Isolation, cultivation and molecular characterization of a new Trypanosoma equiperdum strain in Mongolia OAK
Suganuma, Keisuke; Narantsatsral, Sandagdorj; Battur, Banzragch; Yamasaki, Shino; Otgonsuren, Davaajav; Musinguzi, Simon Peter; Davaasuren, Batdorj; Battsetseg, Badgar; Inoue, Noboru.
Background: Trypanosoma equiperdum causes dourine via sexual transmission in Equidae. T. equiperdum is classified under the subgenus Trypanozoon along with the T. brucei sspp. and T. evansi; however, the species classification of Trypanozoon remains a controversial topic due to the limited number of T. equiperdum reference strains. In addition, it is possible that some were misclassified T. evansi strains. Thus, there is a strong need for a new T. equiperdum strain directly isolated from the genital mucosa of a horse with a clinically-and parasitologically-confirmed dourine infection. Methods: Trypanosomes isolated from the urethral tract of a stallion with suspected dourine, were directly cultivated using soft agarose media at 37 degrees C in 5 % CO2. For...

In vitro inhibitory effect of fosmidomycin on the asexual growth of Babesia bovis and Babesia bigemina OAK
Sivakumar, T; Aboulaila, M. R. A.; Khukhuu, A; Iseki, H; Alhassan, A; Yokoyama, Naoaki; Igarashi, Ikuo; 横山, 直明; 五十嵐, 郁男.
The in vitro inhibitory effect of fosmidomycin was evaluated against the asexual growth of bovine Babesia parasites (Babesia bovis and Babesia bigemina). In the cultures with these parasites, severe reduction of parasite growth was observed by the addition of 1.25 μg/ml of the final drug concentration. Under these conditions, B. bigemina was completely cleared until the third day of cultivation. On the other hand, B. bovis was completely eliminated on the fourth day when the concentration was 6.25 μg/ml in the culture. These parasites failed to grow again when a drug-free medium was substituted for the subsequent cultivation. The IC50 values of fosmidomycin against B. bovis were determined as 0.88 and 0.63 μg/ml in the in vitro cultures with...

In vitro conservation of apical meristem-tip of Melia azedarach L. (Meliaceae) under slow-growth conditions: (with 1 table & 2 figures) Phyton
Scocchi,AM; Mroginski,LA.
Studies on in vitro storage of Melia azedarach -"paradise tree"- under slow-growth conditions were carried out to develop an efficient protocol for the conservation of the genetic diversity of the crop. The response to different growth retardation treatments was examined using clone E. In vitro cultures of apical meristem-tips (0.2-0.3 mm in length) could be effectively maintained for 12 months at 4ºC on ¼ MS basal medium supplemented with 0.5 µM BA. Shoot proliferation was observed on 67 % of the recovered apical meristem-tips, after 12 months at 4ºC. Rooting of the regenerated shoots was achieved by reculturing them on MS + 17.5 µM IBA, during 4 days and subsequently transferred to MS medium lacking growth-regulators for 30 days. Complete plants were...

Dry matter partitioning differences between shoots and roots in two contrasting genotypes of orchids and their relationship with endogenous levels of auxins, cytokinins and abscisic acid Rev. Bras. Fisiol. Veg.
PERES,LÁZARO E. P.; MAJEROWICZ,NIDIA; KERBAUY,GILBERTO B..
Epiphytic orchids have been considered an interesting model for plant development and may have unusual features related to the hormonal control of shoot and root growth. In the present experiment we have studied two genotypes of Catasetum fimbriatum Lindl. (Orchidaceae) with an inverse pattern of shoot and root dry matter accumulation. Whereas the CFC1 genotype directs 60% of dry matter towards the shoot system, this same amount is directed towards the root system in the CFC4 genotype. Differences in the endogenous content of cytokinins, IAA and ABA, as well as differences in IAA conjugation and oxidation in shoots and roots were observed in the two genotypes. These differences were correlated with their dry matter partitioning patterns. Thus, the genotype...

Tetzner,Tatiane Almeida Drummond; Saraiva,Naiara Zoccal; Perecin,Felipe; Niciura,Simone Cristina Méo; Ferreira,Christina Ramires; Oliveira,Clara Slade; Garcia,Joaquim Mansano.
Embryo quality is influenced by the culture conditions that affect in vitro maturation (IVM), fertilization (IVF) and culture (IVC) rates. The present study investigated the feasibility of producing bovine embryos after the replacement of fetal calf serum (FCS) and bovine serum albumin (BSA) by ovalbumin (OVA). The IVM and IVC medium were supplemented with 10% FCS, 4 mg/mL BSA, or 4 mg/mL OVA. The IVF medium was supplemented with 6 mg/mL BSA or OVA. For IVM, supplementation with FCS, BSA, and OVA did not affect nuclear maturation or cortical granule migration. Higher rates of formation of two pronuclei were obtained when FCS was employed for IVM (79.97%), regardless of the supplement used for IVF, and when BSA was used for IVF (59.4%), regardless of the...

Plant regeneration from proroplasts of alfalfa (Medicago sativa) via somatic embryogenesis Scientia Agricola
Monteiro,Mariza; Appezzato-da-Glória,Beatriz; Valarini,Maria José; Oliveira,Carlos Alberto de; Vieira,Maria Lucia Carneiro.
Alfalfa is one of the most frequently studied species from the production of tissue culture-derived embryos point of view. In this study, five alfalfa cultivars were analyzed with reference to their ability to regenerate plants from protoplast cultures via somatic embryogenesis. Plant regeneration from leaf-derived protoplasts isolated from the cultivar Rangelander was achieved using a protocol defined for alfalfa cell suspension-derived embryogenesis. Because of its high efficiency, this procedure is recommended for protoplast electroporation-mediated genetic transformation of alfalfa.

RESPONSE TO in vitro SALT STRESS IN SUGARCANE IS CONDITIONED BY CONCENTRATION AND CONDITION OF EXPOSURE TO NaCI Acta biol.Colomb.
CAVALCANTE GRANJA,Manuela Maria; LACERDA E MEDEIROS,Maria Jaislanny; MEDEIROS DE ANDRADE SILVA,Marina; CAMARA,Terezinha; WILLADINO,Lilia; ULISSES,Claudia.
ABSTRACT Salinity is one of the major environmental stress factors that affect crop productivity, as well as interfering with plant growth and development, resulting in reduced production quality. Given this, we highlight the importance of research in response to plants subjected to salt stress in order to assess the physiological and biochemical behavior of genotypes, with the objective of selecting the more tolerant. One way to ensure the uniformity of the response of the plants is through in vitro cultivation, which allows control of the cultivation conditions. Therefore, the objective of this work was to evaluate the response of two commercial varieties of sugar cane exposed to saline stress in different conditions (gradual and abrupt). Two varieties...
