Background: Trypanosoma equiperdum causes dourine via sexual transmission in Equidae. T. equiperdum is classified under the subgenus Trypanozoon along with the T. brucei sspp. and T. evansi; however, the species classification of Trypanozoon remains a controversial topic due to the limited number of T. equiperdum reference strains. In addition, it is possible that some were misclassified T. evansi strains. Thus, there is a strong need for a new T. equiperdum strain directly isolated from the genital mucosa of a horse with a clinically-and parasitologically-confirmed dourine infection. Methods: Trypanosomes isolated from the urethral tract of a stallion with suspected dourine, were directly cultivated using soft agarose media at 37 degrees C in 5 % CO2. For... |

The in vitro inhibitory effect of fosmidomycin was evaluated against the asexual growth of bovine Babesia parasites (Babesia bovis and Babesia bigemina). In the cultures with these parasites, severe reduction of parasite growth was observed by the addition of 1.25 μg/ml of the final drug concentration. Under these conditions, B. bigemina was completely cleared until the third day of cultivation. On the other hand, B. bovis was completely eliminated on the fourth day when the concentration was 6.25 μg/ml in the culture. These parasites failed to grow again when a drug-free medium was substituted for the subsequent cultivation. The IC50 values of fosmidomycin against B. bovis were determined as 0.88 and 0.63 μg/ml in the in vitro cultures with... |

Studies on in vitro storage of Melia azedarach -"paradise tree"- under slow-growth conditions were carried out to develop an efficient protocol for the conservation of the genetic diversity of the crop. The response to different growth retardation treatments was examined using clone E. In vitro cultures of apical meristem-tips (0.2-0.3 mm in length) could be effectively maintained for 12 months at 4ºC on ¼ MS basal medium supplemented with 0.5 µM BA. Shoot proliferation was observed on 67 % of the recovered apical meristem-tips, after 12 months at 4ºC. Rooting of the regenerated shoots was achieved by reculturing them on MS + 17.5 µM IBA, during 4 days and subsequently transferred to MS medium lacking growth-regulators for 30 days. Complete plants were... |

The present work aimed at maximizing the number of plantlets obtained by the micropropagation of pineapple (Ananas comosus (L.) Merrill) cv. Pérola. Changes in benzylaminopurine (BAP) concentration, type of medium (liquid or solidified) and the type of explant in the proliferation phase were evaluated. Slips were used as the explant source, which consisted of axillary buds obtained after careful excision of the leaves. A Sterilization was done in the hood with ethanol (70%), for three minutes, followed by calcium hypochlorite (2%), for fifteen minutes, and three washes in sterile water. The explants were introduced in MS medium supplemented with 2mg L-1 BAP and maintained in a growth room at a 16h photoperiod (40 mmol.m-2.s-1), 27 ± 2ºC. After eight weeks,... |

Abstract Considering there is no illustrated scale of the principle anomalies observed for banana seeds, the objective of this guide is to cover in detail, for the first time, the categories to classify the most common types of anomalies observed in banana embryos and seeds, as well as in vitro embryo rescue and culture, taking into account the experience acquired by the banana breeding program at Embrapa in Brazil. Four classes to classify the seeds were proposed and with regards to classifying the embryos, eight classes were established. |

The present study examined the effects of green tea polyphenols (GTP), insulin-like growth factor-I (IGF-I) and glucose on oocyte in vitro maturation, subsequent embryo development and blastocyst quality in bovine. Cumulus-oocyte complexes (COC) were aspirated from the ovaries and cultured in synthetic oviduct fluid supplemented with MEM amino acids (SOFaa) media supplemented with one of the following supplements: GTP (0, 10, 15 and 20 µM), IGF-I (0, 50, 100 and 150 ng/mL) or glucose (0, 1.5, 5.6 and 20 mM) for 24 h. The results showed that oocytes cultured in media supplemented with 15 µM GTP, 100 ng/mL IGF-I and 5.6 mM glucose, in separate experiments, have higher cleavage and blastocyst rates compared with oocytes cultured in media without or with other... |

Embryo quality is influenced by the culture conditions that affect in vitro maturation (IVM), fertilization (IVF) and culture (IVC) rates. The present study investigated the feasibility of producing bovine embryos after the replacement of fetal calf serum (FCS) and bovine serum albumin (BSA) by ovalbumin (OVA). The IVM and IVC medium were supplemented with 10% FCS, 4 mg/mL BSA, or 4 mg/mL OVA. The IVF medium was supplemented with 6 mg/mL BSA or OVA. For IVM, supplementation with FCS, BSA, and OVA did not affect nuclear maturation or cortical granule migration. Higher rates of formation of two pronuclei were obtained when FCS was employed for IVM (79.97%), regardless of the supplement used for IVF, and when BSA was used for IVF (59.4%), regardless of the... |

ABSTRACT This study developed a methodology for callus induction in leaf segments of B. verbascifolia and evaluated the bioactive phenolic compounds production. Leaf explants were cultured in MS medium with 30 g L-1 sucrose, solidified with 7 g L- 1 agar supplemented with 2,4-D (0; 4.52; 9.05; 18.10 µM) and BAP (0; 4.44; 8.88; 17.75 µM ) in the presence and absence of light. Forty-five days after inoculation we assessed the percentage of callus induction, color, consistency, fresh and dry matter, total phenols, flavonoids, tannins contents, and chromatographic profile by HPLC-DAD method. Callus induction occurred only in medium with growth regulators. Maximal induction (100%) was found in medium containing 2,4-D combined with BAP in the presence and... |

ABSTRACT Salinity is one of the major environmental stress factors that affect crop productivity, as well as interfering with plant growth and development, resulting in reduced production quality. Given this, we highlight the importance of research in response to plants subjected to salt stress in order to assess the physiological and biochemical behavior of genotypes, with the objective of selecting the more tolerant. One way to ensure the uniformity of the response of the plants is through in vitro cultivation, which allows control of the cultivation conditions. Therefore, the objective of this work was to evaluate the response of two commercial varieties of sugar cane exposed to saline stress in different conditions (gradual and abrupt). Two varieties... |

ABSTRACTCattleya intermediais an Atlantic Forest species endemic to Brazil that is classed as vulnerable on the list of threatened species. In this study, C. intermedia plantlets were micropropagated in an asymbiotic culture and the influence of different concentrations of sucrose (15, 30, 45 and 60 g L-1, plus a zero sucrose medium) and macronutrient salts (complete Murashige and Skoog (MS) medium and half MS medium (with half-strength macronutrients)) on survival and development of the plantlets was evaluated. In all treatments 100% plantlet survival was achieved. The integrated analysis of height of aerial part, number of leaves per plantlet, fresh mass, number of roots per plantlet and length of the longest root showed that the plantlets exhibited... |
