males. However, sex determination in this species is complex, impacted by both genetic and environmental factors. Hence, production of monosex populations is not yet a routine operation, although in some cases almost 100% female batches have been obtained; ii) chromosome set...

Genetic investigation of swimbladder inflation anomalies in the European sea bass, Dicentrarchus labrax L. ArchiMer
Peruzzi, Stefano; Westgaard, Jon-ivar; Chatain, Beatrice.
Although the actiology of swimbladder inflation anomalies in important aquaculture species such as European sea bass D. labrax is not fully determined, culture conditions are commonly suggested as main contributory factors. Little information is available on whether swimbladder inflation has a genetic basis for its expression too. In this work, 24 full-sibling sea bass families from a 4 dams x 6 sires factorial crossing were reared under communal conditions. The larvae developing normal and abnormal (uninflated or hyper-inflated) swimbladders were genotyped at four microsatellite loci, Labrax-3, Labrax-13, Labrax-17, Labrax29, and allocated to the individual breeders. Out of 273 offspring, 97% could be assigned to a single parental pair. The genotype and...

Genetic variation for carcass quality traits in cultured sea bass (Dicentrarchus labrax) ArchiMer
Saillant, Eric; Dupont-nivet, Mathilde; Sabourault, Marie; Haffray, Pierrick; Laureau, Stanislas; Vidal, Marie-odile; Chatain, Beatrice.
Genetic parameters for carcass quality traits were estimated in 27 families of sea bass (3 dams x 9 sires factorial mating design), raised mixed in the same tanks starting before hatching. O. spring parentage was determined a posteriori using 6 microsatellite loci. Carcass quality traits were recorded at 818 days post fertilization (mean standard length: 32.6 +/- 3.1 cm). Genetic parameters were estimated from the sire half sib variance and covariance components. Heritability of body weight (BW) and carcass processing traits (standardized to body weight) percent head weight (Head%), percent viscera weight (Viscera%) and percent visceral fat weight (VisceFat%) were relatively high ranging from 0.48 +/- 0.15 (Viscera%) to 0.87 +/- 0.23 (Head%); the estimate...

Heritabilities and GxE interactions for growth in the European sea bass (Dicentrarchus labrax L.) using a marker-based pedigree ArchiMer
Dupont Nivet, M; Vandeputte, Marc; Vergnet, Alain; Merdy, O; Haffray, P; Chavanne, H; Chatain, Beatrice.
253 full-sib families from 33 males and 23 females of European seabass were produced in a partly factorial mating design. All fish were reared in the same tank during 14 months, then 7000 of them were dispatched in four farms to different locations (France, Israel, Italy, Portugal) representing a wide variety of environmental conditions. Around 400 g mean weight, 1177 to 1667 fish in each site were weighed and length was measured. Condition factor (K) was calculated. Pedigrees were redrawn a posteriori using microsatellites markers: parental origin could be retraced for 99.2% of fish. Due to a high incidence of deformities, the useful sample size was reduced to 491-670 fish per site. Maternal effects were small. Using a simple animal model, heritability...

Nitrogenous and phosphorous waste production in a flow-through land-based farm of European seabass (Dicentrarchus labrax) ArchiMer
Lemarie, Gilles; Martin, Jean-louis; Dutto, Gilbert; Garidou, C.
A study on nitrogenous and phosphorous waste production in European seabass (Dicentrarchus labrax) was carried out in a flow-through land-based farm. The main objectives were (i) to calculate nitrogenous (N) and phosphorous (P) waste production (particulate and dissolved) from in situ measurements for different rearing ponds with specific fish biomasses, fish sizes and flow rates, (ii) to establish nitrogenous and phosphorous waste production budgets, ratios and equations, and (iii) to compare, for the whole farm, in situ measurements to estimate N and P waste production from waste equations. Waste production was manifest by an increase in concentrations of total ammonia nitrogen, total Kjeldhal nitrogen, particulate nitrogen, dissolved phosphorus and...

