Genetic diversity of Xanthomonas axonopodis pv. citri based on plasmid profile and pulsed field gel electrophoresis Genet. Mol. Biol.

Xanthomonas axonopodis pv. citri strains that cause disease in citrus were investigated by pulsed field and plasmid profile analysis. For the first method, genomic DNA was digested by the rare-cutting enzymes Xba I and Vsp I. The strains evaluated were collected in seven different States of Brazil and in Argentina, Bolivia, Paraguay and Uruguay. Genetic variability was found among strains of X. axonopodis pv. citri from different geographical areas Argentina, Bolivia and Uruguay, with similarities varying from 0.62 to 0.83. The tropical mosquito, Aedes aegypti is the most important domestic vector of urban yellow fever and dengue. Genetic population studies on this vector are important because they may lead to new tools for surveillance. An analysis of genetic structure was conducted among populations of A. aegypti from 11 localities in four demographic regions within six Brazilian federal states. Markers included 21 random amplified polymorphic DNA (RAPD) loci. RAPD markers were detected among populations and cluster analysis revealed two main groups. Genetic variability of Herpailurus yagouaroundi, Puma concolor and Panthera onca (Mammalia, Felidae) studied using Felis catus microsatellites Genet. Mol. Biol.

We used four microsatellite loci (Fca08, Fca45, Fca77 and Fca96) from the domestic cat, Felis catus, to investigate genetic variability in specimens of Herpailurus yagouaroundi (jaguarundi, otter cat, eyra), Puma concolor (cougar, mountain lion, puma) and Panthera onca (jaguar) held in various Brazilian zoos. Samples of DNA from the cats were PCR amplified and then sequenced before being analyzed using the CERVUS program. Our results show a mean polymorphic information content (PIC) of 0.83 for H. yagouaroundi, 0.66 for P. concolor and 0.69 for P. onca and a mean of 10.3 alleles for the Fca08 locus, 5.3 for Fca 45, 9 for Fca 77 and 14 for Fca 96. These results indicate a relatively high level of genetic diversity for the specimens studied.

Six species of Trifolium (T. polymorphum Poir., T. riograndense Burkart, T. argentinense Speg., T. medium L., T. pratense L. and T. repens L.) were analyzed using inter-simple sequence repeats (ISSR) markers. Six selected primers generated 186 polymerase chain reaction (PCR) products exploring 112 loci in 34 genotypes analyzed with molecular sizes ranging from 200 to 1300 bp. These primers were able to discriminate among and within species, with the PCR products being on average 41.6% species-specific and 59.9% polymorphic at the within species level. The genetic diversity of Jamunapari goats (Capra hircus) was investigated using an optimized non-radioactive polymerase chain reaction single-strand conformation polymorphism (PCR-SSCP) method to detect <FONT FACE=Symbol>a</font>-lactalbumin polymorphism in a sample of 50 goats. Our data show that PCR-SSCP is an appropriate tool for evaluating genetic variability in Jamunapari goats. Analysis of cultured catfish from six farms in Tamaulipas, Mexico was achieved using a combination of microsatellite PCR analysis and semiautomatic fluoresce-based detection, in order to provide a first assessment of the genetic variability on cultured catfish in Mexico. Five microsatellites showed extensive polymorphism with allele numbers ranging from 10 and 20. Overall observed heterozygosity at each locus ranged between 0.76 and 0.91 and the average polymorphic information content (PIC) for the five loci was 0.811, indicating that these loci can be used for studies of paternity identification, linkage and population genetics. The genetic relationships among 81 maize accessions consisting 79 landraces and two improved varieties, maintained by farmers in southern Brazil were investigated using Random Amplified Polymorphic DNA (RAPD). Thirty-two highly informative primers amplified 255 markers of which 184 (72.2%) were polymorphics. Based on the RAPD markers, a dendrogram was constructed using the UPGMA method. The range of genetic similarity was from 0.78 to 0.91. The molecular data grouped the accessions into two main clusters, which were correlated according to kernel colors. Population structure analysis of Pimelodus maculatus (Pisces, Siluriformes) from the Tietê and Paranapanema Rivers (Brazil) Genet. Mol. Biol.

Pimelodus maculatus populations from the Tietê and Paranapanema rivers were sampled and had their genetic structure analyzed by using RAPD markers, with the aim of contributing to future conservation studies. The proportion of polymorphic loci was greater than 50% in the populations of both rivers. Genetic diversity data showed that, in spite of its nine hydroelectric plants, the Tietê river population was genetically homogeneous, whereas the Paranapanema river population was structured. This might be due to the presence of high waterfalls distributed all along its course. These data may serve as indicators for future conservation studies on the Tietê and Paranapanema rivers.

In the present work, isoenzyme electrophoresis was used to analyze the variability and phenetic relationships among seven American species of genus Prosopis belonging to three different sections: P. argentina (Monilicarpa), P. glandulosa, P. velutina, P. flexuosa, P. ruscifolia, P. kuntzei (Algarobia), and P. reptans (Strombocarpa). Heat tolerance is measured at tissue level by cellular membrane thermostability (CMT) and at the whole plant level by the heat tolerance index (HTI). Eight upland cotton cultivars and 15 crosses were used to determine the type and extent of genetic variability associated with the expression of these traits between and within environments. Heat stress and non-stress conditions were used as the CMT environments and years for HTI. In this study, the microsatellite technique was used to evaluate the genetic variability in populations of collared and white-lipped peccaries kept in captivity. Six primers developed for domestic pigs were used and amplified in both species. They revealed the presence of five polymorphic loci and one monomorphic locus. The polymorphic loci included 4 of the 16 alleles in collared peccaries, and 3 of the 10 alleles in the white-lipped peccaries. Gene variation and population genetic structure of the Neotropical sawfly Digelasinus diversipes were measured by allozyme analyses using starch gel electrophoresis. Cocoon masses were collected in Eugenia glazioviana (Myrtaceae) stems, in two areas of the "Estação Ecológica Jataí" (Luiz Antônio, SP, Brazil - 21°25' S, 47°50' W), in 2000 and 2001. Coconut palms of the Tall group were introduced to Brazil from the Cape Verde Islands in 1553. The present study sought to evaluate the genetic diversity among and within Brazilian Tall coconut populations. Samples were collected of 195 trees from 10 populations. Genetic diversity was accessed by investigating 13 simple sequence repeats (SSR) loci. This provided a total of 68 alleles, ranging from 2 to 13 alleles per locus, with an average of 5.23. The mean values of gene diversity (He) and observed heterozygosity (Ho) were 0.459 and 0.443, respectively.
