Quantitative trait loci mapping of pubescence density and flowering time of insect-resistant soybean (Glycine max L. Merr.) Genet. Mol. Biol.
Komatsu,Kunihiko; Okuda,Shiori; Takahashi,Masakazu; Matsunaga,Ryoichi; Nakazawa,Yoshinori.
Analysis of antibiosis resistance to common cutworm (Spodoptera litura Fabricius) in soybean (Glycine max (L.) Merr.) has progressed significantly, but the immediate cause remains unknown. We performed quantitative trait loci (QTL) analysis of pubescence density and plant development stage because these factors are assumed to be the immediate cause of resistance to cutworm. The QTLs for pubescence appeared to be identical to the previously detected the Pd1 and Ps loci controlling pubescence density. We found no candidate loci for flowering time QTLs, although one could be identical to the gene governing the long-juvenile trait or to the E6 loci controlling maturity. None of the QTLs overlapped with the QTLs for antibiosis resistance.

Metabolomics analysis of wild type Arabidopsis thaliana plants, under control and drought stress conditions revealed several metabolic pathways that are induced under water deficit. The metabolic response to drought stress is also associated with ABA dependent and independent pathways, allowing a better understanding of the molecular mechanisms in this model plant. Through combining an in silico approach and gene expression analysis by quantitative real-time PCR, the present work aims at identifying genes of soybean metabolic pathways potentially associated with water deficit. Digital expression patterns of Arabidopsis genes, which were selected based on the basis of literature reports, were evaluated under drought stress condition by Genevestigator. Genes...

Method optimization for proteomic analysis of soybean leaf: improvements in identification of new and low-abundance proteins Genet. Mol. Biol.
Mesquita,Rosilene Oliveira; Soares,Eduardo de Almeida; Barros,Everaldo Gonçalves de; Loureiro,Marcelo Ehlers.
The most critical step in any proteomic study is protein extraction and sample preparation. Better solubilization increases the separation and resolution of gels, allowing identification of a higher number of proteins and more accurate quantitation of differences in gene expression. Despite the existence of published results for the optimization of proteomic analyses of soybean seeds, no comparable data are available for proteomic studies of soybean leaf tissue. In this work we have tested the effects of modification of a TCA-acetone method on the resolution of 2-DE gels of leaves and roots of soybean. Better focusing was obtained when both mercaptoethanol and dithiothreitol were used in the extraction buffer simultaneously. Increasing the number of washes...

Genetic parameters and selection strategies for soybean genotypes resistant to the stink bug-complex Genet. Mol. Biol.
Godoi,Cláudio Roberto Cardoso de; Pinheiro,José Baldin.
Soybean genotypes resistant to stink bugs are derived from complex breeding processes obtained through indirect selection. The aim of the present work was to estimate genetic parameters for guiding selection strategies towards resistant genotypes, based on those traits associated with responses to pod-attacking stink bugs, such as the grain filling period (GFP), leaf retention (LR), percentage index of pod damage (PIPD) and percentage of spotted seeds (PSS). We assessed the parental lines IAC-100 (resistant) and FT-Estrela (susceptible), the progenies F2 and F4, 30 progenies F2:3, 30 progenies BC1F2:3 and 30 progenies BC2F2:3, besides the cultivars BRS Celeste and MGBR-46 (Conquista). Three field experiments, using randomized complete block design with...

Osorio,Marina Borges; Bücker-Neto,Lauro; Castilhos,Graciela; Turchetto-Zolet,Andreia Carina; Wiebke-Strohm,Beatriz; Bodanese-Zanettini,Maria Helena; Margis-Pinheiro,Márcia.
Environmental stresses caused by either abiotic or biotic factors greatly affect agriculture. As for soybean [Glycine max (L.) Merril], one of the most important crop species in the world, the situation is not different. In order to deal with these stresses, plants have evolved a variety of sophisticated molecular mechanisms, to which the transcriptional regulation of target-genes by transcription factors is crucial. Even though the involvement of several transcription factor families has been widely reported in stress response, there still is a lot to be uncovered, especially in soybean. Therefore, the objective of this study was to investigate the role of bHLH and trihelix-GT transcription factors in soybean responses to environmental stresses. Gene...