High diversity of skin-associated bacterial communities of marine fishes is promoted by their high variability among body parts, individuals and species ArchiMer
Chiarello, Marlene; Villeger, Sebastien; Bouvier, Corinne; Bettarel, Yvan; Bouvier, Thierry.
Animal-associated microbiotas form complex communities, which are suspected to play crucial functions for their host fitness. However, the biodiversity of these communities, including their differences between host species and individuals, has been scarcely studied, especially in case of skin-associated communities. In addition, the intraindividual variability (i.e. between body parts) has never been assessed to date. The objective of this study was to characterize skin bacterial communities of two teleostean fish species, namely the European seabass (Dicentrarchus labrax) and gilthead seabream (Sparus aurata), using a high-throughput DNA sequencing method. In order to focus on intrinsic factors of host-associated bacterial community variability,...

The European sea bass: a key marine fish model in the wild and in aquaculture ArchiMer
Vandeputte, Marc; Gagnaire, P.‐a.; Allal, Francois.
The European sea bass (Dicentrarchus labrax L.) is a marine fish of key economic and cultural importance in Europe. It is now more an aquaculture than a fisheries species (>96% of the production in 2016), although modern rearing techniques date back only from the late 1980s. It also has high interest for evolutionary studies, as it is composed of two semispecies (Atlantic and Mediterranean lineages) that have come into secondary contact following the last glaciation. Based on quantitative genetics studies of most traits of interest over the past 10–15 years, selective breeding programs are now applied to this species, which is at the beginning of its domestication process. The availability of a good quality reference genome has accelerated the...

Consistency in European seabass coping styles: A life history approach ArchiMer
Ferrari, Sebastien; Millot, Sandie; Leguay, Didier; Chatain, Beatrice; Begout, Marie-laure.
Recent years have seen a growth of interest in the consistent differences in individual behaviour over time and contexts constituting so-called “individual coping styles”. An understanding of this inter-individual variation is essential to improve our knowledge of the adaptive value of behaviour. Coping styles may have implications in diverse fields, so the development of appropriate screening methods for each species appears to be the most effective way to extend our knowledge and to incorporate behavioural responses into selection-based breeding programmes, to improve the domestication and welfare of farmed fish. We tested 30 juvenile seabass (Dicentrarchus labrax) at least twice in individual-based tests (feeding recovery in isolation, aggressiveness,...

Sea bass is a major species in Mediterranean aquaculture, but has a distribution area ranging from North Atlantic to South Mediterranean, with a population structure previously revealed by population genetics. To test the farming performances of wild sea bass populations, we produced a partial diallel cross mating scheme, using sires originating from North Atlantic (NAT), South Atlantic (SAT), West Mediterranean (WEM), North-East Mediterranean (NEM) and South-East Mediterranean (SEM). Fifteen sires per origin were mated in a full-factorial design using artificial fertilization with 9 NAT dams and 17 WEM dams, producing 10 population crosses and 1950 potential full-sib families. All fish were reared together, then tagged at an average weight of 20 g and...

Effects de le temperature et du regime alimentaire sur certains parametres de l'immunite cellulaire du loup de mer en elevage ArchiMer
Deschaux, P; Peres, G.
The data showed that antibody response and NK cells activity of a teleostean fish Dicentrarchus labrax (Loup de mer) were maximal when animals were bred at a temperature equal to 18 degree C. A proteinoprive diet enhanced T cytotoxic response and diminished NK cell activity. A fast of 15 days had no effect on these two cellular responses.

Experiences de reproduction artificielle de Loup Dicentrarchus labrax (Linne 1758). ArchiMer
Barnabe, G.; Tournamille, J.-c..
Mature eggs were expressed from captured female and fertilised artificially with slightly bloody sperm, in dry conditions or in a humid atmosphere, both at 13 degree C. The best incubation conditions proved to be 13 degree C, with streptomycin and penicillin. Another sample was kept at 18 degree C. Hatching occurred after 4 or 5 days, larvae showing positive phototropism. The system of renewal of water and artificial food is described. The embryonic reserve drop of lipid disappeared between day 14 and 16. Observations on larvae were like previous ones, but live larvae had larger mouths than had fixed specimens, and it was found that temperatures of 13 degree C or 18 degree C were suitable beyond the egg stage. A sudden change in temperature caused by a...

