Can the hemoglobin characteristics of vesicomyid clam species influence their distribution in deep-sea sulfide-rich sediments? A case study in the Angola Basin ArchiMer

Vesicomyids live in endosymbiosis with sulfur-oxidizing bacteria and therefore need hydrogen sulfide to survive. They can nevertheless live in a wide range of sulfide and oxygen levels and depths, which may explain the exceptional diversity of this clam family in deep-sea habitats. In the Gulf of Guinea, nine species of vesicomyid clams are known to live in cold-seep areas with pockmarks from 600 to 3200 m deep, as well as in the organic-rich sediments of the Congo deep-sea fan at 5000 m deep. Bacteria from deep-sea hydrothermal vents constitute an attractive source of bioactive molecules. In particular, exopolysaccharides (EPS) produced by these bacteria become a renewable source of both biocompatible and biodegradable molecules. The low molecular weight (LMW) derivatives of the GY785 EPS produced by the deep-sea hydrothermal vent strain Alteromonas infernus have previously displayed some biological properties, similar to those of glycosaminoglycans (GAG), explored in cancer and tissue engineering. UPLC-MSE Profiling of Phytoplankton Metabolites: Application to the Identification of Pigments and Structural Analysis of Metabolites in Porphyridium purpureum ArchiMer

A fast and high-resolution UPLC-MSE analysis was used to identify phytoplankton pigments in an ethanol extract of Porphyridium purpureum (Pp) devoid of phycobiliproteins. In a first step, 22 standard pigments were analyzed by UPLC-MSE to build a database including retention time and accurate masses of parent and fragment ions. Using this database, seven pigments or derivatives previously reported in Pp were unequivocally identified: beta,beta-carotene, chlorophyll a, zeaxanthin, chlorophyllide a, pheophorbide a, pheophytin a, and cryptoxanthin. Structural data on a bacterial exopolysaccharide produced by a deep-sea Alteromonas macleodii strain ArchiMer

Some marine bacteria collected around deep-sea hydrothermal vents are able to produce, in laboratory conditions, complex and innovative exopolysaccharides. In a previous study, the mesophilic strain Alteromonas macleodii subsp. fijiensis biovar deepsane was collected on the East Pacific Rise at 2600m depth. It was isolated from a polychaete annelid Alvinella pompejana and is able to synthesise and excrete the exopolysaccharide deepsane. Naja naja snake venom presents abundant thermostable peptides. Many of them possess useful pharmacological activity that may be employed for drug development. For the proteomic characterization of such toxins, in the present study, Naja naja venom solution was heated up to 100°C for 10, 30, 60, 120, 180 and 300 minutes and protein fractions of non-heated and heated venom were analyzed by two-dimensional nano-liquid chromatography coupled online with tandem mass spectrometry. Molecular and morphological characterization of hydrochar produced by microwave-assisted hydrothermal carbonization of cellulose PAB

The objective of this work was to characterize the morphology and molecular composition of the hydrochar produced by microwave-assisted hydrothermal carbonization of cellulose. The produced hydrochar consists mainly of aggregate microspheres with about 2.0 µm in diameter, with aliphatic and aromatic structures and the presence of carbonyl functional groups. The aromatic groups are formed mainly by benzofuran-like structures, being chemically different from common cellulose char. Microwave-assisted hydrothermal carbonization yields a functionalized carbon-rich material similar to that produced by the conventional hydrothermal process.

ABSTRACT Acylated anthocyanins from a purple-fleshed sweet potato (PFSP), obtained by organic cultivation in Brazil, were characterized after separation by a high performance liquid chromatography-diode array detector (HPLC-PDA). These anthocyanins were manually collected at the detector output, concentrated and injected into a high resolution mass spectrometer (ESI-QTOF-MS2). Twenty-two acylated anthocyanins were detected. In tropical soils with intensive agriculture an increasing sulfur deficiency has been verified in several crops. The low available S in these soils is caused by the continuous use of concentrated NPK fertilizers. Production of single superphosphate labeled with 34S Scientia Agricola

Single superphosphate is currently one of the mostly used fertilizers as an alternative source for phosphorus and sulphur. Sulphur presents four stable isotopes (32S, 33S, 34S, and 36S) with natural abundances of 95.00; 0.76; 4.22; and 0.014% in atoms, respectively. Single superphosphate labeled with the 34S isotope was obtained from a chemical reaction in stoichiometric amounts between Ca(H2PO4)2 and Ca34SO4.2H2O. Calcium sulphate (Ca34SO4.2H2O) was enriched with 5.85 ± 0.01 atoms % of 34S. The Ca(H2PO4)2 reagent was obtained from a reaction between CaCl2.2H2O and H3PO4. 100 isolates of phytopathogenic bacteria of rice, Xanthomonas oryzae, were identified by matrix-assisted laser desorption ionization–time of-flight mass spectrometry (MALDI-TOF MS). The results showed the whole-cell protein spectrum profile of mass spectra of Xanthomonas oryzae pv.oryzae,and X. oryzae pv. oryzicola shared the common spectra peak. However, X. oryzae pv.oryzae presented difference mass spectra at 2641.55, 3729.83, 4919.52, 5608.13, 9857.82, 12480.64 and 18737.92 m/z.whereas X. oryzae pv. oryzicola presented unique spectra of 6279.48 and 9179.89 m/z.

The purpose of this work was to improve the separation and yield of pure β- and α-trypsin isoforms by ion-exchange chromatography and to characterize some physical-chemical properties of these isoforms. Purification of trypsin isoforms was performed by ion-exchange chromatography in 0.1 mol/L tris-HC buffer, pH 7.10 at 4ºC. The sample loading, salt concentration, flow rate and pH of mobile phase were varied to determine their effects on the resolution of the separation. The resolution was optimized mainly between β- and α-trypsin. Pure isoforms were obtained by chromatographying 100 mg of commercial trypsin during seven days, yielding 51 mg of high purity β-trypsin and 13 mg of α-trypsin partially pure, with small amounts of contaminating of ψ-trypsin....

Proteomic analysis of whey from bovine colostrum and mature milk BABT

The aim of this study was to standardize a methodology to obtain two-dimensional (2D) maps of whey proteins from bovine colostrum and mature milk, using two-dimensional electrophoresis, in order to identify the minor proteins by matrix-assisted laser desorption/ionization tandem time-of-flight mass spectrometry (MALDI TOF/TOF MS). A total of 38 proteins were identified, 20 spots in the colostrum whey and 18 in the mature milk whey; 5 of them were identified for the first time.