Soares-Cavalcanti,Nina M.; Belarmino,Luis C.; Kido,Ederson A.; Wanderley-Nogueira,Ana C.; Bezerra-Neto,João P.; Cavalcanti-Lira,Rafaela; Pandolfi,Valesca; Nepomuceno,Alexandre L.; Abdelnoor,Ricardo V.; Nascimento,Leandro C.; Benko-Iseppon,Ana M..
Plants experience various environmental stresses, but tolerance to these adverse conditions is a very complex phenomenon. The present research aimed to evaluate a set of genes involved in osmotic response, comparing soybean and medicago with the well-described Arabidopsis thaliana model plant. Based on 103 Arabidopsis proteins from 27 categories of osmotic stress response, comparative analyses against Genosoja and Medicago truncatula databases allowed the identification of 1,088 soybean and 1,210 Medicago sequences. The analysis showed a high number of sequences and high diversity, comprising genes from all categories in both organisms. Genes with unknown function were among the most representative, followed by transcription factors, ion transport...

Plants have the ability to recognize and respond to a multitude of pathogens, resulting in a massive reprogramming of the plant to activate defense responses including Resistance (R) and Pathogenesis-Related (PR) genes. Abiotic stresses can also activate PR genes and enhance pathogen resistance, representing valuable genes for breeding purposes. The present work offers an overview of soybean R and PR genes present in the GENOSOJA (Brazilian Soybean Genome Consortium) platform, regarding their structure, abundance, evolution and role in the plantpathogen metabolic pathway, as compared with Medicago and Arabidopsis. Searches revealed 3,065 R candidates (756 in Soybean, 1,142 in Medicago and 1,167 in Arabidopsis), and PR candidates matching to 1,261 sequences...

Inheritance of a long juvenile period under short-day conditions in soybean Genet. Mol. Biol.
Carpentieri-Pípolo,Valéria; Almeida,Leones Alves de; Kiihl,Romeu Afonso de S..
The long juvenile period (LJP) characteristic of soybean (Glycine max L. Merrill) cultivars delays flowering under short-day conditions. This trait may be important in increasing the adaptation range of soybean to low latitudes and provides greater flexibility for sowing times within the same latitude. The inheritance of the long juvenile period was studied in the MG/BR22 (Garimpo) soybean cultivar to provide knowledge to support the development of cultivars adapted to short day conditions. Four cultivars (‘Paraná’, ‘Bossier’, ‘Bragg’, and ‘Davis’) were crossed among each other and with MG/BR 22 (‘Garimpo’). The study was conducted under short-day conditions (early sowing) in a greenhouse and in the field, in Londrina, Paraná, (23°22’ south latitude). The...

Genetics of resistance to powdery mildew (Microsphaera diffusa) in Brazilian soybean populations Genet. Mol. Biol.
Gonçalves,Elaine Cristine Piffer; Di Mauro,Antonio Orlando; Centurion,Maria Aparecida Pessôa da Cruz.
Crosses between resistant and susceptible soybean cultivars were performed and the F2 populations were obtained to study the inheritance of soybean resistance to powdery mildew and to estimate the number and action of genes related to resistance. The reaction to powdery mildew was studied in a greenhouse and pots carrying plants with symptoms were distributed among the pots carrying the genotypes to be tested as a source of inoculum. Individual plants were scored according to the method of Yorinori (1997), with modifications, and classified as resistant or susceptible. The results showed that adult soybeans plants can present resistance to powdery mildew, which is controlled by one major gene with a dominant effect.

Genetic relationships between Chinese, Japanese, and Brazilian soybean gene pools revealed by simple sequence repeat (SSR) markers Genet. Mol. Biol.
Yamanaka,Naoki; Sato,Hiroyuki; Yang,Zhenyu; Xu,Dong He; Catelli,Lizandra Lucy; Binneck,Eliseu; Arias,Carlos Alberto Arrabal; Abdelnoor,Ricardo Vilela; Nepomuceno,Alexandre Lima.
An understanding of the relationship of geographically different soybean gene pools, based on selectively neutral DNA markers would be useful for the selection of divergent parental cultivars for use in breeding. We assessed the relationships of 194 Chinese, 59 Japanese, and 19 Brazilian soybean cultivars (n = 272) using 12 simple sequence repeat (SSR) markers. Quantification Theory III and clustering analyses showed that the Chinese and Japanese cultivars were genetically quite distant to each other but not independent, while Brazilian cultivars were distantly related to the cultivars from the other two countries and formed a cluster that was distant from the other two gene pool clusters. Our results indicated that the Brazilian soybean gene pool is...