Early individual electronic identification of sea bass using RFID microtags: A first example of early phenotyping of sex-related growth ArchiMer
Ferrari, Sebastien; Chatain, Beatrice; Cousin, Xavier; Leguay, Didier; Vergnet, Alain; Vidal, Marie-odile; Vandeputte, Marc; Begout, Marie-laure.
Although individual electronic tagging using PIT (Passive Integrated Transponder) tags is well established, it is mainly used for fish > 60 mm in length. Since electronic tagging is an ideal identification method, we used RFID (Radio Frequency Identification) microtags (6 mm in length, 1 mm in diameter, 10 mg in mass) to characterize individual fish from the early stages of their development and throughout their lifecycles. We used sea bass, Dicentrarchus labrax, (105 day-old fish weighing between 100 and 1100 mg) and studied the effects of intra-coelomic tagging in half the population using different endpoints including survival and tag reading, growth over 6 months and swimming responses. Are sex ratios in wild European sea bass (Dicentrarchus labrax) populations biased? ArchiMer
Vandeputte, Marc; Quillet, Edwige; Chatain, Beatrice.
Sex ratios in farmed European sea bass are highly biased towards males (75 to 95%), which is problematic for aquaculture. In this mini-review, we re-analyse fisheries literature data about sex ratios in wild sea bass from 13 population samples, representing altogether 4889 individuals covering the major part of the distribution range of the species. We find that as a whole, the sex ratio of wild populations is biased towards females (59.4% females, p < 0.001), but that the sex ratio of the younger fish (< 30 cm total length) is balanced (52.0% females, p = 0.15), while the sex ratio of the older fish is heavily biased towards females (69.5% females, p < 0.01). Possible causes of these differences (differential longevity, biased sampling) are...

Modulation des performances cardio-respiratoires du bar europeen (Dicentrarchus labrax) par l'apport d'acides gras alimentaires: une etude integree des effets cardiaques in vivo au courant calcique de type L des myocytes ventriculaires ArchiMer
Chatelier, Aurelien.
Diet quality, and particularly fatty acids composition, is an environmental factor that is emerging as a significant source of physiological diversity. Indeed, numerous recent studies show that the fatty acid composition of the diet influences cardiac and metabolic performances of organisms. Because it is involved in the control and regulation of internal flow of matter and energy, the heart is believed to play a central role in environmental adaptation in fish. Within the double context of adaptation of individuals to their ecological niche and species evolution, understanding sources of interindividual variability and particularly fatty acid influences on the cardiorespiratory system is essential. The objective of the study was to investigate in the sea...

Utilisation de bactéries lactiques probiotiques pour prémunir les poissons d'élevage contre des vibrions pathogènes ArchiMer
Lamari, Faouzi.
Diseases cause major production losses in fish farms. Bacteria of the genus Vibrio are known as opportunistic pathogens, and have been associated with mass-mortality episodes. To cope with these disease outbreaks, fish farmers often resort to the use of antibiotics. However, preventive overuse has led to the emergence of resistant strains and frequent treatment failures. Probiotic lactic acid bacteria are increasingly used to improve the quality of seed production in aquaculture, while limiting antibiotic treatments. Microbial strains must fulfil several criteria to be evaluated as potential probiotics. In a first step, we selected and characterized 55 strains of lactic acid bacteria isolated from fish hatchery. The first selection criterion focused on the...

Husbandry stress during early life stages affects the stress response and health status of juvenile sea bass, Dicentrarchus labrax ArchiMer
Varsamos, S; Flik, G; Pepin, Jean-francois; Bonga, S; Breuil, Gilles.
In aquaculture management it is important to establish objective criteria to assess health and welfare of the fish. Here we show that European sea bass (Dicentrarchus labrax) confronted with husbandry-associated stress (tank cleaning, i.e. scrubbing, and water temperature variation) during early life stages show poorer survival and disease resistance as juveniles. We evaluated several parameters for stress (plasma cortisol, glucose and lactate, hydromineral status), growth performance, the immune response (plasma IgM levels) and the effects of a nodavirus challenge. Principal component analysis allowed the establishment of a stress panel including plasma cortisol, osmolality, IgM levels and weight. Sea bass juveniles reared during early life in high and...