Genetic analysis of soybean resistance to Fusarium solani f.sp. glycines Genet. Mol. Biol.
Fronza,Vanoli; Vello,Natal Antonio; Camargo,Luis Eduardo Aranha.
In order to study the genetic control of soybean resistance to sudden death syndrome (SDS), a 5 x 5 diallel with the F2 generation, without the reciprocals, was carried out in a greenhouse. The following parents were used: Forrest, MG/BR-46 (Conquista), IAC-4, FT-Cristalina, and FT-Estrela. The first two cultivars are more resistant to SDS than IAC-4, which is considered to be moderately resistant to SDS, and the last two cultivars are highly susceptible. The fungus was inoculated with three colonized sorghum grains placed at the bottom of the holes with two soybean seeds. Single plants were evaluated between 14 and 37 days after emergency based on foliar severity symptoms (1-5) of SDS. The disease incidence and a disease index were also calculated for...

The genetic base of Brazilian soybean cultivars: evolution over time and breeding implications Genet. Mol. Biol.
Wysmierski,Philip Traldi; Vello,Natal Antonio.
Genetic diversity is essential for crop breeding and one way to estimate it is through the concept of genetic base, which can be defined as the number of ancestors and their relative genetic contributions (RGC) to each cultivar. The RGC can be estimated through the coefficient of parentage between the ancestors and cultivars. Previous studies determined that the genetic base of Brazilian soybean was very narrow. The objective of this work was to evaluate the pedigree of 444 Brazilian soybean cultivars to estimate their genetic base. The cultivars were divided according to their release dates and according to their origin (public or private), and the genetic base for each group was also estimated. We found 60 ancestors, of which the top four (CNS, S-100,...

Cell wall, lignin and fatty acid-related transcriptome in soybean: achieving gene expression patterns for bioenergy legume Genet. Mol. Biol.
Pestana-Calsa,Maria Clara; Pacheco,Cinthya Mirella; Castro,Renata Cruz de; Almeida,Renata Rodrigues de; Lira,Nayara Patrícia Vieira de; Calsa Junior,Tercilio.
Increasing efforts to preserve environmental resources have included the development of more efficient technologies to produce energy from renewable sources such as plant biomass, notably through biofuels and cellulosic residues. The relevance of the soybean industry is due mostly to oil and protein production which, although interdependent, results from coordinated gene expression in primary metabolism. Concerning biomass and biodiesel, a comprehensive analysis of gene regulation associated with cell wall components (as polysaccharides and lignin) and fatty acid metabolism may be very useful for finding new strategies in soybean breeding for the expanding bioenergy industry. Searching the Genosoja transcriptional database for enzymes and proteins directly...

Soybean [Glycine max (L.) Merril], one of the most important crop species in the world, is very susceptible to abiotic and biotic stress. Soybean plants have developed a variety of molecular mechanisms that help them survive stressful conditions. Hybrid proline-rich proteins (HyPRPs) constitute a family of cell-wall proteins with a variable N-terminal domain and conserved C-terminal domain that is phylogenetically related to non-specific lipid transfer proteins. Members of the HyPRP family are involved in basic cellular processes and their expression and activity are modulated by environmental factors. In this study, microarray analysis and real time RT-qPCR were used to identify putative HyPRP genes in the soybean genome and to assess their expression in...

The study of tolerance mechanisms for drought stress in soybean is fundamental to the understanding and development of tolerant varieties. Using in silico analysis, four marker genes involved in the classical ABA-dependent and ABA-independent pathways of drought response were identified in the Glycine max genome in the present work. The expression profiles of the marker genes ERD1-like, GmaxRD20A-like, GmaxRD22-like and GmaxRD29B-like were investigated by qPCR in root samples of drought sensitive and tolerant soybean cultivars (BR 16 and Embrapa 48, respectively), submitted to water deficit conditions in hydroponic and pot-based systems. Among the four putative soybean homologs to Arabidopsis genes investigated herein, only GmaxRD29B-like was not regulated...
